Accepted 23-III-2021. ABSTRACT Introduction: Rhinella schneideri is a toad widely distributed in South America and its poison is characterized by inducing cardiotoxicity and neurotoxicity. Objective: In this work, we investigated pharmacological strate- gies to attenuate the peripheral neurotoxicity induced by R. schneideri poison in avian neuromuscular prepara- tion. Methods: The experiments were carried out using isolated chick biventer cervicis preparation subjected to field stimulation for muscle twitches recordings or exposed to acetylcholine and potassium chloride for contrac- ture responses. Results: Poison (10 μg/ml) produced complete neuromuscular blockade in chick biventer cervi- cis preparation within approximately 70 min incubation (times for 50 and 90 % blockade: 15 ± 3 min and 40 ± 2 min, respectively; P < 0.05, N= 5); contracture responses to exogenous acetylcholine and KCl were unaffected by poison indicating no specificity with postsynaptic receptors or myotoxicity, respectively. Poison (10 μg/ml)- induced neuromuscular blockade was not prevented by heparin (5 and 150 IU/ml) under pre- or post-treatment conditions. Incubation at low temperature (23-25 °C) abolished the neuromuscular blockade; after raising the temperature to 37 °C, the complete neuromuscular blockade was slightly slower than that seen in preparations directly incubated at 37 °C (times for 50 and 90 % blockade: 23 ± 2 min and 60 ± 2.5 min, respectively; P < 0.05, N= 4). Neostigmine (3.3 μM) did not reverse the neuromuscular blockade in BC preparation whereas 3,4-diaminopyridine (91.6 μM) produced a partial and sustained reversal of the twitch responses (29 ± 7.8 % of maximal reversal reached in approximately 40 min incubation; P < 0.05, N= 4). Conclusions: R. schneideri poison induces potent peripheral neurotoxicity in vitro which can be partially reversible by 3,4-diaminopyridine.     www.nature.com/scientificreports OPEN The impact of multiple climatic and geographic factors on the chemical defences of Asian toads (Bufo gargarizans Cantor) Yueting Cao, Keke Cui, Hongye Pan, Jiheng Wu & Longhu Wang* Chemical defences are widespread in nature, yet we know little about whether and how climatic and geographic factors afect their evolution. In this study, we investigated the natural variation in the concentration and composition of the main bufogenin toxin in adult Asian toads (Bufo gargarizans Cantor) captured in twenty-two regions. Moreover, we explored the relative importance of eight climatic factors (average temperature, maximum temperature, minimum temperature, average relative humidity, 20–20 time precipitation, maximum continuous precipitation, maximum ground temperature, and minimum ground temperature) in regulating toxin production. We found that compared to toads captured from central and southwestern China, toads from eastern China secreted higher concentrations of cinobufagin (CBG) and resibufogenin (RBG) but lower concentrations of telocinobufagin (TBG) and cinobufotalin (CFL). All 8 climatic variables had signifcant efects on bufogenin production (ri>0.5), while the plastic response of bufogenin toxin to various climate factors was highly variable. The most important climatic driver of total bufogenin production was precipitation: the bufogenin concentration increased with increasing precipitation. This study indicated that the evolution of phenotypic plasticity in chemical defences may depend at least partly on the geographic variation of defensive toxins and their climatic context. Chemical defences is ubiquitous in nature and can serve for deterring predators, competitors, parasites, and path- ogens1–3. Many species can secrete a large diversity of toxic defensive compounds for self-protection.

In this review, we present a new theory of the etiology and pathogenesis of preeclampsia that is based on the interaction of Na/K-ATPase and its endogenous ligands in- cluding marinobufagenin. The signaling pathway of marinobufagenin involves an inhibition of transcriptional factor Fli1, a negative regulator of collagen synthesis, followed by the deposition of collagen in the vascular tissues and altered vascular functions. Email: [email protected] Received: 09 Jul 2013, Revised and Accepted: 14 Aug 2013 ABSTRACT Bufadienolides are a type of cardiac glycoside originally isolated from the traditional Chinese drug Chan’Su which increases the contractile force of the heart by inhibiting the enzyme Na+/K+–ATPase. They also show toxic activities to livestock. They are widely used in traditional remedies for the treatment of several ailments, such as infections, rheumatism, inflammation, disorders associated with the central nervous system, as antineoplastic and anticancer component. Structural changes in functionality could significantly alter their cytotoxic activities. The novel oxy-functionalized derivatives of bufalin obtained could provide new platforms for combinatorial synthesis and other further investigations for the development of new bufadienolides antitumor drugs. In this review, naturally occurring bufadienolides which are isolated from both plant and animal sources are reviewed and compiled with respect to their structural changes and medicinal utility.     Herpetologia no Brasil II Princípios bioativos da pele de anfíbios: panorama atual e perspectivas Carlos Alberto Schwartz, Mariana de Souza Castro, Osmindo Rodrigues Pires Júnior, Natan Medeiros Maciel, Elisabeth Nogueira Ferroni Schwartz e Antonio Sebben Resumo A secreção cutânea dos anfíbios contém uma infinidade de substâncias biologicamente ativas empregadas em três funções principais: proteção contra o desenvolvimento de microrganismos patogênicos, como defesa passiva contra o ataque de predadores carnívoros, e, ainda, como feromônios. Considerando as espécies conhecidas na atualidade, o número de espécies que tiveram secreções cutâneas estudadas até o presente é estimado em menos de 20% do total. Mesmo nessas, o conhecimento a respeito da composição química e das propriedades farmacológicas das secreções cutâneas é apenas parcial, o que indica que mesmo as espécies já estudadas devem ser pesquisadas à luz das novas metodologias disponíveis. No presente artigo, apresentamos o panorama atual dessa área, considerações a respeito de problemas metodológicos enfrentados e suas perspectivas. Palavras-chave: Toxinas, Venenos, Amphibia, Antibióticos, Secreção Cutânea. Abstract The amphibian skin secretions contain a great variety of biologically active compounds used in three main functions: protection against the development of microorganisms, passive defense against predators and, even as pheromones. Considering the species currently known, the number of species that had the skin secretion studied till now answer less than 20% of the total. Even in these species, the knowledge about the chemical composition and the pharmacological properties are just partial. Therefore the species already studied must be evaluated in the light of recent methodologies. Herein we present the currently outlook in these multidisciplinary area, considerations concerning the methodological problems, and perspectives.
