Metode yang digunakan untuk identifikasi ikan yang sangat cepat dan akurat sangat diperlukan. DNA barcoding adalah teknik identifikasi baru dengan pendekatan molekular. Gen CO1 diamplifikasi dan produk PCR disekuensing serta dianalisis dengan menggunakan software bioinformatika. Pengeditan hasil sekuensing dan penentuan komposisi nukleotida dianalisis dengan software Mega5. Urutan DNA disejajarkan dengan ClustalW vers. 1.4. Sekuens dibandingkan dengan data GenBank menggunakan BLAST (Basic Local Aligment Search Tools) dan BOLDSystems. Pohon filogenetik dibuat dengan menggunakan metode Neighbor_Joining. Salah satu contoh identifikasi ikan dari genus Mystus yang telah dikonfirmasi adalah Mystus nigriceps menjadi Mystus singaringan. Kata Kunci : Identifikasi, Mystus, DNA barcode ABSTRACT The fish of the Bagridae Family in Indonesia reach until 60 species and one of them is from the genus Msytus. The method is used for fast and accurate species identification was needed. DNA barcoding was a new identification method with molecularly approach. The CO1 gene was amplified and PCR products were sequenced and analyzed using bioinformatics software. Editing of sequencing results and determining the nucleotide composition were analyzed with Mega5 software. The DNA sequence was aligned with ClustalW vers. 1.4. Sequences are compared with GenBank data using BLAST (Basic Local Aligment Search Tools) and BOLDSystems. Phylogenetic tree was made using Neighbor_Joining method. One example of the identification of fish from the confirmed Mystus genus is Mystus nigriceps being the Mystus singaringan. Two vessels of a domestic shipping line appear to have served as a transport vector for the Asian green mussel Perna viridis (Linnaeus, 1758) between regions. This species is indigenous in the western but not in the eastern part of the archipelago, separated historically by the Sunda Shelf. The green mussels collected from the hulls of the ferries when in eastern Indonesia showed a significantly lower body condition index than similar-sized individuals from three different western-Indonesian mussel populations. This was presumably due to reduced food supply during the ships’ voyages. Although this transport- induced food shortage may initially limit the invasive potential (through reduced reproductive rates) of the translocated individuals, the risk that the species will extend its distributional range further into eastern Indonesia is high.

Phytoplankton pigment analysis by using High Performance Liquid Chromatographer (HPLC) resulted in a set of pigments of chlorophyll-a (Chl-a), chlorophyll-b (Chl-b), chlorophyll-c (Chl-c), lutein, zeaxanthin, fucoxanthin, peridinin, diadinoxanthin, and ß-carotene. Linear multi regression and multivariate principal component analysis (PCA) showed that algae pigments correlate positively with nutrients and are not significantly correlated with underwater light and water transparency, suggesting important roles of nutrients for phytoplankton development in tropical estuaries.

The observation result shows the variability of sea surface chlorophyll-a in Lampung Bay seems to be correlated with the monsoonal system that influences on the variability of the meteorological and oceanographic situation. In the wet season (northwest monsoon), the concentration of chlorophyll-a is high. It was suspected due to high precipitation during this season in which the coastal area was enriched by the nutrient load from the upper land area through the rivers and coastal discharge. While in the dry season (southeast monsoon), high concen- tration of the chlorophyll-a is stimulated by increasing nutrient concentration due to the upwelling. During these seasons, the abundance of the pelagic fish was high within this region. Pasir Putih I no. 1 Ancol Timur-Jakarta Utara 14430, Indonesia Received May 23, 2012; Accepted October 30, 2012 ABSTRACT This research aims to investigate the quality of the Tambelan island waters as primary information to support these islands as a marine conservation area. Some chemical properties of seawater such as pH and DO (Dissolved Oxygen) were identified by portable instrumentations. Furthermore inorganic nutrients phosphate, nitrate, and silicate contents were measured according to the methods developed by Morris and Riley in 1963 and then modified by Strickland and Parsons in 1968. The results of the research show that the pH values were in the range of 8.06 to 8.30, and the DO level amounted to 5.26 to 6.31 mg/L. Further results from this research show that the content of phosphate and nitrate ranged from 0.001 to 0.004 mg/L and 0.005 to 0.018 mg/L, respectively. Meanwhile, the content of silicate was found to be 0.063 to 0.134 mg/L in the archipelagic waters of Tambelan. The values of the above parameters are in accordance with the range of the seawater quality as recommended by the Ministry of Environment of the Republic of Indonesia.

Selain itu di lokasi ini terdapat situs arkeologi maritim, sehingga kawasan ini merupakan kawasan yang cocok untuk diusulkan menjadi kawasan marine eco archeo park. Penelitian awal kualitas perairan dilakukan pada Mei 2014 secara purposive sampling dengan menggunakan alat multiparameter secara insitu dan analisis sampel air di laboratorium. Parameter yang diukur yaitu salinitas, pH, turbiditas, padatan tersuspensi (TSS), Biological Oxygen Demand (BOD5), nitrat, tembaga dan nikel. Analisis data dilakukan secara deskriptif dengan menggunakan analisis Principal Component Analysis. Hasil laboratorium menunjukkan bahwa semua parameter masih sesuai dengan KepmenLH no 51 tahun 2004, kecuali nilai tembaga pada saat pengukuran tidak sesuai, walaupun secara keseluruhan perairan Selat Dampier pada saat pengukuran masih dalam kondisi baik sebagai daerah taman nasional dan wisata bahari.
