A. 3 Author for correspondence ( [email protected] ) Communicating Editor: Javier Francisco-Ortega Abstract— Previous attempts to resolve relationships among the primary lineages of Campanulidae (e.g. Apiales, Asterales, Dipsacales) have mostly been unconvincing, and the placement of a number of smaller groups (e.g. Bruniaceae, Columelliaceae, Escalloniaceae) remains uncertain. Here we build on a recent analysis of an incomplete data set that was assembled from the literature for a set of 50 campanulid taxa. To this data set we first added newly generated DNA sequence data for the same set of genes and taxa. Second, we sequenced three additional cpDNA coding regions (ca. 8,000 bp) for the same set of 50 campanulid taxa. Finally, we assembled the most comprehensive sample of cam- panulid diversity to date, including ca. 17,000 bp of cpDNA for 122 campanulid taxa and five outgroups. Simply filling in missing data in the 50-taxon data set (rendering it 94% complete) resulted in a topology that was similar to earlier studies, but with little additional resolution or confidence. -

ABSTRACT Wood anatomical features examined in five species of Myodocarpus, four spe cies of Delarbrea, and the single member of Pseudosciadium (Araliaceae) using light and scanning electron microscopy support the hypotheses that they are a closely related, monophyletic group, and that they form an ancient lineage that has survived and diversified on the island of New Caledonia, where all except two of the 17 species belonging to these genera are endemic. Delarbrea and Pseudosciadium have similar wood sttucture; the wood of Myodocarpus is distinctly more primitive (sensu BAILEY). These genera differ markedly in wood structure from other Araliaceae in the presence of apotta- cheal (diffuse and diffuse-in-aggregates) axial parenchyma, and wood features KEYWORDS Myodocarpus, do not suppott earlier suggestions that Myodocarpus is intermediate between Delarbrea, Araliaceae and Apiaceae. Within Myodocarpus two groups of species can be Pseudosciadiu m, distinguished on wood characters, which correspond to the species with Araliaceae, wood anatomy, simple vs. pinnately compound leaves. The wide-ranging D. paradoxa subsp. vasicentric tracheids, paradoxa is the only taxon studied with numerous vasicentric tracheids, often phylogeny, New Caledonia. -

Plant Biodiversity Science, Discovery, and Conservation: Case Studies from Australasia and the Pacific Craig Costion School of Earth and Environmental Sciences Department of Ecology and Evolutionary Biology University of Adelaide Adelaide, SA 5005 Thesis by publication submitted for the degree of Doctor of Philosophy in Ecology and Evolutionary Biology July 2011 ABSTRACT This thesis advances plant biodiversity knowledge in three separate bioregions, Micronesia, the Queensland Wet Tropics, and South Australia. A systematic treatment of the endemic flora of Micronesia is presented for the first time thus advancing alpha taxonomy for the Micronesia-Polynesia biodiversity hotspot region. The recognized species boundaries are used in combination with all known botanical collections as a basis for assessing the degree of threat for the endemic plants of the Palau archipelago located at the western most edge of Micronesia’s Caroline Islands. A preliminary assessment is conducted utilizing the IUCN red list Criteria followed by a new proposed alternative methodology that enables a degree of threat to be established utilizing existing data. Historical records and archaeological evidence are reviewed to establish the minimum extent of deforestation on the islands of Palau since the arrival of humans. This enabled a quantification of population declines of the majority of plants endemic to the archipelago. In the state of South Australia, the importance of establishing concepts of endemism is emphasized even further. A thorough scientific assessment is presented on the state’s proposed biological corridor reserve network. The report highlights the exclusion from the reserve system of one of the state’s most important hotspots of plant endemism that is highly threatened from habitat fragmentation and promotes the use of biodiversity indices to guide conservation priorities in setting up reserve networks. -

It is largely congruent with the phylogeny of angiosperms reconstructed from chloroplast genes atpB, matK, and rbcL, and nuclear 18S rDNA. The basalmost lineage consists of Amborella and Nymphaeales (including Hydatellaceae). Austrobaileyales follow this clade and are sister to the mesangiosperms, which include Chloranthaceae, Ceratophyllum, magnoliids, monocots, and eudicots. With the exception of Chloranthaceae being sister to Ceratophyllum, relationships among these five lineages are not well supported. In eudicots, Ranunculales, Sabiales, Proteales, Trochodendrales, Buxales, Gunnerales, Saxifragales, Vitales, Berberidopsidales, and Dilleniales form a basal grade of lines that diverged before the diversification of rosids and asterids. Within rosids, the COM (Celastrales–Oxalidales–Malpighiales) clade is sister to malvids (or rosid II), instead of to the nitrogen-fixing clade as found in all previous large-scale molecular analyses of angiosperms. Santalales and Caryophyllales are members of an expanded asterid clade. -

Our research included pollen sampled from 417 herbarium specimens representing 158 species belonging to 125 genera distributed among all major lineages of Apiales. The pollen was mechanically isolated, acetolysed, suspended in pure glycerine and mounted on paraffin-sealed slides for light microscopy investigation. Although most of the analysed traits were highly homoplastic and showed significant overlap even between distantly related lineages, we were able to construct a taxonomic key based on characters that bear the strongest phylogenetic signal: P/E ratio, mesocolpium shape observed in polar view and ectocolpus length relative to polar diameter. -
