Immune Checkpoint Inhibitors

Much of the recent advancement in the field of tumor immunology and immunotherapy has been generated by the recognition that immune checkpoint proteins can be blocked by human . Immune checkpoint pathways involve both costimulatory and inhibitory proteins. The costimulatory receptors transduce signals to promote immunity against pathogens. The inhibitory receptors, on the other hand, negatively regulate T- cell activation to prevent excessive inflammation. Immunomodulatory antibodies directed against cytotoxic T lymphocyte antigen-4 (CTLA-4) and programmed death-1 (PD-1), have demonstrated significant promise in the treatment of melanoma and other tumor types. These agents are commonly called “checkpoint inhibitors” because they block normally negative regulators of immunity. Numerous other immune checkpoint pathways that could be the target of novel therapies have been identified and are currently under development for clinical testing.
