A mis padres Agradecimientos Llegado este momento y dado que la lectura de los agradecimientos siempre parece que suscita un interés especial voy a intentar aunar esfuerzos para recordar todo lo vivido y compartido a lo largo de estos años. Y es que, a pesar de pertenecer al mundo de la ciencia y de la tecnología, la vida está llena de vivencias y emociones y supongo que, será por ello, por lo que en los últimos meses me han animado diciendo “Venga Ana, que ya se ve la luz al final del túnel”, y es que esta tesis como una parte importante de mi vida ha estado llena de luces y sombras. Entre las sombras, la incertidumbre ante un nuevo experimento, ensayos que no salen a la primera a pesar de que sus autores en los artículos de referencia no señalan ningún contratiempo, equipos que se rompen en el momento en el que necesitas utilizarlos y un sin fin más de cosas, que a día de hoy, una vez completado el trabajo se empiezan a ver desde otra perspectiva. Las luces por el contrario sois todos vosotros, los que de una forma u otra habéis compartido conmigo esta experiencia. Gracias a mis directoras de tesis, Puri y Marta, por haber confiado en mí desde el principio, por guiarme y adentrarme en el mundo de la investigación, por compartir conmigo vuestro bagaje científico y personal. El aprendizaje obtenido seguro que en el futuro forma implícitamente parte de mi personalidad. Gracias a todos mis compañeros de laboratorio, a los que estaban incluso antes de que yo llegara. David, es difícil agradecerte en palabras todo el apoyo que he recibido de tu parte a lo largo de estos años. Gracias por hacerme fácil lo difícil, gracias por tu tiempo, por enseñarme en los inicios, por tu paciencia y por tu amistad. Álvaro, gracias por hacer más distendidas las horas de trabajo en el laboratorio y en el despacho, que sepas que me he convertido en toda una experta resolviendo jeroglíficos. Pilar, gracias por tu disponibilidad, por ser flash resolviendo dudas y contestando mails, no sabes lo que eso se valora. Lauri, gracias por ser una amiga, por contagiar buen rollo, encontrarse con un carácter como el tuyo en el laboratorio hace más llevadero cualquier trabajo a realizar. Gracias por tu apoyo y tu escucha desde el principio de los principios. Chini, no sabía bien donde ubicarte, porque eres de los de antes, de los de ahora y de los de siempre, así que nunca mejor dicho, gracias por estar siempre ahí con una sonrisa. Gracias a todos los compañeros que estaban de paso pero que han dejado una huella imborrable a su marcha. Gracias a Celia, fue corto pero intenso, es difícil compenetrarse también en tan poco tiempo, solo es posible con un carácter tan cercano como el tuyo. Gracias por tu energía positiva, por tu alegría, por tu saber escuchar y comprender y por tener ese gran corazón. Tao, yīqiān xie xie, por qué? Pues por todo, porque aún no me explico como fuimos capaces de entendernos a la perfección hablando en distinto idioma. Gracias por contagiarme de alegría, por transmitirme parte de tu filosofía de vida oriental, que sepas que me ha servido de mucha ayuda a lo largo de estos años. Gracias por enseñarme tu cultura y por estar siempre a mi lado a pesar de la distancia. En definitiva, gracias por ser tan buen amigo. A Elisa, gracias por tu amistad, dulzura y generosidad, espérame en Cerdeña que en una de estas me dejo caer por allí. Ruti, gracias por las risas y confidencias que hemos compartido tanto en la uni como fuera, tu forma de ser resume lo buena gente que eres. Crespi, gracias por abrirme los ojos, por fin sé “quién es mi papi”☺. Gracias por tu buen rollo, por saber aunar fiesta de noche y minuciosidad en el labo durante el día, eres todo un crack! Y finalmente, y no por ello menos importante, gracias a los compañeros que me han acompañado hasta el final. Cascan, nos unió la enología y al final el vino y la química han seguido en nuestras vidas. Gracias por tus consejos, tus ánimos, por ofrecerme tu ayuda siempre que lo he necesitado y por ser un amigo. Marivel, vales un montón y por eso creo que he aprendido tanto contigo, como diría José Miguel, incluso hasta hablo un poco de guacamayo. Muchas gracias por tener siempre un ratito para mi y por los ratos de cháchara que hemos pasado, como a ninguna de las dos nos gusta hablar…A Pilar y Cristina, compañeras de labo y de despacho, porque tanto los buenos momentos como los días en que parece que se tuerce todo, cuando se comparten son más llevaderos y vosotras habéis estado ahí. The contribution by the numerous grape-must-associated non-Saccharomyces yeasts to wine fermentation has been debated extensively. These yeasts, naturally present in all wine fermentations, are metabolically active and their metabolites can impact on wine quality. Although often seen as a source of microbial spoilage, there is substantial contrary evidence pointing to a positive contribution by these yeasts. The role of non-Saccharomyces yeasts in wine fermentation is therefore receiving increasing attention by wine microbiologists in Old and New World wine producing countries. Species that have been investigated for wine production thus far include those from the Candida, Kloeckera, Hanseniaspora, Zygosaccharomyces, Schizosaccharomyces, Torulaspora, Brettanomyces, Saccharomycodes, Pichia and Williopsis genera. In this review the use and role of non-Saccharomyces yeast in wine production is presented and research trends are discussed. This is of concern to white wine producers and frning with bentonite to remove protein is an established winery practice. However, such methods are inconvenient, non-specific and can result in extensive flavour loss. In this study, proteins responsible for hazp, have been isolated and characterized, and their interaction with wine polysaccharides has been explored. An examination of the use of peptidases to remove such proæins was also undertaken. To get a reliable estimate of the concentration of grapo juice and wine proteins it was necessary to determine their amino acid composition. The commonly used Bradford dye- binding assay grossly underestimates the protein concentration by 50 to 807o and in addition, grape and wine phenolic compounds were found to strongly interfere.

HEATHERBELL 2 The effect of bentonite fining on the total proteins and the heat-unstable (80°C, 6 h; 4°C, 12 h) proteins in GewLirztraminer, White Riesling, Sauvignon blanc, and Sylvaner wines was investigated. Protein molecular weights (MW), isoelectric points (pl), and glycoproteins were determined by using lithium dodecyl sulfate polyacrylamide gel electrophoresis (LDS PAGE) and two-dimensional isoelectric focusing-lithium dodecyl sulfate (IF-LDS) electrophoretic techniques with silver staining as well as protein blotting for glycoprotein detection. Rel- ative concentrations of proteins in stained gels were determined by laser scanning densitometry. Bentonite fining tends to remove the higher pl (5.8- 8.0) and intermediate MW (32 000- 45 000) protein fractions first. How- ever, these represent only a small proportion of the soluble proteins. In general, it is necessary to remove the lower pl (4.1 - 5.8), lower MW (12 600 and 20 000- 30 000) fractions, which contain glycoproteins and represent the major component of the proteins, to protein stabilize wines to heat testing. Protein fractions with MW of 60 000 to 65 000 and having a wide range of pl (4.1 - 8.0) were highly resistant to removal by bentonite fining and remained in protein-stabilized wine. In addition, trace amounts of fractions with MW of 28 000 may remain in GewiJrztraminer and White Riesling wines and 25 000 in Sauvignon blanc wines. Unstable proteins precipitated by heat tests were recovered and analyzed.

The contribution by the numerous grape-must-associated non-Saccharomyces yeasts to wine fermentation has been debated extensively. These yeasts, naturally present in all wine fermentations, are metabolically active and their metabolites can impact on wine quality. Although often seen as a source of microbial spoilage, there is substantial contrary evidence pointing to a positive contribution by these yeasts. The role of non-Saccharomyces yeasts in wine fermentation is therefore receiving increasing attention by wine microbiologists in Old and New World wine producing countries. Species that have been investigated for wine production thus far include those from the Candida, Kloeckera, Hanseniaspora, Zygosaccharomyces, Schizosaccharomyces, Torulaspora, Brettanomyces, Saccharomycodes, Pichia and Williopsis genera. In this review the use and role of non-Saccharomyces yeast in wine production is presented and research trends are discussed.
