The Solanaceae family is one of the largest and well-distributed plant families in the world. It contains species of agricultural and economical importance, such as Solanum tuberosum, Solanum melongena, Solanum lycopersicum, Nicotiana tabacum, and Capsicum annuum. In Puerto Rico, there are ’46 species of Solanaceae of which six are endemic: Brunfelsia densifolia, Brunfelsia lactea, Brunfelsia portoricensis, Goetzea elegans, Solanum ensifolium, and Solanum woodburyi. Our objective was to use DNA barcoding to identify the Solanaceae species in Puerto Rico, including the endemics, and to assess the species relationships between them. To accomplish our objective, two chloroplast regions (psbA-trnH and matK) and a nuclear region [internal transcribed spacer (ITS)] were assessed. Pairwise distance and phylogenetic analysis demonstrate that DNA barcoding can be used to discriminate at the species level among these taxa in Puerto Rico. For all three markers, the genus that showed the highest pairwise distance between represented species was Solanum, whereas the genus that displayed the least was Capsicum. Phylogenetic trees of single and concatenated regions were generated from sequences obtained in this study and from data downloaded from the National Center for Biotechnology Information database.

Jagatheeswari Department of Botany, Annamalai University, Annamalai Nagar, Chidambaram, Tamil Nadu, India E-mail address: [email protected] ABSTRACT Solanum genus namely Solanum seaforthianum Andr. belongs to the Solanaceae family, and comprises only dioeciously species. These plants are distributed between 29º and 40º south. All species of this genus are diploid with chromosome numbers of 2n = 24, 28 and 30. According to literature, the basic chromosome number in this genus is x = 12, 14 and 15. Solanum genus with a chromosome complement of 2n = 30 has a symmetric karyotype with a median and sub median centromere position. Because ancestral species have a symmetric karyotype, it seems that x = 12 is the initial basic chromosome number in this genus and the x = 14 and x = 15 derived from x = 12. So it seems that diploid phenomena played an important role in evolution and speciation. INTRODUCTION The genus Solanum L. belongs to the family solanaceae in the order Solanales. It is one plant genus of great importance for food security in most countries of the developing world. Some of its members are cultivated mostly as vegetable crops (Kochlar, 1981), while the active principles such as solinidine and other steroids extracted from the roots and leaves of some species are useful pharmaceuticals (Simmonds and Choudhury, 1976). This plant was named after Lord Seaforth (Francis Mackenzie Humberston) who was Governor of Barbados in the West Indies from 1801 to 1806. It was he, who was responsible for the introduction of this and many other plant species into Britain as living plants for gardens from the West Indies. This is another plant species that was introduced for ornamental purposes and has now escaped into the urban areas where it will be found scrambling in hedges and shrubs and the natural areas. Solanum seaforthianum is a scrambling invasive vine which will grow in sun to partial shade and is generally found climbing amongst shrubs and trees in woodland areas and forest margins where it can attain a height of up to 7 metres or more. It is often found on anthills and along stream and river banks where there is sufficient light. All parts of the plant and the seed are poisonous to humans if ingested. The seed is mainly dispersed by birds to which it is not toxic, but it has been reported to be toxic to livestock. Abstract A data set derived from gene sequences of ndhF is used to deduce phylogenetic relationships among the subgenera of Solanum, among related genera of the tribe Solaneae, and within selected Solanum clades. Complete ndhF sequences were obtained for 12 species of Solanum, representing five of the seven subgenera. ndhF sequences also were obtained from species of Capsicum, Cyphomandra, Datura, Jaltomata, Lycopersicon, Nicotiana, and Physalis. Results of a parsimony analysis of these data indicate that the Solanum species form a monophyletic clade with Capsicum rather weakly supported as the sister group. LycojJersicon and Cyphomandra are included within the Solanum clade. Three to four clades can be identified within Solanum. The subgenera Minon and Potaloe are probably not monophyletic as currently defined. Solanum wendlandii, a species with recurved prickles but no stellate hairs, does not belong with the other spiny solanums in subgenus Leptosternonurn. These results cast doubt on the utility of traditional char~lcters such as anther shape and hair morphology in defining high-level infrageneric groups in Solanum. Introduction Solanum, with approximately 1000 to 1400 species (D'Arcy, 1991), is one of the largest genera of flowering plants.
