Introduction to the Microscope
.Care .Parts .Focusing • Always carry with 2 hands • Only use lens paper for cleaning • Do not force knobs • Always store covered • Keep objects clear of desk and cords Eyepiece
Body Tube
Revolving Nosepiece Arm Objective Lens
Stage Stage Clips Coarse Focus Diaphragm Fine Focus Light
Base COMPOUND MICROSCOPE
You are looking through 2 lenses at one time.
Which lenses??? o The eye piece and oObjective Lens
•Eyepiece or Ocular Lens: 10X magnification
•Body Tube: Separates the eyepiece lens from the objective lens
•Revolving nose piece:
Holds the objective lenses; Allows them to rotate
OBJECTIVES Scanning Objective Lens: 4X Low Power Objective Lens: 10X* Standard = Always 10X
High Power Objective Lens: 40X or 44X* Depends on the microscope STAGE: Where the slide is placed
STAGE CLIPS: Holds the slide in place
DIAPHRAGM: Regulates (CONTROLS) the amount of light passing through the opening of the stage
MIRROR: (Light source) Has a concave surface; concentrates & reflects light up through the body tube
COARSE ADJUSMENT KNOB
Moves the stage (or body tube) QUICKLY up and down for FOCUSING
FINE ADJUSTMENT KNOB: Moves the stage (or body tube) SLOWLY for SHARPENING the image
ARM: Supports the body tube
Base: Supports the microscope
SLIDE: Holds the specimen
COVER SLIP: Protects the specimen and FLATTENS it (light must be able to pass through the specimen) TOTAL MAGNIFICATION
Multiply the eye piece by the objective lens to find out how many times the specimen is magnified
EX: Eyepiece X Low Power Objective (10X*) X (10X) = 100X Total Magnification *standard magnification=always 10X Total Magnification continued….
Another example:
Eye piece X High Power Objective
(10X*) X (40X) = 400X Total Magnification
*standard magnification=always 10X
• Place the Slide on the Microscope • Use Stage Clips • Click Nosepiece to the lowest (shortest) setting • Look into the Eyepiece • Use the Coarse Focus • Follow steps to focus using low power • Click the nosepiece to the longest objective • Do NOT use the Coarse Focusing Knob • Use the Fine Focus Knob to bring the slide
What can you find on your slide?
