He has statutory authority to examine and report to Parliament on whether departments and the bodies they fund have used their resources efficiently, effectively, and with economy. Our studies evaluate the value for money of public spending, nationally and locally. Our recommendations and reports on good practice help government improve public services, and our work led to audited savings of £1.15 billion in 2014. BIS’s capital investment in science projects Summary 5 Summary 1 The government invests in science to support economic growth, improve national productivity and help the UK take the lead in new markets. The Department for Business, Innovation & Skills (BIS) has overall responsibility for the government’s spending on science, technology and engineering. It also provides funding for a wide range of scientific disciplines and industry sectors with the aim of developing and maintaining the UK’s science and research capability. 2 In 2014-15, BIS allocated £1.1 billion of capital funding to science (Figure 1). This covered its expenditure on major national projects such as oceanographic research ships, supercomputers and research institutes, capital funding for large national research facilities such as particle accelerators, and the UK’s participation in international programmes such as the European Space Agency. It also covered capital funding allocated by the Higher Education Funding Council for England (HEFCE) for laboratories and research facilities in universities. Source: National Audit Offi ce 6 Summary BIS’s capital investment in science projects 3 In December 2014 BIS announced plans for a further £5.9 billion of capital expenditure on science between 2016 and 2021. This included £800 million for new projects, more than £1.2 billion for ongoing national and international projects and a £900 million fund to respond to new challenges as they emerge. It also included plans to spend around £3 billion on the underlying laboratory infrastructure in universities or research institutes. The 2015 Spending Review confirmed this level of future funding. Under the Haldane principle, the government does not take decisions on whether to fund specific projects or individual research proposals. However, ministers have the final say in decisions that involve large-scale, long-term commitments such as the construction of large research facilities. 4 We last reported on the government’s spending on large science facilities in 2007.1 The report was generally positive about BIS’s delivery of a significant capital programme and how it and the research councils had set about prioritising project proposals. Our report did, however, raise concerns about how BIS assessed the ongoing costs of projects and the impact of meeting those costs on the balance of activities funded by the research councils. We also concluded that BIS needed to give more attention to specifying from the start how to assess and measure the success of individual projects. 5 A report published in 2013 by the House of Lords Science and Technology Committee2 raised concerns that the cost of running large-scale infrastructure had not been budgeted for. It also concluded that the potential of the UK’s large-scale scientific resources is being compromised by the lack of a long-term strategic investment plan. In response, BIS launched a public consultation in 2014 to identify strategic priorities for science investment. 6 Reports published in 2015 by the House of Commons Science and Technology Committee and by Sir Paul Nurse acknowledged that the UK research base is world-leading, producing excellent research in a competitive system.3,4 However, they also emphasised the importance of ensuring capital investments are accompanied by sufficient resource funding. Between 2010 and 2014, science resource funding fell in real terms. DNPEP has implicated roles in protein and peptide metabolism, as well as the renin-angiotensin system in blood pressure regulation. Despite previous enzyme and substrate characterization, structural details of DNPEP regarding ligand recognition and catalytic mechanism remain to be delineated. Results: The crystal structure of human DNPEP complexed with zinc and a substrate analogue aspartate-β- hydroxamate reveals a dodecameric machinery built by domain-swapped dimers, in agreement with electron microscopy data. A structural comparison with bacterial homologues identifies unifying catalytic features among the poorly understood M18 enzymes. The bound ligands in the active site also reveal the coordination mode of the binuclear zinc centre and a substrate specificity pocket for acidic amino acids. Conclusions: The DNPEP structure provides a molecular framework to understand its catalysis that is mediated by active site loop swapping, a mechanism likely adopted in other M18 and M42 metallopeptidases that form dodecameric complexes as a self-compartmentalization strategy. Small differences in the substrate binding pocket such as shape and positive charges, the latter conferred by a basic lysine residue, further provide the key to distinguishing substrate preference.

McPhee1, Lin Huang1 & David M.J. Lilley1 Kink turns (k-turns) are widespread elements in RNA that mediate tertiary contacts by kinking the helical axis. We have found that the ability of k-turns to undergo ion-induced folding is conferred by a single base pair that follows the conserved AG pairs, that is, the 3b3n position. A Watson–Crick pair leads to an inability to fold in metal ions alone, while 3n ¼ Gor3b¼ C (but not both) permits folding. Crystallographic study reveals two hydrated metal ions coordinated to O6 of G3n and G2n of Kt-7. Removal of either atom impairs Mg2 þ - induced folding in solution. While SAM-I riboswitches have 3b3n sequences that would predispose them to ion-induced folding, U4 snRNA are strongly biased to an inability to such folding. Thus riboswitch sequences allow folding to occur independently of protein binding, while U4 should remain unfolded until bound by protein. In this work, we focus in OgpA from A. muciniphila,anO-glycopeptidase that exclusively hydrolyzes the peptide bond N-terminal to serine or threonine residues substituted with an O-glycan. We determine the high-resolution X-ray crystal structures of the unliganded form of OgpA, the complex with the glycodrosocin O-glycopeptide substrate and its product, providing a comprehensive set of snapshots of the enzyme along the catalytic cycle. In combination with O-glycopeptide chemistry, enzyme kinetics, and computational methods we unveil the molecular mechanism of O-glycan recognition and specificity for OgpA. We have determined the structure with a number of ligands related to NADH, but in each case only the ADP moiety is observed. The adenosine adopts an anti conformation, and forms multiple hydrogen bonds across the width of the sugar edge of the penultimate C:G base pair of the helix preceding the bulge, and the observed contacts have been confirmed by mutagenesis and calorimetry.
