Characterization of Wines Produced from Fungus Resistant Grapes
Characterization of Wines Produced from Fungus Resistant Grapes Claudia Koraimanna, Erich Leitnera, Uwe Oppermannb a Graz University of Technology, [email protected], Graz, Austria b Shimadzu Europe GmbH, [email protected], Duisburg, Germany Introduction The vineyards of the Styrian wine region are located in the southern part of Austria close to the Slovenian border covering approximately 4.500 hectares. Predominantly steep slopes are cultivated with inclinations up to 60%. This area is a classical cool-climate region with an average annual temperature of 10.5°C and precipitation of approximately 1.000 mm per year. Main rainfall months are especially May, June, July & ,August and September in which 55 to 70% of the annual precipitation falls. The mechanical management of the steep slopes is difficult and quite dangerous, too. The establishment of fungus- resistant grape varietals can clearly reduce plant protection efforts and also brings economic advantages. First experiments with fungus resistant grape varieties date back to the early '80s. Most varieties of the initial trials were not accepted by the consumers due to their sensory properties. The recent generation of fungus-resistant grape varietals show much more compromising results and a higher acceptance on the market. Sensory Analysis Analytical Methods Sensory analysis was performed by a trained panel with specific Aroma compounds were analyzed by HS-SPME using a 2 cm 50/30 µm expertise in wine tasting consisting of 12 members. All panelists DVB/Carboxen/PDMS fiber with different techniques and parameters. The aroma are members of the Austrian Wine quality board which is substances except Isobutylmethoxypyrazine were quantified by the standard responsible for the Austrian wine quality banderol, where a addition procedure with GC-MS (SIM) at 5 different concentration levels adjusted to sensory judgment for each quality wine is mandatory. -
A Molecular Marker Set Combining a Retrotransposon Insertion and SSR Polymorphisms Is Useful for Assessing Diversity in Vitis
Received: 17 August 2020 y Accepted: 11 May 2021 y Published: 25 June 2021 DOI:10.20870/oeno-one.2021.55.2.4473 A molecular marker set combining a retrotransposon insertion and SSR polymorphisms is useful for assessing diversity in Vitis Frédérique Pelsy, Lucie Bevilacqua, Sophie Blanc and Didier Merdinoglu Université de Strasbourg, INRAE, SVQV UMR-A 1131, F-68000 Colmar, France *corresponding author: [email protected] Associate editor: Laurent Jean-Marie Torregrosa ABSTRACT Molecular markers, based on DNA polymorphisms, are useful tools for identifying individuals, establishing phylogenetic relationships, managing collections of genetic material or assisting breeding. In the present study, we developed a marker set to differentiate Vitis species, grapevine varieties or clones belonging to the same variety. This novel marker set combines, in four PCR amplifications, the presence/absence of a remarkable retrotransposon, Tvv1-Δ3460, inserted at its single locus and the SSR polymorphism present within its two LTRs. By studying a collection of Vitaceae accessions, we showed the prevalence of two allelic forms of Tvv1-Δ3460 - one of which was partially truncated - in Vitis species. Out of the twenty-five studiedVitis species, the insertion of a Tvv1-Δ3460 element was detected in twenty, including Vitis vinifera. The homozygous vs heterozygous state of the element insertion was determined by amplifying the empty site. Additionally, each Tvv1-Δ3460 LTRs included a microsatellite sequence useful for designing markers based on LTR length. The LTR-SSR markers distinguished most of the fifty-two cultivars and revealed polymorphism within five of the seven varieties studied. Pinot Pedigree Diagram
PINOT PEDIGREE DIAGRAM No fewer than 156 western European grape varieties make up this huge pedigree of natural crosses, with savagnin, gouais blanc and pinot playing the major roles. Given the number of varieties that are currently unknown (?), this pedigree is just one of the possible interpretations of all direct parent–offspring relationships that have been discovered by DNA parentage analyses (Pinot and teroldego, which have a grandparent–grandchild relationship, are included to illustrate the unexpected link between Pinot and syrah). It strongly challenges the commonly assumed independent origins of western European varieties and argues in favour of a small core set of founder varieties that have given birth through natural crosses to the significant biodiversity we know today. For the sake of clarity, trebbiano toscano and folle blanche appear twice in the diagram. Gatefold_Pinot.indd 1 23/07/2012 15:10 ? Gänsfüsser ? Chatus ? César ? ? ? Abstract : Two specialists, NEGRUL (1946) et LEVADOUX (1948) proposed for Europe vine-plants cultiva- ted a classification in natural groups issued from ecogeographical areas relatively closed in their situations and evo- lutions. These ampelographs are complementary in their propositions, Russian for Eastern Europe and French for Western forms. Middle-East is the probable origin of mediterranean viticulture with its vine-varieties then the local wild forms in more septentrional vineyard extensions. After sortotypes or ecogeogroups definitions, anterior structural propositions of French vine-varieties in eco-geo- graphical groups are developped by individual integration forms in taxonomic and morphologic unities. The pre- sent propositions are not immovable nor definitive but as such they may be useful in recent genetic advancements. They can confirm or not some new biochemical diagnosis.
