Maul 2 ~The Salk Institute for Biological Studies, La Jolla, California 92037; 2Wistar Institute of Anatomy and Biology, Philadelphia, Pennsylvania 19104; 3Howard Hughes Medical Institute, 4Institute for Human Gene Therapy, University of Pennsylvania, Philadelphia, Pennsylvania 19104 USA Wild-type PML and at least four other novel proteins are localized within discrete nuclear structures known as PODs. We demonstrate here that during adenovirus infection, immediate early viral proteins from the E1 and E4 transcription units associate with the POD, which in turn undergoes a dramatic morphological change. During this process, the auto-antigen Sp-100 and NDP55 but not PML, relocate from the POD to the viral inclusion bodies, the sites of adenovirus DNA replication and late RNA transcription. The E4-ORF3 11-kD protein alone will induce this reorganization and reciprocally, viruses carrying mutations in the E4-domain fail to do so. These same viral mutants are defective in viral replication as well as the accumulation of late viral mRNAs and host cell transcription shutoff. We show that interferon (INF) treatment enhances the expression of PML, reduces or blocks PODs reorganization, and inhibits BrdU incorporation into viral inclusion bodies. In addition, cell lines engineered to overexpress PML prevent PODs from viral-induced reorganization and block or severely delay adenovirns replication.

HAROLD SAMUEL GINSBERG May 27, 1917–February 2, 2003 Elected to the NAS, 1982 Harry Ginsberg was one of the founders of modern virology. He began his career with research in epidemi- ology, describing the pathogenesis of viral infections, the course of infections, and their outcomes. This was followed by the isolation of novel viruses. Harry was associated with the identification of several adenovirus serotypes that caused acute respiratory diseases, atypical pneumonia, and respiratory illnesses common to children. His lifelong study of the adenoviruses made this group of viruses one of the most intensively explored and better understood classes of viruses. Having identified the adenoviruses and their pathogenic consequences, Harry changed his research direction, By Arnold J. Levine helping to create the field of the molecular biology of animal viruses. His research uncovered each of the phases of viral replication, and he was among the first to apply chemotherapy to interrupt virus replication and prevent pathogenesis. In addition, he studied how a virus could inhibit the synthesis of cellular RNA and proteins and how viruses redirected cellular metabolism for the propagation of these agents. H e then changed his research direction for a third time, employing molecular genetics to uncover the functions of adenovirus genes and their regulation. As it became clear that some adenovirus serotypes could cause cancers in animals, Harry’s research group prepared the antiserum from tumor-bearing animals to unravel the viral tumor antigens.

What was the more benign or more confined progen- itor of HIV? What is its relationship to other viruses with similar physical or pathological properties? How rapidly do variations of HIV evolve?
