No Conflicts of Interest related to the issues discussed in this meeting have been identified during the screening process, and no interests were declared orally by the members at the beginning of this meeting. 4. Scientific topic(s) for discussion 4.1. Evaluation of the food enzyme beta-galactosidase from Klyuveromyces lactis data package DSM (EFSA-2021-00182) The WG discussed the draft opinion and concluded that additional information is required from the applicant. Once the additional information is received, the draft opinion will be discussed at a future WG meeting. 4.2. Evaluation of the food enzyme 3-phytase from Aspergillus niger (strain PHY93-08) (EFSA-2016-00575) The WG discussed the additional information from the applicant and needs further discussion. 4.3. Evaluation of the food enzyme pectinesterase from a genetically modified strain of Aspergillus niger (FLZSC) (EFSA-2015-00087) The WG discussed the additional information from the applicant and needs further discussion. 4.4. Evaluation of the food enzyme alpha-amylase from Aspergillus oryzae (AE-AA) (EFSA-2014-00913) The WG discussed the draft opinion and concluded that additional information is required from the applicant. On the basis of sequence similarity all known glycosidases have been classified into 90 families. In many cases proteins of different families have common evolution origin. It makes necessary to combine the corresponding families into a superfamily. Results: Using of the PSI-BLAST program we found significant sequence similarity of several glycosidase families, two of which includes enzymes with the α galactosidase activity. Sequence homology, common catalytic mechanism, folding similarities, and composition of the active center allowed us to group three of these families – GH27, GH31, and GH36 – into the α-galactosidase superfamily. Phylogenetic analysis of this superfamily revealed polyphyletic origin of GH36 family, which could be divided into four families. Glycosidases of the α-galactosidase superfamily have a distant relationship with proteins belonging to families GH13, GH70, and GH77 of glycosidases, as well as with two families of predicted glycosidases. Introduction Glycoside hydrolases or glycosidases (EC 3.2.1.-) are a widespread group of enzymes, hydrolyzing the glycosidic bonds between two carbohydrates or between a carbohydrate and an aglycone moiety. A large multiplicity of these enzymes is a consequence of the extensive variety of their natural substrates: di-, oligo-, and polysaccharides.

However, lately, three new systems have been dened: the threonine-based proteasome system, the glutamate-glutamine system of eqolisin, and the serine-glutamate-aspartate system of sedolisin. However, it remains largely unclear how soil microbial communities respond to long-term, moderate warming. In this study, we used Illumina sequencing and microarrays (GeoChip 5.0) to analyze taxonomic and functional gene compositions of the soil microbial community after 14 years of warming (at 0.8–1.0 °C for 10 years and then 1.5–2.0 °C for 4 years) in a Californian grassland. Long-term warming had no detectable effect on the taxonomic composition of soil bacterial community, nor on any plant or abiotic soil variables. In contrast, functional gene compositions differed between warming and control for bacterial, archaeal, and fungal communities. Functional genes associated with labile carbon (C) degradation increased in relative abundance in the warming treatment, whereas those associated with recalcitrant C degradation decreased. A number of functional genes associated with nitrogen (N) cycling (e.g., denitrifying genes encoding nitrate-, nitrite-, and nitrous oxidereductases) decreased, whereas nifH gene encoding nitrogenase increased in the 1234567890():,; warming treatment. These results suggest that microbial functional potentials are more sensitive to long-term moderate warming than the taxonomic composition of microbial community.

Biocatalytic production of fructose from inulin employing inulinase is the most promising alternative for fructose production. For commercial production, use of immobilized inulinase is advantageous as it offers reutilization of enzyme and increase in stability. In order to meet the demand of concentrated fructose syrup, inulin hydrolysis at high substrate loading is essential. Results: Inulinase was immobilized on chitosan particles and employed for fructose production by inulin hydrolysis. Fourier transform infrared spectroscopy (FTIR) analysis confirmed linkage of inulinase with chitosan particles. Immobilized biocatalyst displayed significant increase in thermostability at 60 and 65 °C. Statistical model was proposed with an objective of optimizing enzymatic inulin hydrolytic process. At high substrate loading (17.5 % inulin), using 9.9 U/g immobilized inulinase at 60 °C in 12 h, maximum sugar yield was 171.1 ± 0.3 mg/ml and productivity was 14.25 g/l/h. Immobilized enzyme was reused for ten cycles. Raw inulin from chicory and asparagus was extracted and supplied in 17.5 % for enzymatic hydrolysis as a replacement of pure inulin. More than 70 % chicory inulin and 85 % asparagus inulin were hydrolyzed under optimized parameters at 60 °C. Results of high performance liquid chromatography confirmed the release of fructose after inulin hydrolysis.
