Clough, Co-Chair Associate Professor Youfu Zhao, Co-Chair Professor Glen L. Hartman ABSTRACT Sclerotinia stem rot, also referred to as white mold, poses a threat to soybean production world-wide. The causal agent, Sclerotinia sclerotiorum (Lib.) de Bary, can survive in harsh environments and has an extremely broad host range. No naturally complete resistance has been reported to exist in soybean, which makes it a compelling problem. Studying the molecular interactions between soybean and S. sclerotiorum is a promising approach to identify the soybeans genes that are controlling the quantitative resistance and, on the other hand, to discover pathogenicity and virulence factors in S. sclerotiorum which could be considered as targets to disable and weaken the aggressiveness of the pathogen. This study utilized RNA-sequencing to characterize the transcriptomes of the leaves of a susceptible soybean line (AC) and an AC transgenic line (OxO) which contains an enzyme that degrades the oxalic acid (OA) produced by S. sclerotiorum thereby conferring resistance. The leaves were infected with S. sclerotiorum and samples were collected at 4 and 8 hours post inoculation (hpi) to characterize this interaction and to examine the key determinants of resistance and infection at these early infection stages. More than 600 soybean genes were detected with a fairly stringent cutoff as being significantly differentially expressed in response to S. -

Accepted 11 June, 2012 Sclerotinia crown and stem rot (SCSR) of alfalfa caused by Sclerotinia trifoliorum is one of the main constraints for efficient alfalfa cultivation in temperate climate all over the world. The resistance of 200 alfalfa accessions to Sclerotinia crown and stem rot was evaluated during 2010 to 2011 in the field nursery established in 2009. The resistance of alfalfa accessions germinating seeds to mycelium of Sclerotinia trifoliorum and oxalic acid (OA) concentrations of 10, 20, 30 mM was screened under laboratory conditions. The statistically significant differences at P<0.05 were determined among evaluated alfalfa accessions resistance to all screened factors. The reactions of alfalfa accessions to disease under field conditions showed that majority of the non-adapted accessions were heavily diseased, whereas the resistant accessions had only negligible disease severity. The germination of accessions seeds at oxalic acid concentration of 30 mM showed strong correlation (r = -0.817**, P<0.01) with SCSR severity in 2011. Among OA concentrations, this one showed the highest correlation rate with SCSR as well as was the least time consuming method. This method of seeds germination on mycelium of S. -

Sclerotinia sclerotiorum, causal agent of Sclerotinia Stem Rot (SSR) in oilseed brassicas, is the disease of most concern to SE US winter carinata, having the potential to reduce yield and ultimately farm-gate income. A disease management plan is currently in development which will include the use of a disease forecasting system. Implementation of a disease forecasting system necessitates determining the optimal environmental conditions for various life-stages of SE US isolates of this pathogen for use in validating or modifying such a system. This is because temperature requirements and optimums for disease onset and subsequent development may vary depending on the geographical origin of S. sclerotiorum isolates. We investigated the optimal conditions necessary for three life-stages; conditioning of sclerotia, germination of apothecia, and ascospore infection of dehiscent carinata petals. Multiple isolates of S. sclerotiorum were tested, collected from winter carinata or canola grown in the SE US. -

GROVES3 Research Branch, Department of Agriculture, Ottawa, Canada Received October 10, 1962 Abstract Three new species of Botryotinia on Caltha palustris L., Ranunculus septen- trionalis Poir., and Ficaria verna Huds. (Ranunculaceae) are described as B. calthae Hennebert and Elliott, B. ranunculi Hennebert and Groves, and B. ficariarum Hennebert. Each of the three species has a Botrytis state of the 73. cinerea complex, and they thus constitute additions to the species already segregated from that complex, i.e. Botryotinia fuckeliana, 73. convoluta, B. draytoni, and B. pelargonii. The Botrytis state of B. ficariarum can be distinguished morp hologically. While B. ranunculi is a North American species and B. ficariarum an European one, B. calthae is reported from both continents. Introduction Botryotinia, a genus of the Sclerotiniaceae, was erected by Whetzel (1945) for four species formerly assigned to Sclerotinia Fuckel, but which differ from the true Sclerotinia species mainly in their erumpent, planoconvexoid sclerotia which are firmly attached to the substrate, and in the possession of a conidial state belonging to the form-genus Botrytis Pers. The type species is Botryotinia convoluta (Drayton) Whetzel in which the conidial state is a Botrytis of the cinerea type or B. cinerea sensu lato, and the other species included were Botryotinia fuckeliana (De Bary) Whetzel, of which the conidial state is Botrytis cinerea Pers. or B. cinerea sensu stricto, and Botryotinia ricini (Godfrey) Whetz. and B. porri (v. Beyma) Whetz. In the latter two species the conidial states would not be considered Botrytis species of the cinerea type. -

Elbieta Patkowska, Mirosaw Konopiski University of Life Sciences in Lublin Abstract. Salsify has a remarkable taste and nutritious values. It is a rich source of inulin – a glycoside which has a positive effect on human and animal organisms. The paper pre- sents studies on the species composition of soil-borne fungi infecting the roots of Tragopogon porrifolius var. sativus cultivated with the use of oats, tansy phacelia and spring vetch as cover crops. In a field experiment the cover crops formed abundant green mass before winter and it constituted a natural mulch on the surface of the plough land. It was managed in two ways: 1) mixed with the soil as a result of spring ploughing, or 2) mixed with the soil as a result of pre-winter ploughing. The conventional cultivation of salsify, i.e. without cover crops, constituted the control. The studies established the number and health status of four-week-old salsify seedlings and roots with necrotic signs. A laboratory mycological analysis made it possible to determine the quantitative and qualitative composition of fungi infecting the underground parts of Tragopogon porri- folius var. sativus. The emergences and the proportion of infected salsify seedlings varied and depended on the species of the mulching plant. The smallest number of infected seed- lings was obtained after the mulch with oats, slightly more after the application of spring vetch or tansy phacelia as cover crops, and the most in the control. -

Here it is provided the updated checklist of the alien vascular fora of Tuscany. Together with those taxa that are considered alien to the Tuscan vascular fora amounting to 510 units, also locally alien taxa and doubtfully aliens are reported in three additional checklists. The analysis of invasiveness shows that 241 taxa are casual, 219 naturalized and 50 invasive. Moreover, 13 taxa are new for the vascular fora of Tuscany, of which one is also new for the Euromediterranean area and two are new for the Mediterranean basin.
