WOLNE RODNIKI W CHEMII 1. Poj ęcie i rodzaje rodników Rodniki, wolne rodniki (termin angielski – radicals, free radicals ) według definicji Mi ędzynarodowej Unii Chemii Czystej i Stosowanej (IUPAC - International Union of Pure • and Applied Chemistry ) są to indywidua chemiczne (atomy lub cz ąsteczki), na przyklad CH 3, • • • • • SnH 3, NO, NO 2, Cl , H zawieraj ące niesparowane elektrony, tj. układy o spinie elektronowym ró Ŝnym od 0 (IUPAC Gold Book, http://goldbook.iupac.org/R05066.html ). Najprostszym rodnikiem jest atom wodoru. Słowo „rodnik”, pochodz ące od łaci ńskiego „radix”, oznaczaj ącego korze ń, funkcjonuje od około 200 lat [1-3]. Rodniki maj ą du Ŝy wpływ na nasze Ŝycie i zdrowie. S ą obecne w przyrodzie - powstaj ą w atmosferze ziemskiej pod wpływem promieniowania słonecznego, podczas procesów spalania, znajduj ą si ę równie Ŝ w dymie od papierosów (oszacowana liczba rodników w dymie papierosowym z jednego wydechu wynosi 10 14 !) Przyczyniaj ą si ę do starzenia organizmów Ŝywych i rozkładu Ŝywno ści ale wykorzystujemy je te Ŝ w wielu syntezach organicznych, np. do produkcji lub modyfikacji tworzyw sztucznych. Dawniej powszechnie stosowano termin „wolne rodniki” poniewa Ŝ „rodniki” były jednocze śnie synonimem podstawników zwi ązanych z cz ąsteczk ą zwi ązku chemicznego za pomoc ą wi ąza ń kowalencyjnych. Chcia Ŝ zgodnie z zaleceniami nomenklaturowymi, nie jest konieczne stosowanie okre ślenia „wolne” przed słowem „rodniki”, to jednak w aktualnej literaturze naukowej funkcjonuj ą obydwa terminy. Wyraz „wolny” u Ŝywa si ę te Ŝ w celu podkre ślenia braku wyst ępowania par rodnikowych. Pary rodników (ang. radical pair, geminate pair ) mog ą wyst ępowa ć dzi ęki efektowi „klatki” np. w rozpuszczalnikach. Atom z niesparowanym elektronem zwany jest centrum rodnikowym lub centrum aktywnym . W niektórych układach rodnikowych np. aromatycznych niesparowany elektron jest zdelokalizowany, co przyczynia si ę do jego znacznej trwało ści [4-8]. Rodniki powstaj ą łatwo ze zwi ązków chemicznych zawieraj ących nietrwałe wi ązania kowalencyjne, przykładem których mog ą by ć nadtlenki alkilowe zawieraj ące słabe ugrupowanie nadtlenkowe ulegaj ące p ękaniu zarówno pod wpływem ciepła jak i światła: ∆ lub h ν • • R1–O–O–R2 R1–O + R 2–O (1) Innymi czynnikami wywołuj ącymi reakcje tworzenia rodników s ą: promieniowanie radiacyjne, wyładowania elektryczne, ultrad źwi ęki, siły mechaniczne, a tak Ŝe zwi ązki chemiczne lub biologiczne (np. enzymy). Historyczne znaczenie miało odkrycie wzgl ędnie trwałego rodnika trifenylometanowego powstaj ącego w reakcji trifenylochlorometanu ze srebrem metalicznym: C Cl + Ag + AgCl (3) Rodnik trifenylometanowy nie jest struktur ą płask ą (pier ścienie le Ŝą w płaszczyznach odchylonych od siebie o ok. 30 °). Ze wzgl ędu na przeszkody steryczne, w reakcji rekombinacji (dimeryzacji) ulega on jednocze śnie przegrupowaniu. Rodniki, w których niesparowany elektron zajmuje orbital s lub p są nazywane rodnikami typu σ lub π. W zale Ŝno ści od rodzaju atomu, na którym zlokalizowany jest elektron, mówimy o rodnikach w ęglowych, tlenowych lub rodnikach innych pierwiastków. Ogólnie mo Ŝna podzieli ć rodniki na nieorganiczne i organiczne. • Najprostszy rodnik organiczny – metylowy ( CH 3) ma budow ę płask ą (4), co wynika z 2 hybrydyzacji sp atomu w ęgla, przy czym niezhybrydyzowany orbital 2p z z pojedy ńczym elektronem jest prostopadły do płaszczyzny, w której le Ŝą atomy wodoru i w ęgla [5, 7]. Podstawienie atomu wodoru przez elektroujemny atom (np. chlorowiec) lub grup ę elektronodonorow ą (np. -OH, -NH 2) powoduje przekształcenie struktury płaskiej w piramidaln ą (5). H C H (4) C (5) H F F F Elektron w rodnikach opisywany jest przez pojedy ńczo zaj ęty orbital molekularny SOMO (ang. Singly Occupied Molecular Orbital ), którego energia le Ŝy pomi ędzy orbitalami HOMO ( Highest-energy Occupied Molecular Orbital ) i LUMO ( Lowest-energy Unoccupied Molecular Orbital ). Rodniki mo Ŝna te Ŝ podzieli ć na rodniki o charakterze nukleofilowym i elektrofilowym. Ma to wpływ na oddziaływania z cz ąsteczkami nierodnikowymi drugiego substratu, czyli jego orbitalami HOMO i LUMO (Rys. 1). Energia rodników elektrofilowych jest bli Ŝsza energii HOMO, st ąd tworzenie orbitali molekularnych przedstawionych na rys. 1a. W wyniku reakcji chemicznej rodników o charakterze nukleofilowym (o wy Ŝszej energii SOMO) z innymi cz ąsteczkami dominuj ą oddziaływania z LUMO (rys. 1b). Ten sam rodnik mo Ŝe odgrywa ć rol ę czynnika nukleofilowego lub elektrofilowego w zale Ŝno ści od charakteru drugiego reagenta. Oddziaływania elektrofilowe i nukleofilowe rodników wzrastaj ą gdy energia mi ędzy odpowiednimi orbitalami maleje [5,7]. (a) oddziaływania elektrofilowe (b) oddziaływania nukleofilowe E Rys. 1. Schemat poziomów energetycznych oraz oddziaływania orbitalu SOMO i HOMO (a), SOMO i LUMO (b) [5] Rodniki maj ą wła ściwo ści paramagnetyczne (niezerowy moment magnetyczny, dzi ęki któremu zachodzi porz ądkowanie si ę spinów elektronów zgodnie z liniami zewn ętrznego pola magnetycznego) w odró Ŝnieniu od diamagnetyków, których wszystkie elektrony s ą sparowane. Przenikalno ść magnetyczna ( µ) paramagnetyków jest nieco większa od jedno ści (w przypadku ferromagnetyków µ >> 1). Znane s ą te Ŝ układy, w których wyst ępuj ą dwa centra rodnikowe (tj. dwa niesparowane elektrony), które nie reaguj ą nawzajem ze sob ą. Nazywane s ą birodnikami (biradicals ). W przypadku wyst ępowania niesparowanych elektronów w dwóch stanach o ró Ŝnej multipletowo ści stosuje si ę termin dirodnik ( diradical ). Odkrycie i pierwsze syntezy stabilnych birodników zawdzi ęczamy Wilhelmowi Schlenklowi (1879 – 1943, niemiecki chemik) i Mosesowi Gombergowi (1879 – 1943, prof. University of Michigan, pochodzenia rosyjskiego). O O O O O O O (8) (9) (10) Szczególnym przypadkiem birodników s ą karbeny i nitreny. Karbeny (ang. carbenes ) o wzorze ogólnym :CR 1R2 (np. :CH 2, :CCl 2) są cz ąsteczkami, w których atom w ęgla tworzy dwa wi ązania kowalencyjne z s ąsiednimi atomami. S ą bardzo reaktywnymi zwi ązkami organicznymi (cz ęsto s ą zwi ązkami przej ściowymi, które mo Ŝna otrzyma ć w bardzo niskich temperaturach) wyst ępuj ącymi w stanie singletowym lub trypletowym. Karben w stanie trypletowym jest birodnikiem o hybrydyzacji sp, natomiast w stanie singletowym - sp 2.
