A todos vocês, os meus sinceros agradecimentos. 7 RESUMO Visando avaliar aspectos biológicos e taxonômicos de trematódeos transmitidos por Melanoides tuberculata (Müller, 1774) no Brasil, coletas malacológicas foram realizadas na represa da Pampulha, Belo Horizonte, Minas Gerais, no período entre março de 2006 a julho de 2009. De 3834 exemplares de M. tuberculata coletados e examinados, 279 (7,3%) apresentaram-se infectados por larvas de trematódeos que, após caracterização morfológica preliminar, foram utilizadas para estudos biológicos experimentais. Estágios evolutivos (cercárias, metacercárias, rédias, parasitos adultos, ovos e/ou miracídios) foram estudados a fresco e após processo de fixação, coloração e montagem de lâminas permanentes, seguida de análise morfológica e morfométrica em microscópio de luz. Cercárias do tipo Pleurolofocerca foram encontradas em 268 (7,0%) exemplares de M. tuberculata e utilizadas para a infecção de Poecilia reticulata Peters, 1859. Metacercárias obtidas nas brânquias dos peixes 30 dias após a infecção foram inoculadas por via oral a Mus musculus (Linnaeus, 1758) da linhagem AKR/J e parasitos adultos recuperados no intestino delgado dos camundongos 30 dias após infecção foram identificados como Centrocestus formosanus (Nishigori, 1924). Outro tipo cercariano, do grupo Megalura, foi encontrado em 11 (0,3%) exemplares de M. tuberculata coletados. Estas larvas formaram metacercárias em substrato sólido que foram utilizadas para a infecção de Gallus gallus domesticus (Linnaeus, 1758) jovens. Parasitos adultos obtidos da membrana nictitante e saco conjuntival das aves, 30 dias após infecção, foram identificados como Philophthalmus gralli Mathis & Leger, 1910. Este é o primeiro registro de ambas as espécies de trematódeos em M. tuberculata no Brasil. Palavras-chave: Melanoides tuberculata, trematódeos, Centrocestus formosanus, Philophthalmus gralli, infecção experimental, represa da Pampulha, Brasil 8 ABSTRACT With the aim to evaluate some biological and taxonomic aspects of trematodes transmitted by Melanoides tuberculata (Müller, 1774) in Brazil, malacological surveys were carried out at Pampulha dam, Belo Horizonte, Minas Gerais State from March 2006 to July 2009. From 3,834 specimens of M. tuberculata collected and examined, 279 (7.3%) were found infected by larvae of trematodes. After preliminary morphological characterization, larvae were used for experimental life cycle studies. Developmental stages (cercariae, metacercariae, rediae, adult parasites, eggs and/or miracidium) were studied alive and after fixation, staining and permanent mounting. The morphological and morphometric analyses were made using a light microscopy. Pleurolophocerca cercaria was found in 268 (7.0%) specimens of M. tuberculata and was used for experimental infection of laboratory reared Poecilia reticulata Peters, 1859. Metacercariae obtained from gills of fishes 30 days after the infection were inoculated by oral route in AKR/J strain of Mus musculus (Linnaeus, 1758). Adult parasites recovered in small intestine of mice, 30 days after the infection, were identified as Centrocestus formosanus (Nishigori, 1924). Another cercarian type, Megalura group, was found in 11 (0.3%) M. tuberculata specimens collected. These larvae encysted in solid substrate and were used for experimental infection of young chicken Gallus gallus domesticus (Linnaeus, 1758). Adult parasites recovered from the nictitant membrane and conjunctival sac of the chickens, 30 days after the infection, were identified as Philophthalmus gralli Mathis & Leger 1910. This is the first report of both fluke species in M. tuberculata from Brazil. Infected snails released megalurous cercariae which readily encysted in characteristically ¯ask-shaped cysts. Adult ¯ukes were recovered from the ocular orbit of experimentally infected domestic ducklings inoculated with excysted metacercariae. The adult and larval stages of the ¯uke are described and compared with those of the other marine-acquired Philophthalmus species. The metacercaria of the Kuwaiti isolate of P. hegeneri diers from those of an American isolate from Batillaria minima in the Gulf of Mexico in the process of encystment, and metacercarial cyst shape resembles those of P. larsoni from a congeneric snail, C. muscarum, from Florida, suggesting a close relationship. The present record signi®cantly extends the known geographical range of P. hegeneri and implicates a new intermediate host. MARCH 1984 Snails of the species Thais (Nucella) lapillus (L) were collected from Scarborough South Bay, and Robin Hoods Bay, North Yorkshire. The presence of the rediae of Parorchia acanthus. NICOLL (Digenea: PHILOPHTHALMIDAE) in T-,. lapillus individuals was previously associated with abnormal shell growth by Feare (1970a). His work has been extended to provide more conclusive evidence of parasitic gigantism in T, larAllus infested with P-,. acanthus-. - The energy increment and soft tissue mass increase associated with shell growth has been calculated for a sample of infested T, lapillus individuals. As reported by Cooley (1958) and Feare (1969) infestation with P_.. acanthus rediae progressively destroys the host gonad. The resultant reproductive saving was estimated for non-infested male and female T, lapillus from Robin Hoods Bay in 1981 and the energy values obtained were compared with estimates of the average energy loss from infested M., laDillus as a result of cercarial production and redial growth. The proportion of the whole body dry mass of infested M, lapillus. individuals contributed by the redial population was generally similar to the gonadal proportion of non-infested femalest but did not follow the same seasonal cycle. The digestive gland of infested dogwhelks was proportionally reduced from that of non-infested females in August only. The growth of redial populations within the hosts through the summer is suggested as a possible cause of host gigantism.

Sao Paulo 52(6):323-327, November-December, 2010 doi: 10.1590/S0036-46652010000600007 Melanoides tuberculata AS INTERMEDIATE HOST OF Philophthalmus gralli IN BRAZIL Hudson Alves PINTO & Alan Lane de MELO SUMMARY Melanoides tuberculata that naturally harbored trematode larvae were collected at the Pampulha dam, Belo Horizonte (Minas Gerais, Brazil), during malacological surveys conducted from 2006 to 2010. From 7,164 specimens of M. tuberculata collected, 25 (0.35%) were infected by cercariae, which have been morphologically characterized as belonging to the Megalurous group, genus Philophthalmus. Excysted metacercariae were used for successful experimental infection of Gallus gallus domesticus, and adult parasites recovered from the nictitating membranes of chickens were identified asPhilophthalmus gralli. Captive animals commonly have infections by direct life cycle parasites, since they are easily transmitted between individuals. However, diagnosing these infections in the laboratory is challenging due to the wide variety of parasite, their life stages and to the variety of available diagnose techniques, being difficult to choose the best one. The present study sampled a group of captive Indian peafowl ( Pavo cristatus ) from S ão Paulo Zoological Park Foundation, São Paulo, Brazil, to test and compare different coproscopical techniques commonly applied in veterinarian clinical analysis laboratories: direct smear, concentrations by sodium chlorite, sucrose, zinc sulphate, faecal sedimentation and formalin-ether followed by modified Ziehl-Neelsen staining. Sensitivity, specificity, predictive values (positive and negative) and Cohen’s kappa index were calculated. In total 108 samples were processed and parasites found were: non- sporulated coccidian oocysts (91.7%), Capillarinae eggs (89.8%), unidentified nematode larvae (75%), Ascarididae eggs (63%), unidentified nematode adults (60.2%), unidentified nematode eggs (42.6%), strongylid-like eggs (42.6%), Cryptosporidium spp.

Importantly, zoonotic helminths that affect human eyes (HIE) may cause blindness with severe socio-economic consequences to human communities. These infections include nematodes, cestodes and trematodes, which may be transmitted by vectors (dirofilariasis, onchocerciasis, thelaziasis), food consumption (sparganosis, trichinellosis) and those acquired indirectly from the environment (ascariasis, echinococcosis, fascioliasis). Adult and/or larval stages of HIE may localize into human ocular tissues externally (i.e., lachrymal glands, eyelids, conjunctival sacs) or into the ocular globe (i.e., intravitreous retina, anterior and or posterior chamber) causing symptoms due to the parasitic localization in the eyes or to the immune reaction they elicit in the host. Unfortunately, data on HIE are scant and mostly limited to case reports from different countries. The biology and epidemiology of the most frequently reported HIE are discussed as well as clinical description of the diseases, diagnostic considerations and video clips on their presentation and surgical treatment. Molecular evolution Herein, we provide the first data on the putative SSU rRNA secondary structures of the Plagiorchiida species.The Taxonomy structures were found to be quite conserved across broad range of species studied, well compatible with those of others eukaryotic SSU rRNA and possessed some peculiarities: cross-shaped structure of the ES6b, additional shortened ES6c2 helix, and elongated ES6a helix and h39 + ES9 region. The secondary structures of variable regions ES3 and ES7 appeared to be tissue-specific while ES6 and ES9 were specific at a family level allowing considering them as promising markers for digenean systematics. Their uniqueness more depends on the length than on the nucleotide diversity of primary sequences which evolutionary rates well differ. The findings have important implications for understanding rRNA evolution, developing molecular taxonomy and systematics of Plagiorchiida as well as for constructing new anthelmintic drugs.
