Interestingly, such exquisite plants originate from minute seeds, going through challenges to germinate and establish in nature. Alternatively, orchid utilization as an economically important plant gradually decreases its natural population, therefore, driving the need for conservation. As with any conservation attempts, broad knowledge is required, including the species’ interaction with other organisms. All orchids establish mycorrhizal symbiosis with certain lineages of fungi to germinate naturally. Since the whole in situ study is considerably complex, in vitro symbiotic germination study is a promising alternative. It serves as a tool for extensive studies at morphophysiological and molecular levels. In addition, it provides insights before reintroduction into its natural habitat. Here we reviewed how mycorrhiza contributes to orchid lifecycles, methods to conduct in vitro study, and how it can be utilized for conservation needs.     Orchid Research Newsletter No. 66 When the Molecular Systematics Laboratory at the Royal Botanic Gardens, Kew, first opened under the leadership of Mark Chase and Mike Bennett as Keeper of the Jodrell, the main target marker was rbcL for family-wide systematic studies of Orchidaceae and other families. Internal transcribed spacers of nuclear ribosomal DNA were used for studies below the family level. Those of us unfortunate to have lived through those days of manual sequencing in the early 1990s know how laborious it was to label nucleotides with radioactive phosphorus 32 or sulfur 35 with all the safety measures that that entailed, pour polyacrylamide gels between two glass plates without even the smallest air bubble, expose the sequencing gel to x-ray film, and then manually call the bases one by one off the autoradiogram, hoping to get 100 bases of the more than 1300 base pairs in the rbcL sequence and then enter them into the computer database. In many cases, the results were ambiguous, which entailed judgment calls. And this did not even include data analysis with software much, much slower than today. All this work produced only a trickle of data and a lack of strong support for many branches leading to the major clades. Manna from heaven came in the form of automated sequencing later that decade. Automated sequencers used the Sanger method, which relied on the introduction of dideoxynucleotides into the growing DNA strand by DNA polymerase, creating fragments separated by size on the gel by electrophoresis. Nucleotides were labeled by fluorescent dyes rather than sulfur 35, and read by a laser.

    Orchid Research Newsletter No. 65 Those who attended the 21st World Orchid Conference in Johannesburg last September were treated to 1) a marvelous show filled with native South African species of Orchidaceae and other indigenous plant families, and 2) some excellent talks on orchid floras, conservation biology, ecology, and systematics. Just to mention a few, Steve Johnson gave a typically superb plenary talk on deceit pollination in South Africa, and Mike Fay intrigued the audience with news of exciting new approaches to conservation genetics. Nicola Flanagan delivered a fascinating lecture on the diversity of Vanilla species and their mycorrhiza in Colombia. Alex Hirtz introduced the incredible orchid diversity of Ecuador as an enticement to attend the 22nd WOC in Guayaquil in 2017. We thank the organizing committee – Gerrit van Ede, Anthony Grohovaz, Catharina Wilfinger, Frik Botha, and Lourens Grobler – and all the others who helped to make all the registrants feel so welcome. The New Year promises at least two other conferences of note. First up is the 17th European Orchid Conference to be held at the Royal Botanic Gardens, Kew, and the Royal Horticultural Society Halls in Vincent Square, London, in April (see details below). Among the distinguished international speakers will be Cássio van den Berg, Ken Cameron, Andre Schuiteman, Mariana Mondragón-Palomino, Kingsley Dixon, Hanne Rasmussen, Phillip Cribb, Tom Mirenda, Henrik Pedersen, Holger Perner, John Elliott, Svante Malmgren, Hildegard Crous, Pepe Portilla, and Andrea Niessen. Then in November the 5th Scientific Conference on Andean Orchids will convene in Colombia, a nation that rivals (Colombians say surpasses) Ecuador in orchid diversity.

    RODOLFO SOLANO-GÓMEZ1*, ANNE DAMON2, GABRIELA CRUZ-LUSTRE3, LAURA JIMÉNEZ-BAUTISTA3, SOCORRO AVENDAÑO-VÁZQUEZ3, VINCENZO BERTOLINI2, RAÚL RIVERA-GARCÍA1 Y GABRIELA CRUZ-GARCÍA1 Botanical Sciences 94 (3): 625-656, 2016 Abstract The Tacaná-Boquerón region (TBR) in Chiapas is considered an area of high biodiversity in Mexico, with DOI: 10.17129/botsci.589 a rich but poorly studied orchid flora, which is an important component of the vegetation and threatened by an accelerated rate of deforestation. By means of fieldwork, and the revision of scientific collections and literature, an orchid checklist for the TBR and adjacent areas was made; using geographic information systems the distribution of this orchid flora was analyzed in the study area. From 1,235 records we report 105 genera and 325 species, two of them determined to infraspecific level and two are natural hybrids; nine species were additions to the Mexican flora; 39 species are considered at risk in Mexico; the national distribution of a further 20 species are restricted to the TBR. This orchid flora represents 24 % and 44 % of the national and state orchid species richness, respectively, is the second richest in Mexico, and only surpassed by the region El Momón-Las Margaritas-Montebello (Chiapas). The greatest orchid richness is concentrated between elevations of 500 to 2,499 m, in areas now given over to permanent agriculture (including coffee and cocoa plantations); in primary forest the richness is lower. The Tacaná Volcano Bio- sphere Reserve is the only protected area in the TBR and the proposal to extend it to the Boquerón peak would promote the protection of the orchid flora growing above 1,000 m in the region.

The Annex to this document has been prepared by UNEP-WCMC. 2. In accordance with paragraphs b) and c) of the section Selection of species to be reviewed in Resolution Conf. 12.8 on Review of Significant Trade in specimens of Appendix-II species, the Plants Committee should select for its review species of priority concern. The selection should be based on recorded trade levels and information available to the Plants Committee, the Secretariat, Parties or other relevant experts. 3. The Secretariat has required UNEP-WCMC to provide a summary of trade data in Appendix-II plant species over the five most recent years, and guidance on the selection of species for review. 4. The Plants Committee is requested to consider the document Review of Significant Trade – Analysis of Trade Trends with notes on the conservation status of selected species – Volume 1. Plants, by UNEP-WCMC (see the Annex to this document, English only) and to recommend a shortlist of species for its Review of Significant Trade.
