Abstract: TheroleofD(EC3.1.4.4)wasstudiedinthechangingofphospholipidcontentduringcottonseed (Gossipiumhirsutum108-G)maturation.

TodefinephospholipaseDactivitycottonseedhomogenateswereincubatedinawater-methanolenvironmentat37°Cforvarious timeintervals(15,30,60,120,and300min).Enzymeactivitywasjudgedfromtheformationofhydrolysisproducts-phosphatidic acidandphosphatidylmethanol.Thephospholipidcontentwasdefinedbyusingthin-layerchromatographywithafollowupanalysis ofquantityinphospholipidfractions.

ItwasshownthatphospholipaseDdisplayedtwofunctionsduringcottonseedmaturation:transferaseand.Hydrolase functioncouldbedefinedfromtheincreaseoftheproduct(phosphatidicacid)concentrationduringthecompletematurationof the seed.Wenoticedthattheconcentrationincreased.Transferaseactivitycouldbedefinedthroughtheincreaseofthe phosphatidylmetanolconcentrationduringtheincubationofphospholipidsinawater-methanolenvironment.Theexperimentaldata showedthatphospholipaseDplaysanimportantroleinphospholipidmetabolism.
