VIII Aditivos alimentares na nutrição de juvenis de piavuçu, Leporinus macrocephalus: desempenho e parâmetros hematológicos Resumo Verificou-se o desempenho e a resposta hematológica de juvenis de piavuçu, Leporinus macrocephalus, alimentados com extrato de alho, extrato de canela e levedura submetidos ao estresse de captura. O experimento foi realizado no Laboratório de Alimentação e Nutrição de Peixes (Aquanut), Ilhéus, Bahia, onde 192 juvenis de piavuçu com peso médio inicial de 3,09 ± 0,8 g foram distribuídos em 16 tanques de fibra de vidro (170 L), com densidade de 12 peixes/tanque em um delineamento inteiramente casualizado, com quatro tratamentos e quatro repetições. Os tratamentos consistiam na adição de 1% de extrato de alho, 1% de extrato de canela, 9x105 UFC.g-1 de levedura Saccharomyces cerevisiae DS-5 e uma dieta controle, sem a inclusão de aditivo. Foram avaliados o desempenho, índices corporais e sobrevivência dos peixes. Após o ensaio de desempenho os peixes foram submetidos ao estresse de captura e o efeito dos aditivos como mitigador do estresse foi avaliado por meio de análises hematológicas. Observou-se maior ganho de peso e melhor conversão alimentar (P≤0,05) nos peixes alimentados com as dietas contendo os extratos de alho e de canela. Foram verificadas alterações nos números de leucócitos logo após o estresse nos peixes de todos os tratamentos. Foram observadas reduções nos números de trombócitos nos peixes suplementados com extrato de alho e dieta controle 24 horas após o estresse. Juvenis de piavuçu alimentados com dietas suplementadas com os extratos de alho e de canela ao nível de 10 g. Kg-1 apresentaram melhor desempenho, entretanto nas concentrações utilizadas esses aditivos não influenciaram nos parâmetros hematológicos quando os peixes foram submetidos ao estresse de captura. Palavras chave: aquicultura, probiótico, fitogênico, desempenho IX Food additives in the nutrition juveniles of piavuçu, Leporinus macrocephalus: performance and hematological parameters Abstract It was verified the juveniles piavuçu’s performance and hematological response, Leporinus macrocephalus fed with garlic extract, cinnamon extract and yeast subjected to capture stress. The experiment was conducted at the laboratory of Food and Nutrition of Fish (Aquanut) Ilhéus, Bahia, where 192 juveniles piavuçu with an average initial weight of 3.09 ± 0.8 g were distributed in 16 tanks (170L), with a density of 12 fish per tank in a completely randomized design with four treatments and four replications. The treatments consisted of the addition of 1% of garlic extract, 1% extract of cinnamon, 9x105 CFU.g-1 of yeast Saccharomyces cerevisiae DS-5 and a control diet without the inclusion of additive. Was evaluated the performance, body index and fish survival. After the performance test the fish were subjected to the stress of capture and the effect of additives such as mitigating stress was assessed by hematological analysis. Was noted more weight gain and improved feed conversion (P ≤ 0.05) in fish fed with diets containing extracts from garlic and cinnamon. Changes were observed in numbers of leukocytes after the stress on the fish in all treatments. Reductions in the number of thrombocytes in fish supplemented with garlic extract and control diet 24 hours after stress. Piavuçu juveniles fed diets supplemented with extracts of garlic and cinnamon to the level of 10 g. Kg-1 had better performance, however the concentrations used in these additives did not affect hematologic parameters when fish were subjected to the stress of capture. X Lista de tabelas Tabela 1. However, bacteria of this genus are isolated from many other habitats, environments, and food products. The taxonomy of this genus is complex when phenotypic identification methods are used because such methods might not correctly identify all the species. On the other hand, molecular methods have proven very reliable, such as using the sequences of concatenated housekeeping genes like gyrB and rpoD or comparing the genomes with the type strains using a genomic index, such as the average nucleotide identity (ANI) or in silico DNA–DNA hybridization (isDDH). So far, 36 species have been described in the genus Aeromonas of which at least 19 are considered emerging pathogens to humans, causing a broad spectrum of infections. Having said that, when classifying 1852 strains that have been reported in various recent clinical cases, 95.4% were identified as only four species: Aeromonas caviae (37.26%), Aeromonas dhakensis (23.49%), Aeromonas veronii (21.54%), and Aeromonas hydrophila (13.07%).

The results obtained were compared to that of the gold standard method, which used 16S rDNA sequences. Methodology and results: Based on the Indonesian government standard identification method SNI7303, we identified 56 out of 95 samples as A. hydrophila. The samples were then screened using the PCR amplification approach developed by Dorsch and Cascón. Of the 56 samples, only 20 samples were found to be positive by either the Dorsch or Cascón methods. DNA from these 20 samples was amplified using common 16S rDNA primers and the sequences compared with available 16S rDNA sequences from the GenBank.

The production of putrescine, diaminopropane, cadaverine, and agmatine are not exactly correlated to the phylogenetic genospecies within 36 strains of the genus Aeromonas (the family Aeromon- adaceae) lacking in triamines. The occurrence of norspermidine was limited but not ubiquitous within the family Vibrionaceae, including 20 strains of Vibrio, Listonella, Photobacterium, and Salini- vibrio. Spermidine was not substituted for the absence of norspermidine in the family. Agmatine was detected only in Photobacterium. Salinivibrio and some strains of Vibrio were devoid of polyamines. Vibrio ("Moritella") marinus contained cadaverine. Within the family Pasteurellaceae, Haemophilus contained cadaverine only and Actinobacillus contained no polyamine. Halomonas, Chromohalobacter, and Zymobacter, belonging to the family Halomonadaceae, ubiquitously con- tained spermidine and sporadically cadaverine and agmatine. Treated hospital waste effluents may contain pathogenic and drug resistant bacteria, which constitutes the most dangerous single risk factor for dissemination of pathogenic and drug resistant organisms to the environment. This thesis focuses on the possibility of persistence and transmission of pathogenic and drug resistant bacteria like Aeromonas and Enterococcus from wastewater to environment and to humans, especially in relation to a sewage treatment process based on waste water recycling. In one study (Paper I), we have shown that a prevalent ampicillin and ciprofloxacin resistant clonal lineage of Enterococcus faecium in Swedish hospital sewage water may be transmitted from hospital patients to hospital sewage water. Recycling of hospital sewage water may in the second step disseminate the disease-causing organisms to the environment and in the third step to the human food chain. In other studies (Papers III, V), we have investigated the persistence of Aeromonas in a duckweed aquaculture based hospital sewage water treatment plant, where sewage grown duckweed is used as fish food.

Aeromonas species are ubiquitous bacteria in terrestrial and aquatic milieus. They are becoming renowned as enteric pathogens of serious public health concern as they acquire a number of virulence determinants that are linked with human diseases, such as gastroenteritis, soft-tissue, muscle infections, septicemia, and skin diseases. Proper sanitary procedures are essential in the prevention of the spread of Aeromonas infections. Oral fluid electrolyte substitution is employed in the prevention of dehydration, and broad-spectrum antibiotics are used in severe Aeromonas outbreaks. This review presents an overview of emerging Aeromonas infections and proposes the need for actions necessary for establishing adequate prevention measures against the infections.
