Entomology March 2009 Thesis presented for the degree of Doctor of Philosophy at Imperial College of Science, Technology and Medicine, and for the Diploma of Imperial College (DIC) Division of Biology, Imperial College London Silwood Park Campus, Ascot, Berkshire, SL5 7PY, UK 1 Abstract Dieback disease caused by Fusarium and Lasiodiplodia species is a major threat to cocoa production in Ghana and elsewhere in West Africa. Current recommendations involve insecticide application to control mirid bugs whose feeding punctures provide entry points for these fungi. Little is known about the true identity of the causal pathogens of this disease. Earlier work implicated F. decemcellulare as the causal agent and more rarely L. theobromae (Cotterell, 1927; Crowdy, 1947). A total of 117 single spore fungal cultures was established from diseased cocoa stems imported from Ghana. On morphological grounds cultures could be designated as either Fusarium or Lasiodiplodia spp. The Fusarium cultures exhibited inter-isolate variability with respect to macroscopic appearance and macro-conidium morphology, suggesting the presence of more than a single species. The isolates were further characterised by PCR amplification and sequencing of the ITS region of rDNA and comparison with authentic reference cultures. Thirty-seven Fusarium isolates were identified to twenty F. chlamydosporum, nine F. solani and four isolates each of F. oxysporum and F. proliferatum. The thirty-six Lasiodiplodia isolates were identified to two species, twenty-seven L. pseudotheobromae and nine L. theobromae. In pathogenicity tests, F. chlamydosporum, F. oxysporum, F. proliferatum, F. solani and L. pseudotheobromae, previously unknown as pathogens of either cocoa or any member of the Malvaceae, caused significant wilting and dieback in Amelonado seedlings similar to that observed in the field. All isolates exhibited optimal growth at 30 ºC on PDA. Disease incidence in 29 and 15 cocoa germplasm lines in the laboratory and greenhouse, respectively, showed reproducible differences in their reaction to necrotic lesion and dieback infection. LCTEEN 37/F was one of the most susceptible genotypes. CATIE 1000, T85/799 and MXC 67 were the most tolerant and could be used in cocoa breeding programmes for resistance to dieback. 2 DECLARATION OF OWN WORK I confirm that this thesis: Pathogen diversity and host resistance in dieback disease of cocoa caused by Fusarium decemcellulare and Lasiodiplodia theobromae is entirely my own work, conducted under the supervision of Dr Simon Archer and Dr Simon Leather. These problems are aptly illustrated in the genus Phoma. This phytopathologically significant fungal genus is currently subdivided into nine sections which are mainly based on a single or just a few morphological characters. However, this subdivision is ambiguous as several of the section-specific characters can occur within a single species. In addition, many teleomorph genera have been linked to Phoma, three of which are recognised here. In this study it is attempted to delineate generic boundaries, and to come to a generic circumscription which is more correct from an evolutionary point of view by means of multilocus sequence typing. Therefore, multiple analyses were conducted utilising sequences obtained from 28S nrDNA (Large Subunit - LSU), 18S nrDNA (Small Subunit - SSU), the Internal Transcribed Spacer regions 1 & 2 and 5.8S nrDNA (ITS), and part of the β-tubulin (TUB) gene region.

They are implicated in the production of antibiotic and other compounds of therapeutic importance. Ecologically they provide several benefits to plants, including protection from plant pathogens. There have been numerous studies on the biodiversity and ecology of endophytic fungi. Some taxa dominate and occur frequently when compared to others due to adaptations or capabilities to produce different primary and secondary metabolites. It is therefore of interest to examine different fungal species and major taxonomic groups to which these fungi belong for bioactive compound production. In the present paper a list of endophytes based on the available literature is reported. More than 800 genera have been reported worldwide. Dominant genera are Alternaria, Aspergillus, Colletotrichum, Fusarium, Penicillium, and Phoma. Most endophyte studies have been on angiosperms followed by gymnosperms. Among the different substrates, leaf endophytes have been studied and analyzed in more detail when compared to other parts. Most investigations are from Asian countries such as China, India, European countries such as Germany, Spain and the UK in addition to major contributions from Brazil and the USA.

In this thesis, my aims were (1) to employ next-generation sequencing (NGS) based metabarcoding for the detection and identification of plant pathogens at the ecosystem scale in New Zealand, (2) to characterise plant pathogen communities, and (3) to determine the environmental drivers of these communities. Smitha Revi1, Madhu Subramanian2 1,2,Department of Agricultural Entomology, Kerala Agricultural University, College of Horticulture, Vellanikkara, Kerala, India Abstract The effect of mycological media on mycelial growth, sporulation, viability of spores and fungal biomass production of Fusarium oxysporum, a native endophytic entomopathogenic fungus was studied. The nutritional requirement varies with the entomopathogenic fungal species and even the fungal strain. In this study, five different media were used and significant variability was observed on different media. Isolate produced maximum mean mycelial growth on SMA medium followed by SMAY and PDA. PDA medium supported maximum sporulation (1.72 x 105spores ml-1) and viable spore count (5x105 cfu ml-1). Maximum mean dry fungal biomass was observed in SDYB (5.811 g). Sporulation is favoured by nutritional conditions that restricted the fungal biomass production in PDB, which recorded significantly lower fungal biomass (1.345 g). Potato dextrose medium was found to be significantly superior and best suited among all media in the present investigation. INTRODUCTION Fusarium is a large genus of hyaline filamentous fungi, which are ubiquitous with cosmopolitan distribution. They belong to the family Nectriaceae of the order Hypocreales within the fungal phylum Ascomycota. They can be found in air, water, plants, insects, soils and organic substrates. Fusarium spp. such as Fusarium oxysporum caused 100% of the mortalities of the insect larvae. Insect biocontrol potential of Fusarium is favored by their excellent soil survivability as saprophytes, and sometimes, insect-pathogenic strains do not exhibit phytopathogenicity.
