Les trois récepteurs sont exprimés par les macrophages au sein de la lésion d’athérosclérose humaine. Cependant, l’expression et le rôle de NOR1 dans les macrophages alternatifs n’ont pas encore été étudiés. En utilisant le modèle in vitro de macrophages primaires alternatifs humains polarisés en présence d’IL-4, nous avons démontré que l’expression de NOR1 est induite dans les macrophages M2, une régulation pas observée chez la souris. D’ailleurs, l’expression de NOR1 est plus importante dans les zones de la lésion athérosclérotique humaine enrichies en macrophages alternatifs CD68+MR+. En utilisant l’approche de diminution de l’expression de NOR1 par un siRNA spécifique, nous démontrons que l’expression de certains marqueurs de la polarisation alternative tels que Mannose Receptor (MR), Interleukin-1 receptor antagonist (IL1Ra), CD200 receptor (CD200R), coagulation factor XIII A1 polypeptide (F13A1), interleukin 10 (IL10) et Peroxisome Proliferator- Activated Receptor (PPAR) est diminuée. Des analyses bioinformatiques ont identifié les gènes F13A1, IL-1Ra, IL-10 et la matrix metallopeptidase 9 (MMP9), comme étant des cibles potentielles de NOR1 dans les macrophages alternatifs humains. De plus, l’expression et l’activité enzymatique de la MMP9 sont induites suite au silencing de NOR1, une régulation confirmée par l’approche de surexpression de NOR1 par infection adénovirale.

The Cellular Mirna, Mir-190, is Upregulated in Type I Ebv Latency by Ebers and Modulates Cellular Mrnas involved in Cell Survival and Viral Reactivation Abstract Epstein-Barr Virus (EBV) is a highly prevalent human pathogen infecting over 90% of the population. Much of the success of the virus is attributed to its ability to maintain latency through different programs in host cells. MicroRNAs (miRNA) are small, non-coding RNAs capable of post-transcriptionally regulating mRNA expression. A microarray comparison of EBV type I latency and type III latency infected cells yielded evidence of differential cellular microRNA expression. I hypothesized that one of these differentially upregulated type I latency miRNAs, miR-190, is important in maintenance of latency I, and miR-190 upregulation is due to viral gene expression.

Örjans group joined us during summer whereas Mikaels group will move physically to their new laboratory, hopefully before summer 2017. During 2016 a position as a University Lecturer in Computational Genomics was filled. Örjan, who comes from SLU, was ranked as number one by the expert reviewers and was offered the position. With all the positive comments from the expert reviewers we took the opportunity to also offer Matthew Webster, who came in at second place, a position as a University Lecturer in Computational Genomics. Thus, in one stroke the number of permanent staff at the Genomic section at IMBIM doubled. Mikael Sellin was recruited as an Associate Senior Lecturer “biträdande universitetslektor (BUL)” in Medical Infection Biology sponsored by the SciLifeLab Uppsala. Mikael was ranked as number one among fifteen national and international applicants. We were pleased that Mikael decided to place his laboratory at IMBIM. BUL positions are a new type of career development positions. In fact, Mikael is the first BUL appointed at the Medical Faculty. Mikael started to work with us in December 2016 but his group will physically move to the B9:3 corridor during spring 2017.
