Analytical Cellular Pathology focuses on the burgeoning applications of physical techniques to the study of cells, tissues and organs in disease to elucidate pathogenesis and further translational research, enhanced diagnosis or prognostication, as well as new and better therapies. Of particular relevance are studies that explore the ways that aberrant biochemical and molecular pathways are transduced into the physical processes that underlie pathogenesis.

The journal appeals to an audience of pathologists, cell biologists and biochemists, as well as biomedical engineers by focusing on new applications of the rapidly evolving array of technologies. Papers should be based on novel concepts, novel methodologies or the novel use of existing methodologies that contribute to the study of pathologic processes, rather than methodological details only. Approaches include tools such as biomedical photonics, surface resonance phenomena and impedance tomography, as well as new approaches for ultra-high resolution magnetic resonance, multispectral, hyperspectral and vibrational mode analysis and computational biology models. Hypothesis generating and hypothesis- testing observational studies are also welcome. The recent expansion of work in the field of breast cancer inevitably will hasten discoveries that will impact patient outcome. This research, that spans basic science, clinical medicine, epidemiology and public policy, poses difficulties for investigators. Not only is it necessary to be facile in comprehending ideas from many disciplines, but also, it is important to understand the public implications of these discoveries.

Breast Disease publishes review issues devoted to in-depth analyses of the scientific and public implications of recent research on a specific problem in breast cancer. Meeting the information needs of scientists, clinicians and policy makers, the reviews not only discuss recent discoveries, but also reflect on their impact in breast cancer research or clinical management.

Breast Disease now also publishes unsolicited peer-reviewed articles. Topics can cover basic cellular and molecular biology, epidemiology, genetics, clinical research, imaging and social and legal issues.

Concentrating on molecular biomarkers in cancer research, Cancer Biomarkers publishes original research findings (and reviews solicited by the Editor-in-Chief) on the identification of markers associated with the disease processes, whether or not they are an integral part of the pathological lesion. The disease markers may include, but are not limited to, genomic, epigenomic, proteomics, cellular and morphologic and genetic factors predisposing to the disease or indicating the occurrence of the disease. Manuscripts on these factors or biomarkers, either in altered forms, abnormal concentrations or with abnormal tissue distribution leading to disease causation, are accepted.

Disease Markers publishes original research findings (and reviews solicited by the Editor-in-Chief) on the identification of markers associated with the disease processes, whether or not they are an integral part of the pathological lesion. The disease markers may be a genetic host factor predisposing to the disease or the occurrence of cell-surface markers, enzymes or other components, either in altered forms, abnormal concentrations or with abnormal tissue distribution. This journal is designed to provide a forum for publications dealing with original observations in this developing field on any aspect of the general topic including:

New Approaches to Cell and Tissue Imaging: Pathology at the Crossroads

The evolution of anatomic pathology from a discipline based upon direct observation of visual light images to whole spectrum multi-modality computer-intensive analysis is entering a phase of accelerated growth. However, the technologic base is expanding more rapidly than our ability to exploit it for diagnostic and experimental pathology. While the theoretical and practical underpinnings of the new technology are complex, the underlying principles are quite simple. Energy interactions with a biological specimen basically only involve four kinds of interactions: absorption, reflection, scatter and secondary emission. This holds true, for example, for both a radio wave perturbing aligned molecular dipoles or UV light shining at a slide. Radiologists have been quicker than pathologists at exploiting this generalization. The reasons for this may be found in the pixel level complexity of a microscopic slide as compared to a typical radiological image. The aim in pathology is reconstructing images from the data with resolution good enough for morphologic analysis at the cellular and tissue level, whereas the aim in radiology is to get enough visualization to identify the lesion or location of a biochemical event. Paradoxically, modern radiology deals with both ends of the spectrum; molecules and organs, while pathologists remain focused (pun intended) on cells and tissue architecture, and study molecular events in that context.

The purpose of this book is to present some of the promising new image-based and related technologies for the study of visualization, characterization, and study of abnormal cells and tissues that have evolved over the past few years and to discuss their current and potential applications in experimental pathology and, where feasible, clinical pathologic diagnosis. Just as the flourishing of molecular biology has led to a paradigm shift that has rejuvenated our field, the convergence of biomedicine, bioengineering, and sophisticated new tools of mathematical analysis will move us towards a towards a more quantitative and analytic discipline.

The authors, all experts in the field, have described this technology in a manner accessible to an audience consisting of pathologists, cell biologists and biochemists as well as biomedical engineers, using their own research as well as literature review to provide descriptions and examples of the use of this new armamentarium. Inevitably, these approaches as well as new techniques for in situ molecular imaging that are outside the scope of this book, as well as evolving non-invasive radiological procedures for screening and diagnosis will lead to a convergence of pathology and radiology. The visual pathologic features of tumors have always been the critical parameters in determining tumor type, however, over the last several decades molecular analysis of tumors has played an increasing role in the diagnostic pathology of tumors. Thus, “molecular pathology” has gradually emerged as a discipline in pathology which exploits the use of nucleic acid based techniques.

The purpose of this Handbook is to discuss the principles and the application of selected existing methods of molecular pathology and its use in the detection of early cancer. It describes the molecular changes that characterize preinvasive neoplastic lesions, molecular targets for early detection, validation of molecular targets and areas of new diagnostic technology that show promise either for early detection of neoplasias or other medically important uses in cancer prevention and care. The book describes recent advances in proteomics, metabolomics, stem cells research, micro RNA and other posttranslational molecular modifications; all these will serve the pressing needs in the development of the practice of personalized medicine.

The book is divided into three sections that follow a sequence from basic mechanisms of tumor development, through the molecular and biological abnormalities found in common types of cancers, to the technologies that are most applicable to detecting these biologic alterations in precancerous lesions. Each chapter discusses the scientific underpinnings of the evolution and progression of tumors in order to enable clinicians and other professionals who manage cancer patients to better understand the disease at the molecular and genetic levels.
