• 1 Male Checklist Male Reproductive System Components of the Male
Male Checklist Male Reproductive System Components of the male Testes; accessory glands and ducts; the penis; and reproductive system the scrotum. Functions of the male The male reproductive system produces sperm cells that reproductive system can be transferred to the female, resulting in fertilization and the formation of a new individual. It also produces sex hormones responsible for the normal development of the adult male body and sexual behavior. Penis The penis functions as the common outlet for semen (sperm cells and glandular secretions) and urine. The penis is also the male copulatory organ, containing tissue that can fill with blood resulting in erection of the penis. Prepuce A fold of skin over the distal end of the penis. Circumcision is the surgical removal of the prepuce. Corpus spongiosum A spongy body consisting of erectile tissue. It surrounds the urethra. Sexual excitement can cause erectile tissue to fill with blood. As a result, the penis becomes erect. Glans penis The expanded, distal end of the corpus spongiosum. It is also called the head of the penis. Bulb of the penis The proximal end of the corpus spongiosum. Bulbospongiosus muscle One of two skeletal muscles surrounding the bulb of the penis. At the end of urination, contraction of the bulbospongiosus muscles forces any remaining urine out of the urethra. During ejaculation, contractions of the bulbospongiosus muscles ejects semen from the penis. Contraction of the bulbospongiosus muscles compresses the corpus spongiosum, helping to maintain an erection. Corpus cavernosum One of two spongy bodies consisting of erectile tissue that (pl., corpora cavernosa) form the sides and front of the penis.

  • Pancreatic B-Cell Replacement: Advances in Protocols Used for Differentiation of Pancreatic Progenitors to B-Like Cells
FOLIA HISTOCHEMICA REVIEW ET CYTOBIOLOGICA Vol. 57, No. 3, 2019 pp. 101–115 Pancreatic b-cell replacement: advances in protocols used for differentiation of pancreatic progenitors to b-like cells Muhammad Waseem Ghani1, Li Ye1, Zhao Yi1, Hammad Ghani2, Muhammad Waseem Birmani1, Aamir Nawab1, Lang Guan Cun1, Liu Bin1, Xiao Mei1 1Department of Livestock Production and Management, Agricultural College, Guangdong Ocean University, Zhanjiang, Guangdong, China 2Nawaz Sharif Medical College University of Gujrat, Punjab, Pakistan Abstract Insulin-producing cells derived from in vitro differentiation of stem cells and non-stem cells by using different factors can spare the need for genetic manipulation and provide a cure for diabetes. In this context, pancreatic progenitors differentiating to b-like cells garner increasing attention as b-cell replacement source. This kind of cell therapy has the potential to cure diabetes, but is still on its way of being clinically useful. The primary restriction for in vitro production of mature and functional b-cells is developing a physiologically relevant in vitro culture system which can mimic in vivo pathways of islet development. In order to achieve this target, different approaches have been attempted for the differentiation of pancreatic stem/progenitor cells to b-like cells. Here, we will review some of the state-of-the-art protocols for the differentiation of pancreatic progenitors and differentiated pancreatic cells into b-like cells with a focus on pancreatic duct cells. • Src Regulation of Cx43 Phosphorylation and Gap Junction Turnover
biomolecules Article Src Regulation of Cx43 Phosphorylation and Gap Junction Turnover Joell L. Solan 1 and Paul D. Lampe 1,2,* 1 Translational Research Program, Fred Hutchinson Cancer Research Center, Seattle, WA 98109, USA; [email protected] 2 Department of Global Health, Pathobiology Program, University of Washington, Seattle, WA 98109, USA * Correspondence: [email protected] Received: 27 October 2020; Accepted: 22 November 2020; Published: 24 November 2020 Abstract: The gap junction protein Connexin43 (Cx43) is highly regulated by phosphorylation at over a dozen sites by probably at least as many kinases. This Cx43 “kinome” plays an important role in gap junction assembly and turnover. We sought to gain a better understanding of the interrelationship of these phosphorylation events particularly related to src activation and Cx43 turnover. Using state-of-the-art live imaging methods, specific inhibitors and many phosphorylation-status specific antibodies, we found phospho-specific domains in gap junction plaques and show evidence that multiple pathways of disassembly exist and can be regulated at the cellular and subcellular level. We found Src activation promotes formation of connexisomes (internalized gap junctions) in a process involving ERK-mediated phosphorylation of S279/282. Proteasome inhibition dramatically and rapidly restored gap junctions in the presence of Src and led to dramatic changes in the Cx43 phospho-profile including to increased Y247, Y265, S279/282, S365, and S373 phosphorylation. Lysosomal inhibition, on the other hand, nearly eliminated phosphorylation on Y247 and Y265 and reduced S368 and S373 while increasing S279/282 phosphorylation levels. We present a model of gap junction disassembly where multiple modes of disassembly are regulated by phosphorylation and can have differential effects on cellular signaling.

Wendy Yang A dissertation submitted in partial fulfillment of the requirements for the degree of Doctor of Philosophy University of Washington 2016 Reading Committee: Vincenzo Cirulli, Chair Laura Crisa Paul D. Lampe Program Authorized to Offer Degree: Pharmacology University of Washington Abstract Role for cell-to-cell communication in stem cell specification toward pancreatic progenitors: relevance to the design of novel therapies for diabetes. Wendy Yang Chair of the Supervisory Committee: Vincenzo Cirulli Metabolism, Endocrinology & Nutrition Pancreatic islets of Langerhans, responsible for the production of hormones such as insulin and glucagon, develop from pluripotent pancreatic progenitors following their specification toward an endocrine phenotype. Based on the established role of cell-to-cell communication as an important mechanism regulating developmental decisions during embryonic life, I investigated the expression pattern of Connexins (Cxs), the building blocks of Gap Junction channels, in the developing human pancreas, and in an in vitro model of pancreatic progenitor differentiation from human embryonic stem cells (hESC). I also investigated the role of β1 integrins and an associated downstream effector, integrin-linked kinase (ILK), on islet development in mice. In a first series of experiments, I investigated the expression pattern of Cxs in the developing human pancreas. Results from these studies revealed that while Cx32 is predominantly expressed in the acinar tissue, Cx36 is primarily expressed in developing islet β- cells. Cx43 exhibited the most interesting expression pattern, being primarily detected in putative islet cell progenitors that delaminate from the pancreatic ductal epithelium and aggregate with developing islet cell clusters.

  • Common Bile Duct Exploration
Education Common Bile Duct Exploration What is a common bile duct exploration? The common bile duct is a tube that connects the liver, gallbladder, and pancreas to the small intestine. It helps deliver fluids for digestion. A common bile duct exploration is a procedure used to see if a stone is blocking the flow of bile from your liver and gallbladder to your intestine. When is it used? When a stone gets stuck in the common bile duct it may cause bile to back up into the liver. This causes jaundice. Jaundice is a condition in which the skin and the whites of the eyes become yellowish. If the stone is not removed, the common bile duct may become infected and need emergency surgery. It can also cause pancreatitis, a reaction in the pancreas that can be life threatening. Common bile duct exploration is often done during surgery to remove the gallbladder. An alternative procedure is an endoscopic retrograde cholangiopancreatography (ERCP). When an ERCP is done, a tube is inserted through your mouth and stomach into the small intestine. The tube can be used to put contrast dye into the duct to look for stones with x-rays. If there are stones, a small opening is made in the common duct to allow the stone or stones to pass into the intestine. You should ask your health care provider about these choices. How do I prepare for a common bile duct exploration? Plan for your care and recovery after the operation. Allow for time to rest and try to find people to help you with your day-to- day duties.

  • New Insights Into the Secretory Functions of Brown Adipose Tissue
243 2 Journal of J Villarroya et al. Secretory functions of brown 243:2 R19–R27 Endocrinology adipose tissue REVIEW New insights into the secretory functions of brown adipose tissue Joan Villarroya, Rubén Cereijo, Aleix Gavaldà-Navarro, Marion Peyrou, Marta Giralt and Francesc Villarroya Departament de Bioquímica i Biomedicina Molecular and Institut de Biomedicina (IBUB), Universitat de Barcelona, Barcelona, Catalonia, Spain CIBER Fisiopatología de la Obesidad y Nutrición, Barcelona, Catalonia, Spain Correspondence should be addressed to F Villarroya: [email protected] Abstract In recent years, an important secretory role of brown adipose tissue (BAT) has emerged, Key Words which is consistent, to some extent, with the earlier recognition of the important f brown adipose tissue secretory role of white fat. The so-called brown adipokines or ‘batokines’ may play an f brown adipokine autocrine role, which may either be positive or negative, in the thermogenic function f batokine of brown adipocytes. Additionally, there is a growing recognition of the signalling f thermogenesis molecules released by brown adipocytes that target sympathetic nerve endings (such as neuregulin-4 and S100b protein), vascular cells (e.g., bone morphogenetic protein-8b), and immune cells (e.g., C-X-C motif chemokine ligand-14) to promote the tissue remodelling associated with the adaptive BAT recruitment in response to thermogenic stimuli. Moreover, existing indications of an endocrine role of BAT are being confirmed through the release of brown adipokines acting on other distant tissues and organs; a recent example is the recognition that BAT-secreted fibroblast growth factor-21 and myostatin target the heart and skeletal muscle, respectively.

  • Changes in Expression of P2X1 Receptors and Connexin 43 in the Rat Myometrium During Pregnancy
Changes in expression of P2X1 receptors and connexin 43 in the rat myometrium during pregnancy Tina Khanam, B.Sc., and Geoffrey Burnstock, Ph.D., D.Sc. Autonomic Neuroscience Centre, Royal Free and University College Medical School, London, United Kingdom Objective: To investigate the expression of P2X1 receptors and connexin 43 in gap junctions between smooth mus- cle cells. Contraction mediated by P2X receptors is known to occur in the bladder and male reproductive tract, and cell–cell coupling of smooth muscle via gap junctions is essential for synchronized rhythmic activity of these tissues. Design: We selected for this study rat myometrial smooth muscle during pregnancy and at postpartum day l. Setting: University medical school. Animal(s): Laboratory rats. Intervention(s): Rats were mated and became pregnant. Main Outcome Measure(s): Immunostaining and fluorescence and confocal microscopy. Result(s): The level of P2X1 receptor expression remained low throughout pregnancy (days 4 to 20) but was greatly up-regulated at day 22 (postpartum day 1). Connexin 43 expression showed a pattern of up-regulation, with progression through pregnancy and peaking near labor, but exhibited a rapid down-regulation after parturition. Conclusion(s): The functional significance of the changes in connexin 43 and P2X1 receptor expression that have been observed is discussed in relation to triggering and modulation of uterine contractility during and after preg- nancy. (Fertil SterilÒ 2007;88(Suppl 2):1174–9. Ó2007 by American Society for Reproductive Medicine.) Key Words: P2X1 receptor, connexin 43, myometrium, rat, confocal microscopy, immunofluorescence A recent article has shown that P2X1 receptors are closely as- connexins and P2 receptor-mediated processes.

PROTEIN SYNTHESIS IN THE ISOLATED GIANT AXON OF THE SQUID* BY A. GIUDITTA,t W.-D. DETTBARN,t AND MIROSLAv BRZIN§ MARINE BIOLOGICAL LABORATORY, WOODS HOLE, MASSACHUSETTS Communicated by David Nachmansohn, February 2, 1968 The work of Weiss and his associates,1-3 and more recently of a number of other investigators,4- has established the occurrence of a flux of materials from the soma of neurons toward the peripheral regions of the axon. It has been postulated that this mechanism would account for the origin of most of the axonal protein, although the time required to cover the distance which separates some axonal tips from their cell bodies would impose severe delays.4 On the other hand, a number of observations7-9 have indicated the occurrence of local mechanisms of synthesis in peripheral axons, as suggested by the kinetics of appearance of individual proteins after axonal transection. In this paper we report the incorporation of radioactive amino acids into the protein fraction of the axoplasm and of the axonal envelope obtained from giant axons of the squid. These axons are isolated essentially free from small fibers and connective tissue, and pure samples of axoplasm may be obtained by extru- sion of the axon. Incorporation of amino acids into axonal protein has recently been reported using systems from mammals'0 and fish."I Materials and Methods.-Giant axons of Loligo pealii were dissected and freed from small fibers: they were tied at both ends. Incubations were carried out at 18-20° in sea water previously filtered through Millipore which contained 5 mM Tris pH 7.8 and 10 Muc/ml of a mixture of 15 C'4-labeled amino acids (New England Nuclear Co., Boston, Mass.).
