Abstract Tra catfish (Pangasianodon hypophthalmus) currently is the most important freshwater indigenous teleost cultured in the aquaculture sector in the Mekong River Delta (MRD) in Vietnam and it is now an icon of aquaculture development in Vietnam and globally. Over decades, the tra catfish industry has developed rapidly to a point where annual production now exceeds one million tonnes, employs nearly 200,000 rural poor, and resulted in export income of US$ 1.4 billion in 2010 (De Silva and Phuong, 2011). This industry however, has been identified as one that is likely to be impacted by predicted climate change scenarios because the MRD has been identified as among the top three most likely seriously affected regions around the world (IPCC, 2007). Predicted changes largely are related to water temperature increases caused by global warming and raised salinity caused by saline intrusion from sea level rise. The current situation highlights the need to assess potential effects of climate change on the tra catfish culture industry and to identify appropriate ways that will allow sustainable development of this important industry. The current study examined the responses of replicate cohorts of juvenile tra catfish to sublethal salinity and temperature change in three trials: (1) effects of raised sublethal salinities (freshwater, 2, 6, 10, 14 and 18‰); (2) effects of temperature levels (24, current ambient (27-28oC), 30, 32, 34, and 36oC); and (3) combined effects of temperature (25, 30, and 35oC) and salinity (freshwater, 6, and 12‰). Indicators for assessing effects of climate change on tra catfish performance included survival rate, growth performance, food conversion ratio, hormonal response (cortisol, growth hormone/IGF-1), osmoregulatory, plasma glucose and plasma ion concentrations, hematological parameters, and growth hormone gene expression patterns. Results of the first trial showed that tra catfish had high survival rates, better growth performance and better FCR values in a salinity range from freshwater to 10‰. Cortisol levels were elevated in the 14 and 18‰ treatments after 6 h and reached a peak after 24-h exposure, and this also led to increases in plasma glucose concentration. While fish in high salinity treatments did recover over a 14 days period, high salinity conditions had long- term negative effects on growth performance and FCR values. In trial 2, low temperature (24oC) significantly affected fish growth rate and FCR value while high sublethal i temperatures led to enhanced growth performance until 34oC where it peaked. Results of the 3rd trial showed that changes in temperature, salinity and their interaction had significant effects on individual growth rate and feed conversion efficiency of fish in the 35°C-6‰ treatment producing optimal conditions for growth while not producing significantly higher FCR values in comparison with a control treatment (freshwater - ambient temperature (25oC)). Tra catfish showed an effective capacity to acclimate successfully to changes in external environment factors (temperature and salinity), that included changing red blood cell count, hemoglobin concentration, metabolic rate, and energy mobilisation. Results of this study suggest that tra catfish can acclimate relatively easily to environmental conditions that have been predicted under current climate change models for the MRD. This provides strong indication that natural tra populations have the potential to adapt to higher salinity and temperature regimes relatively rapidly and therefore may persist successfully in the MRD for the long term. Furthermore, these data suggest that variation in genes underlying stress response to temperature and salinity that currently exist could facilitate development of culture strains that will perform well in the tra culture industry into the future. ii Notes on Thesis Preparation Chapters 2 to 4 of this thesis are presented as either published papers or as submitted manuscripts for publication. Results of the fish landed catch survey in 11 coastal towns of the lake showed a decreasing fish harvest in the open fisheries from 1,420 MT to 460 MT in 1996 to 2011. Inventory of fisherfolk, boat, and gear also decreased to 16%, 7%, and 39%, respectively from 1998 to 2011. The most dominant gear is gill net which is about 53% of the total gear used in the lake with a declining catch per unit effort (CPUE) of 11kg/day to 4 kg/day from 1997 to 2011. Active gear such as motorized push net, ring net, and beach seine also operated in the lake with a CPUE ranging from 48 kg/day to 2,504 kg/day. There were 43 fish species identified in which S. tawilis dominated the catch for the last decade. However, its harvest also declined from 744 to 71 mt in 1996 to 2011. The presence of alien species such as jaguar fish, pangasius, and black-chinned tilapia amplified in 2009.

Catfish constitute a valuable source of dietary protein. Therefore, they are cultured worldwide, and P. hypophthalmus is a food staple in the Mekong area. However, genetic information about the culture stock, is unavailable for breeding improvement, although genetics of the channel catfish, Ictalurus punctatus, has been reported. Toacquiregenomesequencedataasausefulresourcefor marker-assisted breeding, we decoded a draft genome of P. hypophthalmus and performed comparative analyses. Results: Using the Illumina platform, we obtained both nuclear and mitochondrial DNA sequences. Molecular phylogeny using the mitochondrial genome confirmed that P. hypophthalmus is a member of the family Pangasiidae and is nested within a clade including the families Cranoglanididae and Ictaluridae. The nuclear genome was estimated at approximately 700 Mb, assembled into 568 scaffolds with an N50 of 14.29 Mbp, and was estimated to contain ~ 28,600 protein-coding genes, comparable to those of channel catfish and zebrafish. Interestingly, zebrafish produce gadusol, but genes for biosynthesis of this sunscreen compound have been lost from catfish genomes.

    Employing Geographical Information Systems in Fisheries Management in the Mekong River: a case study of Lao PDR Kaviphone Phouthavongs A thesis submitted in partial fulfilment of the requirement for the Degree of Master of Science School of Geosciences University of Sydney June 2006 ABSTRACT The objective of this research is to employ Geographical Information Systems to fisheries management in the Mekong River Basin. The study uses artisanal fisheries practices in Khong district, Champasack province Lao PDR as a case study. The research focuses on integrating indigenous and scientific knowledge in fisheries management; how local communities use indigenous knowledge to access and manage their fish conservation zones; and the contribution of scientific knowledge to fishery co-management practices at village level. Specific attention is paid to how GIS can aid the integration of these two knowledge systems into a sustainable management system for fisheries resources. Fieldwork was conducted in three villages in the Khong district, Champasack province and Catch per Unit of Effort / hydro-acoustic data collected by the Living Aquatic Resources Research Centre was used to analyse and look at the differences and/or similarities between indigenous and scientific knowledge which can supplement each other and be used for small scale fisheries management. The results show that GIS has the potential not only for data storage and visualisation, but also as a tool to combine scientific and indigenous knowledge in digital maps. Integrating indigenous knowledge into a GIS framework can strengthen indigenous knowledge, from un processed data to information that scientists and decision-makers can easily access and use as a supplement to scientific knowledge in aquatic resource decision-making and planning across different levels.

Demidospermus spirophallus n. sp. differed from other congeners mainly because of the morphology of the male copulatory organ (MCO), which exhibited 2½ counterclockwise rings, a tubular accessory piece with one bifurcated end and a weakly sclerotized vagina with sinistral open- ing. Demidospermus prolixus n. sp. presents a counterclockwise-coiled MCO with 1½ rings, an ovate base, a non-articulated groove-like accessory piece serving as an MCO guide, two different hook shapes, inconspicuous tegumental annulations, a non-sclerotized vagina with sinistral opening and the absence of eyes or acces- sory eyespots. Nowadays, three species, Pangasius hypophtahlmus, P. boucorti, and P. djambal, are used in aquaculture. Among the genera in Pangasiidae, Helicophagus was less studied. Although this genus was less preferred than other popular species in Pangasiidae, it still has high commercial price. The present study was conducted to clarify the differences of the exist species in the genus Helicophagus based on biometric analyses. Twenty six specimens, collected from represent rivers in Southeast Asia, used for the material examined. Several type specimens deposited in museums were also added in the analyses. Thirty five characters were designed for measurement on the unique body conformation. Principal component analysis (PCA) was applied to distinguish different species and found strong characters for key identification and description. The results presented the data and information on the diagnosis, description, distribution, and ecology of each species.
