Dr. Yeşim OKAY Araştırmada, günümüzde sadece Ankara–Gölbaşı’nda sınırlı bir alanda yaşamakta olan endemik bitki Centaurea tchihatcheffii Fish.&Mey. (Sevgi Çiçeği) tohumlarının çimlenmeleri üzerine; suda bekletme (10 saat ve 24 saat), GA3 çözeltisinde bekletme (10 ppm ve 100 ppm’lik çözeltilerde 24 saat), 3, 4 ve 5 ay süre ile 0 °C’de katlama ortamında bekletme uygulamalarının ve tohum çimlenme ortamındaki farklı pH derecelerinin (6.5 pH, 7.5 pH ve 8.5 pH) etkilerinin araştırılması amaçlanmıştır. Çalışmada, Ankara-Gölbaşı’ndan 2005 ve 2006 yıllarında toplanan C. tchihatcheffii tohumları kullanılmıştır. GA3 çözeltisinde bekletilen ve 4-5 ay katlamada tutulan tohumlarda çimlenme oranları yüksek bulunmuştur. 8.5 pH derecesindeki ortamlarda ve 2005 yılı tohumlarında çimlenme oranları daha düşük düzeylerde belirlenmiştir. Prof. Dr. Yeşim OKAY The research aims to investigate the effect of soaking in water (10 hours and 24 hours), in GA3 solution (10 ppm and 100 ppm concentrations for 24 hours), stratification for 3, 4 and 5 months at 0°C and effect of different pH in seed germination medium (6.5pH, 7.5pH and 8.5pH) on seed germination of endemic plant C. tchihatcheffii Fish.& Mey. (Sevgi Çiçeği) growing nowadays only in a limitted environment around Ankara- Gölbaşı. In the study, C. tchihatcheffii seeds obtained from Ankara-Gölbaşı in 2005 and 2006 were used. Seed germination rate of seeds at GA3 solution and seeds stratified for 4-5 months were high. Seeds maintained in medium at pH 8.5 and seeds collected in 2005 revealed low levels of germination rate. The chromosome numbers were determined as 2n=2x=16 in C. behen L. and C. solstitialis L.; 2n=2x=18 in C. cynarocephala Wagenitz and C. agregata Fisch. & Mey. ex DC. ssp. agregata; 2n=2x=20 in C. iberica Trev. ex Sprengel; 2n=2x=26 in C. balsamita Lam.; 2n=4x=36 in C. virgata Lam. All of the chromosomes had centromeres at median point (M), median region (m), and submedian region (sm). Sat- ellite chromosomes (sat-chromosomes) were observed only in C. virgata. The findings for each of the analyzed taxa are compared each other and with the results of previous studies. Key words Asteraceae, Centaurea, Chromosome number, Karyotype, Turkey. The number of chromosomes of 11 taxa belonging to the subgenus Cyanus was determined for the first time. As a result of the karyomorphological studies, the number of basic chromosomes was determined to be x = 8, 10, and 12 in annuals and x = 10 and 11 in perennials. The populations are tetraploid in the seven perennial taxa and polyploidy is not rare for this group. On the other hand, all annual taxa are diploid. Considering the asymmetry indices, we can conclude that most taxa have symmetrical karyotypes. The most common karyotype formulas are 40 metacentric chromosomes (m), 20m, and 16m + 4 submetacentric chromosomes, respectively. A satellite was detected in the majority of the taxa, but it was observed to be mainly localized on the short arm of the chromosome. Satellites are located mainly on the second chromosome. Pollen grains of the subgenus Cyanus were tricolporate or syncolporate, subprolate or prolate-spheroidal, 26.15 to 53 µm in polar size, 22.84 to 46.86 µm in equatorial size, isopolar, and radially symmetrical. The only type of pollen grain ornamentation observed was scabrate-perforate. Principal component analysis and cluster analysis were performed to investigate the relationships of the studied Cyanus taxa. As a result of the phenetic analyses, the polar axis, equatorial axis, colpus length, and apocolpium were found to be significant palynomorphological characteristics for taxonomic use. No attempt at phylogenetic reconstruction has been made since recent revisions in circumscription, and previous reconstructions did not include a good representation of species. A new molecular survey is thus needed. Methods Phylogenetic analyses were conducted using sequences of the internal transcribed spacers (ITS) 1 and 2 and the 5.8S gene. Parsimony and Bayesian approaches were used. Key Results A close correlation between geography and the phylogenetic tree based on ITS sequences was found in all the analyses, with three main groups being resolved: (1) comprising the most widely distributed circum- Mediterranean/Eurosiberian sections; (2) the western Mediterranean sections; and (3) the eastern Mediterranean and Irano-Turanian sections. The results show that the sectional classification in current use needs major revision, with many old sections being merged into larger ones.

E-mail [email protected] 1 • Background and Aims The geographic distribution of the genus Plectocephalus comprises a single species in Ethiopia, two in North America and possibly four more in South America, in a striking disjunction that is exceptional for genera of the tribe Cardueae. The enormity of this disjunction cast doubts on the precise taxonomic delineation of the genus, which is not unanimously recognized as a natural entity. Our aims were to define the generic boundaries of Plectocephalus and to formulate a hypothesis that would explain its natural range. • Methods A combined molecular approach, using nuclear ITS and ETS and plastid trnL-trnL-F, rpl32-trnLUAG and ndhF markers, was chosen for phylogenetic reconstruction by maximum parsimony and Bayesian inference. • Key Results Phylogenetic analysis shows that Plectocephalus is a natural genus that includes the African species P. varians, together with all the native South American species, currently classified as Centaurea, C. cachinalensis, C. floccosa and C. tweediei. The recognition of Centaurodendron as an independent genus, which we consider appropriate, would make Plectocephalus paraphyletic.

Accepted 9 August, 2010 Centaurea tchihatcheffii Fisch. and Mey., is a critically endangered endemic species, which grows on limited area around Golbasi district of Ankara province. It has attractive pink, red, purple flowers, the colours of which change with maturation. It has a high potential as out door ornamental plant and as cut flower. Its natural habitat is continuously on decrease because of uncontrolled plucking, intense construction activities and urbanization. The most important reason why the generation of the species is endangered is strong anthropogenic pressure. Therefore, the protection of this plant is of great importance. Study of propagation possibilities for the protection of this species, is of primary importance. This paper reviews C. tchihatcheffii, propagation studies. (Asteraceae). Essential oils obtained by hydrodistillation from different colored flowers of C. tchihatcheffii were separately analyzed by gas chromatography (GC) and gas chromatography/mass spectrometry (GC-MS), simultaneously. The main constituent both oils was found as germacrene D (17.6% and 17.4%). The flower color difference was shown not to play a role in essential oil yield and composition. New fossil evidence and a nearly resolved phylogeny of Cardueae are used here to reconstruct the spatiotemporal evolution of this group. • Methods: We performed maximum parsimony and Bayesian phylogenetic inference based on nuclear ribosomal DNA and chloroplast DNA markers. Divergence times and ancestral area reconstructions for main lineages were estimated using penal- ized likelihood and dispersal–vicariance analyses, respectively, and integrated over the posterior distribution of the phylogeny from the Bayesian Markov chain Monte Carlo analysis to accommodate uncertainty in phylogenetic relationships.
