Abstract Cardiovascular diseases (CVD) are complex diseases where many environmental and genetic factors are involved. Due to this, their study needs to take into account different risk factors and the interactions between them, in order to reduce their incidence and mortality. Thanks to the breakthroughs of the Hap Map and Human genome projects and the development of GWAS and PheWAS, the genetic etiology of the CVD has been extensively investigated the last two decades. However, there is still a big room of knowledge waiting to be discovered and alternative approaches are needed in order to keep advancing in the pathophysiology of CVD. In this thesis, we propose an integrative approach to discover new genetic associations potentially involved in CVD. We chose previous GWAS hits and we focused on phenotypically homogeneous populations, in order to limit as much as possible the heterogeneity of intermediate phenotypes. We also centred our efforts in studying the pleiotropic and gene-gender interaction effects of the genes selected. Another main goal during this thesis was to defend the implementation of personalized genome-based therapy of the results obtained. By using the above-explained approaches, new pleiotropic effects were discovered in the IL- 6R and ABO genes. Within the IL-6R gene we found an antagonistic pleiotropy of this gene when it relates to CRP and lipid trait levels. We also found new pleiotropic effects within the ABO gene, by independently associating the major rs644234*T allele with increased sE- selectin and HDL levels, and decreased ApoE levels. Detection of pleiotropy is a source of new hypothesis and makes easier the development of new drugs, hence improving prevention and personalized medicine within the CVD. In addition, we studied the gene-gender interaction effects, finding some sex-specific associations in two of the genes studied (ABO and GNB3). Further, we centered our efforts in implementing the results obtained during the thesis at the clinical level, with the aim of ameliorating the genetics of personalized medicine. The SNP rs2234246 within the TREM-1 gene was associated with increased levels of its protein and could be used as a predictor or risk biomarker for different diseases (including CVD). Indeed, and due to the high potential of this polymorphism, we applied a European patent and we are planning to start Phase I clinical trials in patients suffering from Septic Shock and Acute Myocardial Infarction. Also the IL-6R haplotype rs4845628*T/rs4537545*C, which is increasing simultaneously CRP, LDL-C and ApoB levels, could be used in the treatment of personalized medicine. One of the side effects of the drug Tocilizumab, which is targeting the IL-6R gene, is the increase of LDL-C levels in patients, which could lead to a risk of suffering from ischaemic heart disease. In order to prevent these side effects, patients carriers of the haplotype rs4845628*T/rs4537545*C could be treated by analogue drugs or different doses of tocilizumab, which could make the treatment more personalized and hence, more secure and precise. Following the goals and approaches synthetized above, we discovered new associations between genes of interest and intermediate phenotypes that are involved in CVD and other complex diseases. Our results help to better understand how the studied genes are exerting their effects at the molecular level, ultimately affecting the outcome of the individuals suffering from CVD. Our results will hopefully be taken into account in future personalized treatments. Résumé Les maladies cardiovasculaires (MCV) sont d’une étiologie complexe et elles sont soumises à de nombreux facteurs environnementaux ainsi que génétiques. Pour pouvoir mieux comprendre leur pathophysiologie et réduire leur incidence et leur mortalité, l’étude des MCV prend en compte beaucoup de facteurs de risques différents en même temps, ainsi que leurs interactions. Les dernières percées technologiques dans le cadre du Projet Génome Humain, ou du Projet HapMap ont permis de développer de nouvelles approches méthodologiques. Les études d’association pangénomiques ou GWAS et les études d’association panphénotypiques ou PheWAS ont révolutionné l’épidémiologie génétique de la dernière décennie. The Architecture of Joseph Michael Gandy (1771-1843) and Sir John Soane (1753-1837): An Exploration Into the Masonic and Occult Imagination of the Late Enlightenment Abstract In examining select works of English architects Joseph Michael Gandy and Sir John Soane, this dissertation is intended to bring to light several important parallels between architectural theory and freemasonry during the late Enlightenment. Both architects developed architectural theories regarding the universal origins of architecture in an attempt to establish order as well as transcend the emerging historicism of the early nineteenth century. There are strong parallels between Soane's use of architectural narrative and his discussion of architectural 'model' in relation to Gandy's understanding of 'trans-historical' architecture. The primary textual sources discussed in this thesis include Soane's Lectures on Architecture, delivered at the Royal Academy from 1809 to 1836, and Gandy's unpublished treatise entitled the Art, Philosophy, and Science of Architecture, circa 1826.

Hobbs Professor Gerald Prince The battlefield crucifixes that lined the Western Front powerfully connected industrialized warfare with the Christian past. This elision of the bloody corporeality of the crucifixion with the bodily suffering wrought by industrial warfare forged a connection between religious belief and modern reality that lies at the heart of my dissertation. Through the poignancy of Christ’s suffering, French Catholics found an explanatory tool for the devastation of the Great War, affirming that the blood of ther F ench dead would soon blossom in rich harvest. This dissertation argues that the story of French Catholicism and the Great War uncovers a complex and often dissonant understanding of the conflict that has become obscured in the uniform narrative of disillusionment and vain sacrifice ot emerge in the last century. Considering the thought to emerge from the French renouveau catholique from 1910 up to 1920, I argue that far from symbolizing the modernist era of nihilism, the war in fact created meaning in a world that had lost touch with its God.

Piper 2016 Abstract This dissertation explores spatio-temporal shifts in twenty-first century French crime narratives, through a series of close readings of contemporary crime films, television, literature, and comics. The works examined rely on the formal properties of the policier genre but adapt its standard conventions, most notably with deviations in the use and function of space. In this dissertation, I demonstrate that the modern policier is one that embraces its spatio-temporal, social, and generic non-fixity. The textual/visual constructions of many hyper-contemporary crime narratives contain multiple modes of decomposition within: a decentralization of space, which moves the action away from the genre’s traditionally urban location to boundless rural spaces and border zones; a de- concentration of the policier genre, through the incorporation of tropes from other literary styles and works; and a devolution of social cohesion and community identity in the narratives. Chapter 1 examines works where historic references and urban legends of the 19th century fantastique literary genre unfold in modern rural locations. The past and the present converge to problematize modern ideals, identity, and community unity in rural spaces where reason is pitted against the supernatural.

    ABSTRACT Title of dissertation: PROUST AND THE DISCOURSE ON HABIT Amy Ross Loeserman, Doctor of Philosophy , 2004 Dissertation directed by: Professor Pierre Verdaguer Department of French and Italian A la recherche du temps perdu b y Marcel Proust is replete with a discourse by the principal character, the narrator Marcel, on the subject of habit (“habitude” in French). This discourse meticulously explores the ubiquitous but concealed role that habit plays with respect to the most s ignificant aspects of life, such as emotions, cognitive processes, and aesthetic experiences, and it explicitly relates not only to the novel’s characters, but to humanity in general. The critical commentary on the novel has largely ignored this subject. This dissertation provides the only comprehensive collection and analysis of the Proustian commentary on habit in A la recherche du temps perdu. It is not by chance that habit was deeply explored in Proust’s novel or that it has been largely overlooked by the critical commentary. Historically, philosophers have paid substantial attention to habit. Habit was a focus of controversial philosophical/psychological theories in 19th century France regarding memory and consciousness, spirit and matter. Pro ust’s commentary was directly related to the prominent philosophical issues of his time. This dissertation discusses the broad meanings of habit, first as developed by Aristotle and St. Thomas; then by French essayists, through Montaigne, Pascal, and the philosophes ; and finally culminating in the great 19th century works on habit by Maine de Biran and Félix Ravaisson. It also reviews substantial contributions on habit made by other French writers and philosophers, notably Stendhal and Alfred Fouillée.

To understand media, we should start not on land but at sea. Te sea has long seemed the place par excellence where history ends and the wild be- gins: the abyss, a vast deep and dark mystery, unrecorded, unknown, un- mapped. Melville called the sea “Inviolate Nature primeval.” It has long been a profoundly unnatural environment for humans in both life and in thought. Seventy- one percent of the earth’s surface has been a sub- lime, uncanny place without limits and beyond understanding, the ulti- mate wasteland. Te ocean was once roiling with dragons, Leviathans, and pirates— a merciless mix of fate, wind, and weather that imperiled anyone brave or foolish enough to risk their life on ship. It is still a very dangerous place, a kind of planetary waste dump and graveyard for many forms of life, including hapless immigrants. Only recently have humans dipped much below its surface, with depth exploration historically having been limited to the shoreline. Both Babylonian and Hebrew ori- gin myths describe creation as the conquest of chaotic uncreated waters (tiamat, tehom). Te Book of Revelation, at the opposite end of the Bible from Genesis, seals this conquest by announcing a new heaven and earth 53 54 CHAPTER TWO in which the sea is no more, abolished as if in a fnal act of spite (Revela- tions 21:1).
