CHONGQING MACHINERY & ELECTRIC CO., LTD Annual Report 2020 Results Highlights Results highlights of Chongqing Machinery & Electric Co., Ltd. (the “Company” or “Chongqing Machinery & Electric”) and its subsidiaries (collectively the “Group”). The revenue of the Group for the year ended 31 December 2020 amounted to approximately RMB6,367.0 million, representing an increase of approximately RMB850.2 million or approximately 15.4% as compared with approximately RMB5,516.8 million for the same period of 2019. Profit attributable to the shareholders of the Company for the year ended 31 December 2020 was approximately RMB183.0 million, representing a decrease of approximately RMB1.8million or approximately 1.0% as compared with approximately RMB184.8 million for the same period of 2019. Basic earnings per share for the year ended 31 December 2020 amounted to approximately RMB0.05 (2019: approximately RMB0.05). The board of directors (the “Board”) of the Company proposed to declare a final dividend of RMB0.01 per share (tax inclusive) for the year ended 31 December 2020 (2019: RMB0.01 (tax inclusive)). 6 CHONGQING MACHINERY & ELECTRIC CO., LTD Annual Report 2020 Chairman’s Statement Dear Shareholders, On behalf of the Board, I am pleased to announce the annual results of the Group for the year ended 31 December 2020 (the “Period” or the “Year”).

    www.nature.com/scientificreports OPEN Role of PARP1‑mediated autophagy in EGFR‑TKI resistance in non‑small cell lung cancer Zhimin Zhang1,4, Xiaojuan Lian2,4, Wei Xie1, Jin Quan2, Maojun Liao1, Yan Wu3, Zhen‑Zhou Yang3,4* & Ge Wang1,4* Resistance to epidermal growth factor receptor‑tyrosine kinase inhibitors (EGFR‑TKIs) has become the main clinical challenge of advanced lung cancer. This research aimed to explore the role of PARP1‑ mediated autophagy in the progression of TKI therapy. PARP1‑mediated autophagy was evaluated in vitro by CCK‑8 assay, clonogenic assay, immunofuorescence, and western blot in the HCC‑827, H1975, and H1299 cells treated with icotinib (Ico), rapamycin, and AZD2281 (olaparib) alone or in combination. Our results and GEO dataset analysis confrmed that PARP1 is expressed at lower levels in TKI‑sensitive cells than in TKI‑resistant cells. Low PARP1 expression and high p62 expression were associated with good outcomes among patients with NSCLC after TKI therapy. AZD2281 and a lysosomal inhibitor reversed resistance to Ico by decreasing PARP1 and LC3 in cells, but an mTOR inhibitor did not decrease Ico resistance. The combination of AZD2281 and Ico exerted a markedly enhanced antitumor efect by reducing PARP1 expression and autophagy in vivo. Knockdown of PARP1 expression reversed the resistance to TKI by the mTOR/Akt/autophagy pathway in HCC‑827IR, H1975, and H1299 cells. PARP1‑mediated autophagy is a key pathway for TKI resistance in NSCLC cells that participates in the resistance to TKIs. Olaparib may serve as a

Chongqing is situated in the east of southwest China, about 2,500km up the Yangtze River from Shanghai. Under its jurisdiction there are 40 districts, cities and counties. It covers an area of 82,000 square kilometres with a total population of 31 million. An estimated 6 million people live in urban Chongqing city. Downtown Chongqing lies at the point where the Yangtze River and the Jialing River merge. Known as the Mountain City, the whole city is built against a backdrop of hills and rivers, characterized by zig-zagging roads and overlapping houses. It is also known as one of the four Furnace Cities for its hot summers and the Foggy City for its misty winters.
