  • Plant and Bacterial Extracts Involved on Arbuscular Mycorrhizal Fungus Stimulation and Pathogens Suppression
Bioagro 27(2): 103-110. 2015 PLANT AND BACTERIAL EXTRACTS INVOLVED ON ARBUSCULAR MYCORRHIZAL FUNGUS STIMULATION AND PATHOGENS SUPPRESSION Andre F. Cruz1 ABSTRACT ‘Amanatsu’ orange (Citrus × natsudaidai Hayata) seedlings were cultivated under aluminum [Al2(SO4)3)], salinity (NaCl) and water stress conditions in green house. ‘Amanatsu’ root extracts (ARE) were obtained from these plants by using a flash chromatograph, and an experiment in vitro to measure the effect of the extracts on hyphal growth of arbuscular mycorrrhizal fungus (AMF) was carried out. The hyphal growth was stimulated by ARE from aluminum and water stressed plants, whereas it was inhibited by ARE from the plants with NaCl. In particular, the hyphal growth in Petri dishes with ARE from aluminum and water stressed plants were around 70 % higher than that with agar only. In a second experiment, the 25 % MeOH eluates of Bahiagrass (Paspalum notatum) and millet (Pennisetum glaucum) root extracts stimulated, in vivo, AMF infection on trifoliate orange (Poncirus trifoliata) rootstocks. A third experiment was carried out to evaluate bacterial extracts on AMF (Gigaspora margarita and Glomus clarum) hyphal growth and soil-borne plant pathogens suppression. The results indicated that the 25 % MeOH fraction could stimulate the hyphal growth whereas mainly the 75 and 100 % MeOH fraction inhibited the pathogen growth in vitro.

  • Protective Interference of Mild Strains of Citrus Tristeza Virus Against a Severe Strain in Morita Navel Orangez
Protective Interference of Mild Strains of Citrus Tristeza Virus Against a Severe Strain in Morita Navel Orangez Hiroyuki Ieki and Akira Yamaguchi ABSTRACT. Nursery plants of citrus tristeza virus (CTV)-free Morita navel orange obtained by thermotherapy were individually preinoculated with mild strains of CTV, collected from citrus trees in orchards, or artificially produced by heat treatment of Morita navel orange infected with a severe strain of seedling yellows CTV. They were challenge-inoculated with a severe strain of seedling yellows CTV by the vector (Toxoptera citricidus) or by side-graft inoculation. In the first experiment, the occurrence of stem pitting on four plants preinoculated with M-10 (HM-55 from Hassaku, Hiroshima Pref.) mild strain was slight 4 yr after challenge-inoculation, and two of four plants showed mild symptoms on indexing to Mexican lime seedlings. Fruit size and production of CTV-free plants and plants preinoculated with mild strain were larger than those of plants with the severe strain, but there was little difference in trunk girth of plants between treatments. In the second experiment, the M-23(A) mild strain produced by heat treatment was effective 2 to 3 yr after challenge-inoculation, based on mild stem pitting observed and mild symptoms on Mexican lime seedlings used to index challenged plants. The M-10 and M-23(A) mild strains appear promising to protect Morita navel orange trees from severe strain challenge under the field conditions. Index words. Artificial production of mild strains of CTV, Toxoptera citricidus, stem pitting. Conversion from Satsuma manda- rin to other varieties such as Has- with CTV has been caried out world- saku, navel orange, Miyauchi-Iyo, wide (1,3,4,7,9, 10, 11, 15, 16,17).

The total contents of synephrine enantiomers in citrus fruit samples were exocarp > mesocarp > endocarp > sarcocarp, suggesting that synephrine content of outer side of citrus fruits was higher than that of the inner side. (R)-Synephrine was detected in exocarp of eleven fresh citrus fruits, except for lemon, lime, and grapefruit samples. (S)-Synephrine was determined in the exocarp of four citrus fruits (mikan, orange, bitter orange, and ponkan samples) and the ratio of (S)-synephrine to total synephrine was 0.5 – 0.9%. The racemization of (R)-synephrine in aqueous solution during heating at 100°C was also examined.

  • OTHER VIRUS DISEASES the Occurrence of Satsuma Dwarf Virus
OTHER VIRUS DISEASES The Occurrence of Satsuma Dwarf Virus in China Zhou Changyong, Zhao Xueyuan, Jiang Yuanhui and He Xinhua ABSTRACT. From 1985 to 1991,84 samples from satsuma mandarins, mandarin hybrids and navel oranges, imported from Japan in recent years, were collected from eight provinces, and indexed for satsuma dwarf virus (SDV) by using the indicator plants, White sesame, Blackeye cowpea and Satisfac- tion kidney bean, and by ELISA. Miyamoto satsuma samples collected trom Huangyan, Zhejiang Province and from Fengjie, Sichuan Province were infected with SDV, while all the others were SDV-free. Nine cultivars of extreme early satsuma, especially those collected from Huangyan and Huaning, Yunnan Province, were infected with citrus tatter leaf virus (CTLV). Other than satsuma, 25 important citrus cultivars collected from the National Citrus Genetic Resources Collection at Chong- qing were found to be SDV-free by ELISA. The results suggest that the occurrence of SDV in China is limited to a few cultivars in a few locations. Satsuma dwarf is an economically Hashimoto, Ichifumi, Imada, Inaba, important virus disease in Japan. The Ishitsuka, Kitaguchi, Mitake, typical symptom, the so-called "boat- Miyamoto, Oomura, Ooura, Tanimoto, shaped leaf," appears on the spring Takabayashi, Toku mori, Ueno, flush. Since the first record in 1950 (5), Wakiyama and Yamakawa No. 3), it has become widespread in almost all early satsuma (Kamei, Matsuyama, the citrus areas of Japan (3). SDV has Miyagawa, Okitsu and Tachima), mid- also been reported from Turkey (1)and dle satsuma (Aoe, Dobashibeni, Korea (4). Satsuma mandarin was im- Hayashi, Kunou, Morita, Mukaiyama, ported into China from Japan, and in Nankan No.
