El objeto de esta Tesis Doctoral ha sido el estudio taxonómico del género Centaurea, cuya diversidad y complejidad en el territorio es alta, por lo que se ha restringido a la sección Seridia (Juss.) DC. y, atin así, el estudio ha requerido 4 años de dedicación para su finalización. La iniciativa se inscribe en el Proyecto Flora iberica, financiado en la actualidad por el Ministerio de Economía y Competitividad. El estudio ha entrañado la realización de numerosas prospecciones en el campo, necesarias para poder abordar aspectos importantes, tales como los estudios cariológicos, palinológicos y moleculares, todos encaminados a apoyar la slntesis taxonómica, que ha requerido además de un exhaustivo estudio de material conservado en herbarios nacionales e internacionales. -

www.nature.com/scientificreports OPEN Evaluation of drought resistance and transcriptome analysis for the identifcation of drought‑responsive genes in Iris germanica Jingwei Zhang1, Dazhuang Huang1*, Xiaojie Zhao1 & Man Zhang2 Iris germanica, a species with very high ornamental value, exhibits the strongest drought resistance among the species in the genus Iris, but the molecular mechanism underlying its drought resistance has not been evaluated. To investigate the gene expression profle changes exhibited by high‑ drought‑resistant I. germanica under drought stress, 10 cultivars with excellent characteristics were included in pot experiments under drought stress conditions, and the changes in the chlorophyll (Chl) content, plasma membrane relative permeability (RP), and superoxide dismutase (SOD), malondialdehyde (MDA), free proline (Pro), and soluble protein (SP) levels in leaves were compared among these cultivars. Based on their drought‑resistance performance, the 10 cultivars were ordered as follows: ‘Little Dream’ > ‘Music Box’ > ‘X’Brassie’ > ‘Blood Stone’ > ‘Cherry Garden’ > ‘Memory of Harvest’ > ‘Immortality’ > ‘White and Gold’ > ‘Tantara’ > ‘Clarence’. Using the high‑drought‑resistant cultivar ‘Little Dream’ as the experimental material, cDNA libraries from leaves and rhizomes treated for 0, 6, 12, 24, and 48 h with 20% polyethylene glycol (PEG)‑6000 to simulate a drought environment were sequenced using the Illumina sequencing platform. We obtained 1, 976, 033 transcripts and 743, 982 unigenes (mean length of 716 bp) through a hierarchical clustering analysis of the resulting transcriptome data. The unigenes were compared against the Nr, Nt, Pfam, KOG/COG, Swiss‑Prot, KEGG, and gene ontology (GO) databases for functional annotation, and the gene expression levels in leaves and rhizomes were compared between the 20% PEG‑6000 stress treated (6, 12, 24, and 48 h) and control (0 h) groups using DESeq2. -

The chromosome numbers were determined as 2n=2x=16 in C. behen L. and C. solstitialis L.; 2n=2x=18 in C. cynarocephala Wagenitz and C. agregata Fisch. & Mey. ex DC. ssp. agregata; 2n=2x=20 in C. iberica Trev. ex Sprengel; 2n=2x=26 in C. balsamita Lam.; 2n=4x=36 in C. virgata Lam. All of the chromosomes had centromeres at median point (M), median region (m), and submedian region (sm). Sat- ellite chromosomes (sat-chromosomes) were observed only in C. virgata. The findings for each of the analyzed taxa are compared each other and with the results of previous studies. Key words Asteraceae, Centaurea, Chromosome number, Karyotype, Turkey. The number of chromosomes of 11 taxa belonging to the subgenus Cyanus was determined for the first time. As a result of the karyomorphological studies, the number of basic chromosomes was determined to be x = 8, 10, and 12 in annuals and x = 10 and 11 in perennials. The populations are tetraploid in the seven perennial taxa and polyploidy is not rare for this group. On the other hand, all annual taxa are diploid. Considering the asymmetry indices, we can conclude that most taxa have symmetrical karyotypes. The most common karyotype formulas are 40 metacentric chromosomes (m), 20m, and 16m + 4 submetacentric chromosomes, respectively. A satellite was detected in the majority of the taxa, but it was observed to be mainly localized on the short arm of the chromosome. Satellites are located mainly on the second chromosome. EBERHARD^ AND ELDREDGE BERMINGHAM Smithsonian Tropical Research Institute, Apartado 2072, Balboa, República de Panamá ABSTRACT.•We present a phylogenetic analysis of relationships among members of the Amazona ochrocephala species complex of parrots, a broadly distributed group in Middle and South America that has been a "taxonomic headache." Mitochondrial DNA sequence data are used to infer phylogenetic relationships among most of the named subspecies in the complex. Sequence-based phylogenies show that Middle American subspecies included in the analysis are reciprocally monophyletic, but subspecies described for South America do not reflect pat- terns of genetic variation. Samples from the lower Amazon cluster with samples collected in western Amazonia•not with samples from Colombia and Venezuela, as was predicted by subspecies classification. All subspecies of the complex are more closely related to one another than to other Amazona species, and division of the complex into three species {A. ochrocephala, A. auropalliata, and A. oratrix) is not supported by our data. Divergence-date estimates suggest that these parrots arrived in Middle America after the Panama land-bridge formed, and then expanded and diversified rapidly. As in Middle America, diversification of the group in South America occurred during the Pleistocene, possibly driven by changes in distribution of forest habitat. Pollen grains of the subgenus Cyanus were tricolporate or syncolporate, subprolate or prolate-spheroidal, 26.15 to 53 µm in polar size, 22.84 to 46.86 µm in equatorial size, isopolar, and radially symmetrical. The only type of pollen grain ornamentation observed was scabrate-perforate. Principal component analysis and cluster analysis were performed to investigate the relationships of the studied Cyanus taxa. As a result of the phenetic analyses, the polar axis, equatorial axis, colpus length, and apocolpium were found to be significant palynomorphological characteristics for taxonomic use. No attempt at phylogenetic reconstruction has been made since recent revisions in circumscription, and previous reconstructions did not include a good representation of species. A new molecular survey is thus needed. Methods Phylogenetic analyses were conducted using sequences of the internal transcribed spacers (ITS) 1 and 2 and the 5.8S gene. Parsimony and Bayesian approaches were used. Key Results A close correlation between geography and the phylogenetic tree based on ITS sequences was found in all the analyses, with three main groups being resolved: (1) comprising the most widely distributed circum- Mediterranean/Eurosiberian sections; (2) the western Mediterranean sections; and (3) the eastern Mediterranean and Irano-Turanian sections. The results show that the sectional classification in current use needs major revision, with many old sections being merged into larger ones. -

(Sevgi Çiçeği) tohumlarının çimlenmeleri üzerine; suda bekletme (10 saat ve 24 saat), GA3 çözeltisinde bekletme (10 ppm ve 100 ppm’lik çözeltilerde 24 saat), 3, 4 ve 5 ay süre ile 0 °C’de katlama ortamında bekletme uygulamalarının ve tohum çimlenme ortamındaki farklı pH derecelerinin (6.5 pH, 7.5 pH ve 8.5 pH) etkilerinin araştırılması amaçlanmıştır. Çalışmada, Ankara-Gölbaşı’ndan 2005 ve 2006 yıllarında toplanan C. tchihatcheffii tohumları kullanılmıştır. GA3 çözeltisinde bekletilen ve 4-5 ay katlamada tutulan tohumlarda çimlenme oranları yüksek bulunmuştur. 8.5 pH derecesindeki ortamlarda ve 2005 yılı tohumlarında çimlenme oranları daha düşük düzeylerde belirlenmiştir.
