  • Anfinsen Experiments
Lecture Notes - 2 7.24/7.88J/5.48J The Protein Folding Problem Handouts: An Anfinsen paper Reading List Anfinsen Experiments The Problem of the title refers to how the amino acid sequence of a polypeptide chain determines the folded three-dimensional organization of the chain: • Does the sequence determine the structure?? • Protein Denaturation • Emergence of the Problem • Ribonuclease A • Refolding of ribonuclease in vitro. A. Protein Denaturation/Inactivation One of the features that identified proteins as distinctive polymers was 1) Unusual phase transition in semi purified proteins When exposed to relatively gentle conditions outside the range of physiological • heat • pH • salt • organic solvents, e.g. alcohols Lets consider the familiar transition I mentioned last week; • Heat denaturation; cooking the white of an egg • Acid denaturation; Milk turning sour Macroscopic changes in bulk solution; scatters visible light, increase in viscosity - white of egg: >>heat; opaque, hard; cool down; no change: Coagulation, Aggregation, precipitation, denaturation: One of the components is egg white lysozyme: activity assay; hydrolysis of bacterial cell walls: Activity remaining vs. temperature on same axes This transition: is Denaturation. These transitions were in general found to be irreversible: activity was not recovered upon cooling: Historically the study of the folding and unfolding of proteins emerged from trying to understand this unusual change of state or phase transitions. B. Emergence of the Protein Folding Problem In the period 1958-1960, the first structure of a protein molecule - myoglobin, the oxygen binding protein of muscle - was solved by John Kendrew in 1958, followed soon after by the related tetrameric red blood cell oxygen binding protein, hemoglobin, solved by Max Perutz, both of the British Medical Research Council Labs in Cambridge, U.K.

  • Peptide Chemistry up to Its Present State
Appendix In this Appendix biographical sketches are compiled of many scientists who have made notable contributions to the development of peptide chemistry up to its present state. We have tried to consider names mainly connected with important events during the earlier periods of peptide history, but could not include all authors mentioned in the text of this book. This is particularly true for the more recent decades when the number of peptide chemists and biologists increased to such an extent that their enumeration would have gone beyond the scope of this Appendix. 250 Appendix Plate 8. Sir Peter Leitch Club AT MT SMART STADIUM, HOME OF THE MIGHTY VODAFONE WARRIORS 16th August 2017 Newsletter #183 Awesome Loyal Fans Supporting the Boys Last Sunday! For Sale: One Used League Club, Average Condition By David Kemeys Former Sunday Star-Times Editor, Former Editor-in-Chief Suburban Newspapers, Long Suffering Warriors Fan O AUCKLAND businessman Paul Davys is in talks with our owner Eric Watson about buying the club, Salthough Watson says he’s in no hurry. He’s had a stake for 17 years, so full marks for persistence, and what’s another year. Watson might be in no hurry, Davys is, saying he’ll give Watson until Friday because he needs to know one way or the other, or he'll walk away. “If he hasn't by the end of the week then it's probably better for the club and me to move on.” Davys has a chunk of the childcare company ChoiceKids, which is as far – or almost identical, depending on your viewpoint – from league as it is possible to get. Doesn’t seem to worry Davys, who says he wants to be hands on, and he is one of the many convinced the culture is wrong, and that attitudes have to improve. “There's a culture problem, fan scan see it, the coaching staff can see it, and the players can see it. But it's hard to change if you don't know how. I've got a fair idea and a fair amount of knowledge on how to build a cul- ture, so that's what I can bring.” No shortage of confidence then.

  • Canadian Olympic Activity Challenge Badminton
CANADIAN OLYMPIC ACTIVITY CHALLENGE Badminton is a racquet sport that started in India in the mid-1800s. Players use a racquet to hit a shuttlecock (also known as a “shuttle” or a “birdie”) over a net. It has been an Olympic sport since the BADMINTON Barcelona 1992 Olympic Games. Athletes compete at the singles, doubles and mixed doubles events. Badminton can be played with either two opposing players (singles) or two opposing pairs (doubles). The players or pairs stand on opposite sides of a net, and take turns serving a shuttlecock with a racquet. The players rally the shuttlecock back and forth over the net, and each side may only hit the shuttlecock once. A player or team wins a point when the shuttlecock lands on their opponent’s side of the court. A game ends when one side wins 21 points. A match is the best of three games. A shuttlecock is cone-shaped and made of overlapping feathers or plastic. The round tip is made of cork. The feathers create drag, causing the shuttle to slow down. However, when it is smashed or served, a shuttle can move faster than a tennis ball. This can make the game very fast-paced and exciting. Badminton players learn a wide variety of strokes like the forehand (like hitting with the palm) or the backhand (like hitting with the knuckles). A player learns which stroke to use in different situations. A badminton player must be very physically fit and able to move quickly in all directions. To learn more about badminton or to find a local club please visit www.badminton.ca FEATURED INDIA During the mid-1800s, India was ruled by Great Britain.

  • Black Sticks Named for Oceania Olympic Qualifier in Hobart
21 September, 2011 Media release Black Sticks named for Oceania Olympic Qualifier in Hobart The Black Sticks national coaches have named their teams for the 2011 Oceania Olympic Qualifier to be played in Hobart from 6-9 October, 2011. The Oceania region has two Olympic allocations courtesy of world rankings for both men and women and with Fiji having pulled out it gives Australia and New Zealand automatic qualification to the London Olympics. The qualifier will now be a three test series between the two rival teams. The national coaches say the qualifier will be more about getting further test experience against Australia and gaining valuable world ranking points. Black Sticks Women’s coach Mark Hager has named an experienced team, selecting the same 18 players who beat Australia in pool play and went on to win bronze at the 2011 Champions Trophy in July in Amsterdam. “This will be a hard fought series against a top team - there is so much rivalry between these two sides. With us ranked sixth and Australia ranked seventh, we will need to win the series in order to hold our position,” says Hager. Black Sticks Men’s coach Shane McLeod has chosen a very experienced men’s team with players like Phil Burrows, Blair Hopping and Dean Couzins – all who rack up a total 730 test caps. This will be the opportunity to take on world champions Australia, which will be invaluable in their build up to the Champions Trophy being held at North Harbour in early December. “This Olympic qualifier forms a key component of our preparation towards Champions Trophy and will allow us to test our game against a world champion, gold medal team.
