Titre : Influence du génotype de porte-greffe dans la signalisation azotée et le développement du greffon chez la Vigne Résumé : Ce travail de recherche a eu pour objectif de caractériser l’influence du génotype de porte- greffe sur le développement du greffon notamment à travers l’étude des mécanismes relatifs à la signalisation azotée et hormonale. Dans ce contexte, deux porte-greffes, 1103 Paulsen (1103P) et Riparia Gloire de Montpellier (RGM), induisant respectivement une forte et faible vigueur du greffon, ont été étudiés. Une analyse du transcriptome racinaire a été effectuée chez des plants greffés impliquant ces deux génotypes dans un système split-root où la disponibilité en azote était hétérogène. Une réponse plus prononcée a été observée pour le génotype RGM, ainsi qu’une régulation temporelle différente entre les deux génotypes, notamment concernant des gènes clés de la réponse au nitrate intervenant dans la régulation de la croissance des racines ou dans la signalisation hormonale. D’un point de vue développemental, une régulation étroite de la production des racines latérales et des rameaux latéraux par la disponibilité en azote a été mise en évidence chez RGM. D’autre part, les propriétés intrinsèques de chaque porte-greffe semblent être conférées au greffon induisant notamment une ramification plus importante de celui-ci lorsque le porte-greffe 1103P est utilisé. La caractérisation fonctionnelle des gènes impliqués dans la voie de biosynthèse des strigolactones et la réalisation de bio-essais ont permis de mettre en évidence la production de composés de type strigolactones chez la Vigne. Ces travaux suggèrent aussi une balance contrastée entre les strigolactones et les cytokinines au sein des deux génotypes, corrélée à leur capacité de contrôle de la croissance du greffon. L’ensemble de ces résultats a permis de mettre en évidence une réponse transcriptomique et développementale à la disponibilité en azote toujours plus prononcée pour le génotype connu pour conférer le moins de vigueur au greffon, RGM. Title: Rootstock genotype impact on nitrogen signalling and development of the scion in Grapevine Abstract: This work aimed to characterize the impact of the rootstock genotype on the scion development especially through the study of the mechanisms related to nitrogen and hormonal signalling. In that context, two rootstocks were studied, 1103 Paulsen (1103P) and Riparia Gloire de Montpellier (RGM), inducing high and low scion vigour, respectively. A transcriptomic analysis was performed on roots of grafted plants involving both genotypes, cultivated in a split-root system where the nitrogen availability was heterogeneous. A more pronounced response was observed for RGM together with a different temporal regulation between both rootstocks, in particular concerning the expression of key genes of the nitrate response involved in root growth regulation or in hormonal signalling. Concerning plant development, a clear impact of the nitrogen availability on the production of lateral roots and shoot branching was highlighted for RGM. Moreover, the intrinsic properties of each rootstock seemed to be conferred to the scion inducing especially a higher shoot branching when the rootstock 1103P was used. The functional characterization of the genes involved in the strigolactone biosynthesis pathway and bioassays highlighted the production of strigolactone-like compounds in Grapevine. These experiments suggest also a contrasted balance between strigolactones and cytokinins within each rootstock genotype, correlated with their ability to control the scion growth. Taking together, these results showed a pronounced transcriptomic and developmental response to nitrogen availability for the genotype conferring the lowest scion vigour, RGM. Puis un jour, entre deux cafés, Michel Hernould me dit « Bon alors, Noé, tu es prêt à te lancer dans la recherche ? ». Je ne saurai résumer ce grand Monsieur. Fin psychologue et toujours à l’écoute de ses étudiants, il a su me donner confiance en moi. A travers cette phrase peut être banale pour lui et grâce aux nombreux conseils qu’il a su me donner, j’ai décidé de me lancer dans cette aventure que représente la Recherche. Phénotypage sans fin, voire même nocturne, expérimentations à faire le weekend, corrections d’articles, de présentations orales ou de manuscrit sans répit, tu as toujours été là pour moi. Tu as aussi été une amie avec qui on peut discuter de tout et de rien. Bien entendu, nos deux logiques bien distinctes nous ont amené à débattre au sujet de nombreuses choses mais je suis assez fier que cela ait toujours été constructif. Je te remercie donc énormément Virginie, pour tout ce que tu m’as apporté et pour ce souvenir intense et captivant que je retiendrai de ces trois années. A mon arrivée dans l’UMR Ecophysiologie et Génomique Fonctionnelle de la Vigne, un petit groupe de personnes a grandement participé à mon intégration.     Dear Sweet Briar alumnae and friends, Friendship and family have been on my mind lately, and so I want to tell you about some new friends I’ve been making this semester. One of them is Ray, who is tall and smart and also known as Love Z, and the other is Blues, who is very personable and a wonderful teacher. My friendship with Ray has grown over time; frankly, I didn’t seem to make much of an impression on him at first. My friendship with Blues blossomed immediately; we seem to be simpatico. As I joked with Merrilee “Mimi” Wroten, the director of Sweet Briar’s acclaimed riding program, maybe I bonded faster with Blues because he’s rather short, and so am I. And if you haven’t already figured it out, Ray, a chestnut warmblood, and Blues, a gray quarter horse, are members of the College’s equine family, and along with Mimi, they’ve been teaching me how to ride. As Sweet Briar’s president, it’s vital for me to learn as much as I can about the Col- lege, its programs and its people. That’s why I taught a course in our inaugural three- week session in the fall of 2018; that’s why I have just instituted collegial gatherings of faculty and staff every Monday evening (called Sweet Briar Hour); and that’s why I’m taking riding lessons, so that I can better understand our equestrian program, its ac- complishments and its needs. Riding also gives me a connection with many members of our Sweet Briar family; a full third of our students identify as riders, whether for competition or recreation.

Vine remarkably hardy, and an unusually strong grower; bunches very large and compact, sometimes shouldered, uniform: many weigh 15 to 20 ounces. Berries large, or larger than Concord; mostly round, light greenish white, semi-transparent, slightly ambered in the sun; skin thin, but tough and does not crack; quality good, has a flavor and aroma peculiarly its own; much liked by most people; very little pulp; melting and sweet to the center; parts freely from the seed, and as it never makes the tongue sore can be freely eaten by those who do not swallow grape seeds. It is very hardy; a NIAGARA vineyard owned by Thomas Beal, of Lindsay, forty miles north of Lake Ontario, stood 35 degrees below zero without injury, and his vines were loaded with a massive burden of fruit this past season. Ripens with Hartford, but hangs firmly on the vine until frost, without shrivelling or withering; enor­ mously productive and a regular bearer. A one year old vine set irf 1878 produced twenty five fine clusters 1879, -j in 1880, a large crop in 1881, in 1882 over 40 poinds of fruit, making the aggregate weight 65 pounds, produced the first four years set in the vineyard.     A History of Grape Growing in Eastern United States By Fred Elmer Gladwin PART I. (Rural New Yorker, vol. XC, no. 5156, 18 April 1931, page 485-486) Probably few records covering the introduction of any fruit in the husbandry of the United States list as many trials and many failures as do those which tell of the experiences encountered by early colonists along the Atlantic seaboard who pioneered in grape development. In fact failure followed failure. Yet after a lapse of time the project would again be revived and strenuous efforts made to avoid the earlier mistakes, if such were recognized. As we now look back on their attempts and study the causes ascribed for the failures, which are many, one can readily understand that they were groping in the dark, but with the indomitable spirit that so often crowns success in most present-day undertakings. In their day there was no recourse to carefully recorded data from institutions devoted to agricultural studies, nor were there available climatic data of temperatures and rainfall, sunshine and cloudiness, nor was the relationship of large bodies of water and vine culture associated. In view of all of the difficulties encountered, many of which increased instead of diminished, it is surprising that they still had faith in an ultimate success. Since much of this early endeavor to introduce grape-growing in the East is concerned with a species not indigenous to the United States, but rather that of the Old World, it perhaps will be well briefly to point out some of the most important differences between this species and our native American grapes.

In 1909 he was appointed a special agent of the New York State Agricultural Experiment Station (NYSAES) and put in charge of the Vineyard Laboratory at Fredonia, NY which came into existence that same year. Later he was made associate in research at the NYSAES, and in 1936 was made chief in research in Pomology, a position he held at the time of his death. His entire period of service with the Experiment Station was spent at the Vineyard Laboratory. This series of columns he authored in 1931 on the history of grape growing in the Eastern United States has been brought together here in an effort to give more prominence to a small part of his work. Daniel Chodowiecki’s allegorical copper plate of 1781 shows unauthorised reprinters and original publishers, respectively as highwaymen and their victims while the Goddess of Justice is asleep. The full title reads: ‘Works of Darkness. A Contribution to the History of the Book Trade in Germany. Presented Allegorically for the Benefit of and as a Warning to All Honest Booksellers.’ The identities of most of the characters have been identified: the bandit chief is the Austrian publisher Johann Thomas von Trattner (1717-1798) who made a fortune by reprinting books from other German- speaking territories. His victims are the publishers Friedrich Nicolai (in the centre), and Philipp Erasmus Reich (fleeing into the background). The small bat-like monster hovering overhead (a position normally reserved for angels in religious paintings!) is modelled on Gerhard van Swieten (1700-1772), an influential adviser and doctor of Maria Theresa of Austria who eased censorship regulations but encouraged the reprinting of foreign books in Austria. Nicolai’s right arm extends the bat monster’s line of gaze and points to the head of the Goddess Justitia, sleeping as if drugged by the poppy blossoms above her head.
