Our project working on “The time capsule analysis of ancient variation stuck in polyploids” is now coming to an end. Allopolyploids originate from the merging of two or more entire genomes and are in most cases immediately isolated from their parents. The com- plexity of the genome ancestry is increased by hybridization with other allopolyploids as well and backcrossing with their parental species. Dactylorhiza (Orchidaceae: Orchidinae) is a genus much affected by allopolyploid speciation and reticulate phylogenetic relationships. Here we use the genetic variation at tens of thousands of genomic positions to trace evolu- tionary history in Dactylorhiza by first delimiting the diploid taxa and establishing their rela- tionship and then grouping 16 allopolyploids by maximum affiliation to their putative parents. The two sibling allopolyploids, D. majalis and D. traunsteineri were studied in more depth to investigate genetic structure and demography. The diploid parents throughout their European distribution were used to create two artificial genomic references for which two pipelines were used to align the polyploid reads and call genotypes/infer genotype likelihood. Two in- dependent origins are suggested for D. traunsteineri and D. majalis, though gene flow does occur. Two other boreal sister genera Nigritella and Gymnadenia are morphologically dis- tinct, though single-marker studies have previously given contradictory results on the inclu- sion/exclusion of Nigritella in Gymnadenia. Thousands of genetic markers were used to un- ravel their phylogenetic status. Lastly, evolutionary and ecological questions were addressed within the genus Epipactis on speciation via transitions from allogamy to autogamy. Genomic inferences based on RADseq (i.e. restriction-site associated DNA sequencing) data have be- come widely applied to answer evolutionary questions in diploid organisms and is included in all four chapters of this PhD, also showing its application to polyploid organisms. We obtain highly congruent evolutionary relationships inferred both with genotype-based and genotype- free analytical methods and develop bioinformatic approaches that can be applied to other naturally occurring, non-model, polyploid complexes. 6 ZUSAMMENFASSUNG Die Untersuchung phylogenetischer Beziehungen von sich kürzlich entwickelten Gruppen stellt eine große Herausforderung dar, insbesondere bei fortlaufender Hybridisierung. Allopo- lyploide Organismen stammen aus der Verschmelzung von zwei oder mehreren Genomen und werden in den meisten Fällen sofort von ihren Eltern isoliert. Die Komplexität der genomi- schen Abstammung wird durch die Hybridisierung mit anderen Allopolyploiden sowie durch Rückkreuzung mit ihren möglicherweise vorkommenden Elternarten erhöht. Die Gattung Dactylorhiza (Orchidaceae: Orchidinae) ist stark von allopolyploider Artbildung und retikulä- ren phylogenetischen Beziehungen betroffen. In dieser Arbeit verwenden wir genetische Va- riation, um die Evolutionsgeschichte in Dactylorhiza zu rekonstruieren, indem wir zunächst die diploiden Taxa abgrenzen, ihre Verwandtschaftsbeziehung festlegen und dann 16 allopo- lyploide Taxa nach maximaler Zugehörigkeit zu ihren mutmaßlichen Eltern gruppieren. Die beiden Geschwister Allopolyploide, D. majalis und D. traunsteineri, wurden untersucht, um die genetische Struktur und Demografie zu untersuchen. Die diploiden Eltern wurden in ihrer gesamten europäischen Verbreitung dazu verwendet, zwei künstliche genomische Referenzen zu erstellen um die polyploiden Werte abzugleichen und Genotypen zu ermitteln bzw. Geno- typ-Wahrscheinlichkeit zu nennen. Für D. traunsteineri und D. majalis wurden zwei unab- hängige Ursprünge ermittelt, obwohl ein Genfluss zwischen beiden Arten herrscht. Zwei wei- tere boreale Schwestergattungen Nigritella und Gymnadenia sind morphologisch unterschied- lich, obwohl Einzelmarkerstudien zuvor widersprüchliche Ergebnisse zum Einschluss / Aus- schluss von Nigritella in Gymnadenia erbracht haben. Tausende genetischer Marker wurden verwendet, um ihren phylogenetischen Status aufzuklären. Schließlich wurden evolutionäre und ökologische Fragen in der Gattung Epipactis zur Artbildung durch einen Übergang von Allogamie zur Autogamie angesprochen. RAD-Daten (Restriktionsstellen Assoziierte DNA) werden in weitem Umfang zur Beantwortung von evolutionären Fragestellungen diploider Organismen angewendet. Die vier Kapitel dieser Doktorarbeit zeigen den Nutzen von RAD- Daten für die Arbeit

    DACTYLORHIZA NEVSKI, THE CORRECT GENERIC NAME OF THE DACTYLORCHIDS By P. F. HUNT and V. S. SUMMERHAYES Royal Botanic Gardens, Kew ABSTRACT The correct generic name for the dactylorchids (marsh and spotted orchids) is shown to be Dactylorhiza Nevski. A list of species of Dactylorhiza is given and the subspecies occurring in the British Isles are indicated. Several new combinations at specific and subspecific rank and five new bigeneric hybrid formulae are published for the first time. In his Species Plantarum (939-944, 1753) Linnaeus divided the genus Orchis into three parts based on the morphology of the roots, namely: Bulbis indivisis, Bulbis palmatis and Bulbis fasciculatis. Some time later, Necker, in his Elementa Botanica (3, 129, 1790), raised these groups to generic level although he actually used the category name 'species naturalis' for them. Orchis L. was retained for Bulbis indivisis whilst Bulbis palmatis and Bulbis fasciculatis became Dactylorhiza Necker. The next important treatment of the genus was by Klinge, in 1898 (Acta Hort. Petrop. 17,148). He recognized two subgenera, namely Eu-orchis, into which he placed the Linnaean Bulbis indivisis, and Dactylorchis which included Bulbis palmatis. This classification was adopted by many later workers, but in 1935, Nevski, in his account of the Orchidaceae for the Flora URSS, substituted Necker's name Dactylorhiza for the second of Klinge's sub­ genera on the ground that it was earlier than Dactylorchis Klinge. Nevski also seems to have excluded Linnaeus's Bulbis fasciculatis, at least by implication. Two years later, however, Nevski evidently decided that the two subgenera were better treated as distinct genera and adopted the generic name Dactylorhiza, making a new combination, D.

Moreover, this examination highlights which species appear to be underprotected and therefore need to be included in some catalogues of protection or be catalogued under some category of protection. Te national and regional catalogues that should be implemented are shown, as well as what species should be included within them. Tis report should be a helpful guideline for environmental policies about orchids conservation in Spain, at least at the regional and national level. Around 76% of the Spanish orchid fora are listed with any fgure of protection or included in any red list, either nationally (about 12-17%) or regionally (72%).

MRKVICKA Zusammenfassung: Die systematische Stellung der Gattung Epipactis in der Subtribus Cephalantherinae wie auch die Stel- lung dieser Subtribus innerhalb der Unterfamilie Neottioideae wird an Beispielen enhvickungsgeschicht- licher Untersuchungen diskutiert. Nach den neuesten molekularen Daten, die aus DNA-Sequenzanalysen gewonnen wurden, ist ein Stammbaum erstellt worden, in dem die Neottioideae in die "epidendroids" eingereiht wurden. Das steht im Widerspruch zu dem in unserer Arbeit praktizierten Klassifikations- System, das in "Die Orcliideen" R. SCHLECHTER in der Bearbeitung von F. BMEGER und K. SENGHAS venvendet wird. Die Ableitung einer Orchideenblüte aus dem Liliiflorae-Erbe ermögliclit eine Differentialdiagnose zwi- schen den Orchidaceae und den Apostasiaceae. Die Apostasiaceae, die viele Autoren als Unterfamilie Apostasioideae zu den Orchidaceae stellen, werden durch vergleichende Blütenanalysen von dieser Familie abgetrennt. Der Entwickiungstendenz des Gynoeceums der Orchideen, durch die es zum Auf- bau eines Rostellums mit seinen Organen kommt, steht die Reduktionstendenz des Gynoeceums der Apostasiaceae, durch die es zu einer Verminderung des ursprünglich trimeren Stigmas kommt, gegen- über. Biologica Nyssana, 7 (2), December 2016: 107-112. Muntele Mic Mountain is located in the southwestern part of Romania and belongs to the Southern Carpathians. Although relatively small, Muntele Mic contains most of typical mountain and high-mountain habitats. The field research regarding the orchid’s family in the Muntele Mic area, have started in the summer of 2009. Owing to easy access (asphalt road that goes to the tourist center of Muntele Mic), although it is classified as part of the European Natura 2000 network (ROSCI0126 Munţii Ţarcu), the area is influenced by negative anthropogenic factors. Although considered to be a very anthropized area, the field research concluded that there are 10 species of orchids growing in this location, of which three: Gymnadenia frivaldii Hampe ex Griseb., Dactylorhiza fuchsii (Druce) Soó and Dactylorhiza saccifera (Brongn.) Soó), were not mentioned in the literature data.
