ON REPRODUCTIVE SYSTEM OF MALE MICE) อาจารย์ที่ปรึกษา : ผชู้ ่วยศาสตราจารย์ ดร.เกรียงศกั ด์ิ เอ้ือมเกบ็ , 105 หน้า. กวาวเครือแดงไดร้ ับการอา้ งวา่ สามารถใช้ในผู้ชายไทยเพื่อการกลบั สู่ภาวะหนุ่มในการ เพ่ิม สมรรถภาพทางเพศ ป้ องกนั มะเร็งตอ่ มลูกหมากและตอ่ มลูกหมากโต เพราะมีสารแอนโดรเจนจาก พืช ในความเป็นจริงการหยอ่ นสมรรถภาพทางเพศคือการที่ไมส่ ามารถรักษาการแข็งตัวของอวัยวะ เพศชายในการมีกิจกรรมทางเพศ ที่มีสาเหตุมาจากสภาพร่างกายและจิตใจ หลายประการ ยาในกลุ่ม ยบั ย้งั พีดีอี -5 ซิลเดนาฟิล ได้รับการรับรองสาหรับการรักษา อาการหยอ่ นสมรรถภาพทางเพศ นอกจากน้นั ยงั พบวา่ ยาน้ีสามารถเพ่ิมระดบั ฮอร์โมนเทสโทสเตอโรน และกระตุน้ เลดิกเซลลส์ ่งผล ใหเ้ กิดการกระตุน้ กระบวนการสร้างตวั อสุจิ อยา่ งไรกต็ าม รายงานผลข้างเคียง ที่พบบอ่ ย ไดแ้ ก่ อาการปวดศีรษะ หนา้ แดง อาหารไมย่ อ่ ย คดั จมูก และรบกวนการมองเห็นภาพสี ดงั น้นั จุดประสงค์ ของการศึกษาคร้ังน้ีเพื่อศึกษาผลของสารสกดั จากรากของกวาวเครือแดงเปรียบเทียบกบั ซิลเดนาฟิล ตอ่ ระบบสืบพันธุ์ของหนู ไมซ์เพศผู้ จากน้นั ผงแหง้ ของรากพืชกวาวเครือแดง จึงถูกสกดั ดว้ ยเอทา นอล และถูก แยกสารสกดั ออกจากกนั โดยซิลิกาเจลคอลมั น์ดว้ ยเทคนิค โครมาโทกราฟี และ สารประกอบที่ถูกแยก ถูกพิสูจน์เอกลักษณ์เพื่อให้ทราบชื่อโดยใช้เครื่องมือโครมาโทรกราฟีแบบ เฟสเคลื่อนที่เป็นของเหลวสมรรถนะสูง (เฮทพีแอลซี ) โดยการเปรียบเทียบกบั สารมาตรฐาน ตอ่ จากน้นั ไดน้ า สารที่สกดั ไดเ้ หล่าน้ีมา ป้อนหนูเพศผู้โดยให้ทางปากในขนาดที่ให้สารสกดั หยาบ ซิลเดนาฟิล แฟลกชันบี แฟลกชันซี และแฟลกชันอี ไดแ้ ก่ 1,250 10 40 50 และ 150 มิลลิกรัมตอ่ กิโลกรัมตอ่ น้า หนกั ตวั ตอ่ วนั ตามลาดับ ส่วนประกอบของสารสกดั เหล่าน้ีพบวา่ เป็นเจนิสติน และไบโอชานินเอ ทุกกลุ่มไดร้ ับการทดลองเป็นเวลา 14 วนั ติดตอ่ กนั เลือดและสเปิร์มได้ถูกเกบ็ เพื่อการวเิ คราะห์ก่อนและหลงั การ ทดลองในทุกกลุ่ม นอกจากน้ี เมื่อสิ้นสุดการทดลองได้ทาการ เกบ็ ส่วนตา่ งๆ ของอวัยวะสืบพันธุ์และอวัยวะที่สาคัญ เพื่อการเปรียบเทียบ การเปลี่ยนแปลงน้า หนกั ตัวสัมพัทธ์ได้ถูกนามาวิเคราะห์ด้วย ผลการศึกษาพบวา่ น้า หนกั ตวั สัมพทั ธ์ของหนูในกลุ่มที่ไดร้ ับ สารท้งั หมด ไมแ่ ตกตา่ งจากกลุ่มควบคุมอยา่ งมีนยั สา คญั ในขณะที่น้า หนกั มา้ มของหนูในกลุ่มที่ ได้รับ แฟลกชันบี ซี และอี จะมีน้า หนกั มากกวา่ กลุ่มควบคุม ในทางตรงขา้ มกบั น้า หนกั ขอ ง (p<0.01) อยา่ งไรกต็ าม น้า หนกั อณั ฑะในกลุ่มที่ไดร้ ับสารสกดั หยาบและกลุ่มไดร้ ับ ซิลเดนาฟิลน้นั ตลอดจนน้า หนกั ของเอพิดิไดมิส ของกลุ่มที่ไดร้ ับ แฟลกชันอีและซี มีน้า หนกั มากกวา่ กลุ่มควบคุม อยา่ งมีนยั สา คญั ทางสถิติ (p<0.01) ยง่ิ ไปกวา่ น้นั หนูที่ภายหลงั การทดสอบสารทุกกลุ่ม มีระดบั ของ เทสโทสเตอโรนสูงข้ึนอยา่ งมีนยั สา คญั เมื่อเทียบกบั ก่อนการใหส้ าร ยกเวน้ กลุ่มควบคุม II นอกจากน้นั ระดบั ของเทสโทสเตอโรนในกลุ่มที่ไดร้ ับแฟลกชนั ซีและอี ยงั สูงกวา่ กลุ่มอื่นๆอยา่ งมี นยั สา คญั และยงั พบวา่ กลุ่มที่ไดร้ ับแฟลกชนั ซีมีจา นวนตวั อสุจิสูงที่สุด การคน้ พบน้ีเป็นหลกั ฐานที่ แสดงวา่ เจนิสติน สารที่ยงั ไมท่ ราบชื่อ และไบโอคานินเอ อาจมีบทบาทสา คญั ในการเพ่ิมระดบั เทส โทสเทอโรน จา นวนตวั อสุจิ และเพ่ิมการเคลื่อนไหวของตวั อสุจิ หรือการรักษาภาวะการมีบุตรยาก ในผู้ชาย หลังจากที่ ได้วิจัยระดับของความปลอดภัยแล้ว ผลการวเิ คราะห์เลือดพบวา่ กลุ่มที่ไดร้ ับ แฟลกชนั ซี อีและซิลเดนาฟิล มีระดบั คอเลสเตอรอลสูงกวา่ กลุ่มควบคุมอยา่ งมีนยั สา คญั ยง่ิ ไปกวา่ น้นั ระดบั เฮโมโกลบินในกลุ่มที่ไดร้ ับแฟลกชนั อี สูงกวา่ กลุ่มที่ไดร้ ับสารสกดั หยาบและกลุ่ม ควบคุมอยา่ งมีนยั สา คญั (p<0.05) สาขาวิชา ชีววิทยา ลายมือชื่อนักศึกษา __________________________ ปีการศึกษา 2556 ลายมือชื่ออาจารย์ที่ปรึกษา ____________________ III WANATKAMON NAKNARONG : THE EFFECTS OF ISOLATED FLAVONOIDS FROM THE TUBEROUS ROOTS OF RED KWAO KREU (Butea superba Roxb.) BUTEA SUPERBA ROXB, TESTOSTERONE, SPERM COUNT, SILDENAFIL, CHOLESTEROL Butea superba Roxb.(BS) has been claimed to use in Thai men for rejuvenation, improve sexual function, prevent prostate cancer and prostatic hyperplasia, because it contains Phytoandrogen. In fact, Erectile dysfunction (ED) is the inability to maintain penile erection for the successful performance of sexual activity which is caused by many physical and psychological factors. The PDE-5 inhibitors, sildenafil, is approved for the treatment of ED. In addition, sildenafil has increased testosterone level and stimulated Leydig cells resulting in an increase of spermatogenesis. However, the most frequent adverse effects reported for sildenafil is headache, flushing, dyspepsia, nasal congestion, and disturbances in color vision. Hence, the objective of this study was to investigate the effects of the tuberous root of Butea superba Roxb extract compared with sildenafil on the reproductive system of male mice. Then, the dried powder of the tuber roots of Butea superba Roxb. was extracted with ethanol and separated by silica gel column chromatography The chemical names of the isolated compounds were identified by High performance Liquid Chromatography (HPLC) compared with standard compounds. Then, these compounds were orally administered to male mice. The crude extract, sildanafil, fraction B, C and D were fed at the doses of 1,250, 10, 40, 50 and 150 mg /kg BW/day respectively. The majority of these extract compounds such as Genistein and IV Biochanin A were elucidated. All groups were treated for 14 consecutive days. Blood and sperms were collected for analysis before (pre-) and after (post-) treatment in all groups. Also, at the end of the experiments, the selected reproductive and vital organs were collected for comparative measurements. A relative change of body weight was also analyzed. The results showed that the relative body weights in all treated groups were not significant different from the control. Whereas, the spleen weight of fraction B, C and E treated groups were significantly heavier, and stomach weight of fraction E group was lighter than those of the control (p<0.01). However, the testes weight of crude extract and sildenafil treated groups, and the epididymis weight of fraction E and C treated group were significantly heavier than those of the control (p<0.01). In addition, the results exhibited that there were significant increases in testosterone levels of all post-treated groups compared to pre-treatment groups, except for that of the control (p<0.01). In addition, the testosterone levels of fraction C and E treated groups were significantly higher than those of other groups (p<0.01). Also, the sperm number of all treated groups were significantly higher than that of the control group (p<0.01) as well as the fraction C which showed the highest number. These findings provide evidence that Genistein, Unknown compound 1 and Biochanin A may play an important role in increasing of testosterone level, sperm number and motility or infertility treatment in men after safety level is investigated. The blood analysis showed that the cholesterol level of fraction C, E and sildenafil were significantly higher than those of the control (p<0.05). Morover, the hemoglobin level of fraction E treated group was significantly higher than those of the control and the crude extract groups (p<0.05). It is a medium sized deciduous tree which is mainly found in ascending of Himalayas up to 900 m height and in peninsular India up to 1200 m height. All parts of plant are important viz . Bark, leaves, flowers, seed etc. This plant is used ethanobotanically in Ayurveda, Unani, Homeopathic and Allopathic system of medicines as astringent, aphrodisiac, antifertility, analgesic, burning sensation, diuretics, diarrhea, dysentery, depurative, filariasis, gout, helminthiasis, leprosy, night blindness, piles, skin diseases, sore throat, snake bite, tumours, ulcer etc. The plant is reported to possess anthelmintic, anti convulsion, anti diarrheal, anti fungal, anti diabetic, hypoglycemic, anti hyperlipidaemic, anti inflammatory, anti oxidant, wound healing, anti fertility and hepatoprotective Activity. All these uses are due to the present of certain phytoconstituent in the plant. The major phytoconstituent reported in this plant are Isobutrin, butrin, butin, butein, chalcones, aurones, isobutyine, palasitrin, 3',4',7- trihydroxyflavone, palsitrin, Coreopsin, isocoreopsin, sulphurein, monospermoside, isomonospermoside, quercetin, prunetin, Lupenone, lupeol.

From this point of view, the biological spectrum, leaf spectrum and conservation status of the current sacred grove vegetation, SBT (Swarga Bauri Than) in Jhargram district of West Bengal, India, have been studied. The area's floristic study revealed that SBT‟s angiosperms were varied and consisted of 307 species belonging to 249 genera, distributed under 79 families of 36 orders as per APG IV. Fabales (12.05%) and Fabaceae (11.73%) are the dominant order and family in terms of species wealth. Biological spectrum indicates that the region enjoys “thero-chamae-cryptophytic” type of phytoclimate. With respect to the spectrum of the leaf size, mesophyll (14.05%) was found to be high followed by notophyll (7.84%), microphyll (7.19%), macrophyll (7.84%), nanophyll (6.86%), leptophyll (6.21%), and megaphyll (2.29%). The study area, being a sacred grove, it has a comparatively undisturbed status, and the protection of germplasm in the grove is based on traditional belief in the social system.

Email: [email protected] Received: 03 May 2018, Revised and Accepted: 12 June 2018 ABSTRACT Objective: Medicinal plants have different chemicals in their roots, stem, leaves, flowers, and fruits. In human history, medicinal plants play a vital role in the traditional medicine because of herbal plants have a great nutritive value, and some of them are also used in medicine. The efficacy of medicinal plants is mainly allied with their components such as secondary metabolites and inorganic acid, and basic radicals considered that prolonged intake can cause health problems if the heavy metals such as mercury, cadmium, and other impurities are the threshold concentrations. Methods: In the present study, the flower extract of Butea superba was analyzed for the inorganic acid and basic radicals and the estimation of total saponins, terpenoids, and alkaloids. The above protocols were done using standard methods. Results: The flower extract of B. superba revealed that the inorganic acid radicals such as chromate, phosphate, and sulfate and the basic radicals such as lead, arsenic, and ferrous showed that positive results. The percentage of total saponins, terpenoids, and alkaloids were compared in the following order, saponins<terpenoids<alkaloids as 4.181±0.017%<79.833±0.351%<107.823±0.105%, respectively.

Kuntze Ashish Mishra*, Saket Verma, , Abhinav Prasoon Mishra Advance Institute of Biotech and Paramedical Sciences, Kanpur, India. ABSTRACT Butea monosperma (Lam.) Kuntze (Syn. Butea frondosa; Family Fabaceae), This is a moderate sized deciduous tree which is widely distributed throughout India, Burma and Ceylon, popularly known as 'dhak' or 'palas', commonly known as ‘Flame of forest’. The family Fabaceae compromises of 630 genera and 18,000 species. It finds use both medicinally and commercially with each part of the plant having utility. This plant species has been found to display a wide variety of biological activities. The plant is traditionally reported to possess astringent, bitter, alterative, aphrodiasiac, anthelmintic, antibacterial and anti-asthamatic properties. Bark yields red juice known as ‘Butea gum’or‘Bengalkino’. Previous phytochemical examination of this plant indicated the presence of various constituents some of these are Cajanin and isoformononetin; Stigmasterol; Butin; two known flavonoids, isobutrin (3, 4, 2', 4'-tetrahydroxychalcone-3, 4'- diglucoside), and the less active butrin (7, 3', 4'- trihydroxyflavanone-7, 3'-diglucoside); free sugars and free amino acids and (-)-3-hydroxy-9- methoxypterocarpan (medicarpin) which were isolated from stem-bark extract; bark; seeds; flower; the petroleum ether extract of flowers and petroleum and ethyl acetate extract of stem bark. Its reported pharmacological properties include anthelmintic, anticonceptive, anticonvulsive, antidiabetic, antidiarrhoeal, antiestrogenic and antifertility, antiinflammatory, antimicrobial, antifungal, antibacterial, antistress, chemopreventive, haemaggultinating, hepatoprotective, radical scavenging, Thyroid inhibitory, antiperoxidative and hypoglycemic effects and wound healing activities. It is powerful astringent and is given in many forms of chronic diarrhoea.
