Load 10 L of the sample per lane in a 4-15% Mini-PROTEAN® TGXTM Precast Gel (Bio-Rad; code no. 456-1085) and carry out electrophoresis. 4) Blot the protein to a nitrocellulose membrane at 60 V for 4 hr. at 4°C in a wet transfer system (Transfer Buffer: 25 mM Tris, 190 mM glycine). See the manufacturer's manual for precise transfer procedure. 5) To reduce nonspecific binding, soak the membrane in 5% skimmed milk (in PBS, pH 7.2) for 20 min. at room temperature. 6) Incubate the membrane with primary antibody diluted with Can Get Signal-1 (TOYOBO; code no. NKB-201) as suggested in the APPLICATION for 1 hr. at room temperature. (The concentration of antibody will depend on the conditions.) 7) Wash the membrane with PBS-T [0.05% Tween-20 in PBS] (5 min. x 3 times). 8) Incubate the membrane with the 1:3000-5000 of goat anti-rabbit IgG-HRP (Santa Cruz; code no. sc-2004) diluted with Can Get Signal-2 (TOYOBO; code no. NKB-201) for 20 min. at room temperature. 9) Wash the membrane with PBS-T (10 min. x 3 times) 10) Wipe excess buffer on the membrane, and then incubate it with appropriate chemiluminescence reagent for 1 min. 11) Expose for 3 min. with ImageQuant LAS 4000 imaging system (Fujifilm). The condition for exposure may vary.
