RNDr. Barbora Mieslerová, Ph.D. Rok obhajoby práce: 2016 Abstrakt: Tato diplomová práce je zaměřená na vybrané invazivní druhy padlí řádu Erysiphales v České republice. Teoretická část je věnována charakteristice řádu Erysiphales včetně popisu životního cyklu, morfologických struktur, geografického rozšíření, epidemiologii a významu padlí. Praktická část se zabývá studiem invazivních druhů padlí na území České republiky. Jsou zde popsány materiály a metodika sběru, tzn. uchování a příprava vzorků a následné měření velikostí konidií, konidioforů, bazální buňky konidioforů, počet distálních buněk na konidioforu, průměr chasmothecií, délka a počet apendixů a počet a velikost vřecek. U každého druhu padlí se sledovala fenologie výskytu v průběhu roku 2015. Morfologické charakteristiky byly srovnávány s údaji z monografie Braun a Cook (2012) a z vědeckých článků a byl určován druh padlí, kterým byla hostitelská rostlina infikována. Všechny druhy padlí se podařilo identifikovat podle monografie padlí (Braun a Cook, 2012). RNDr. Barbora Mieslerová, Ph.D. The year of presentation: 2016 Abstract: The thesis is focused on selected invasive species of powdery mildews of order Erysiphales in the Czech Republic. The theoretical part is focused on the characterization of the order Erysiphales including the description of the life cycle, morphological structures, geographic occurence, epidemiology and importance of powdery mildew. The practical part deals with the study of invasive species of powdery mildew in the Czech Republic. There are described materials and collection methodology, i.e. the preservation and the preparation of samples and subsequent measurement of the size of conidia, conidiophores, basal cell of conidiophores, number of distal cells on a conidiophore, diameter of chasmothecia, length and number of appendices and the number and size of asci described in the thesis. Phenology occurrence was monitored for each species of powdery mildew during 2015. Morphological characteristics were compared with data contained in Braun and Cook Monography (2012) and scientific articles and the kind of powdery mildew which infected the host plant was determined. All powdery mildew species were identified in accordance with Powdery Mildew Monography (Braun and Cook, 2012). Choroby, které způsobují, jsou běžné a snadno rozpoznatelné. Infikované rostliny mají bílé skvrny a později rozsáhlé práškovité povlaky na listech i stoncích. Svrchní část listů je obvykle nejvíce zasažena, ale houba se může objevit i na různých částech nadzemních orgánů rostlin. Jak nemoc postupuje, zvětšují se bílé moučnaté skvrny i hustota napadení. Padlí dobře roste v prostředí s vyšší vlhkostí a při mírných teplotách, což platí i pro Českou republiku. Proto vytváří jak nepohlavní stádia, která jsou charakterizována vznikem konidií, tak pohlavní stádia, která zahrnují vznik kulovitých chasmothecií s přívěsky. Jednotlivé druhy padlí se liší šířkou hostitelského spektra. Mezi nejhojnější druhy padlí řadíme ty, které mají úzké spektrum

These include the uplands of the Clees, Stiperstones and Long Mynd with their associated heath land, forested land such as the Forest of Wyre and the Mortimer Forest, the lowland bogs and meres in the north of the county, and agricultural land scattered with small woodlands and copses. This diversity makes Shropshire unique. The Shropshire Fungus Group has been in existence for 18 years. (Inaugural meeting 6th December 1992. The aim was to produce a fungus flora for the county. This aim has not yet been realised for a number of reasons, chief amongst these are manpower and cost. The group has however collected many records by trawling the archives, contributions from interested individuals/groups, and by field meetings. It is these records that are published here. The first Shropshire checklist was published in 1997. Many more records have now been added and nearly 40,000 of these have now been added to the national British Mycological Society’s database, the Fungus Record Database for Britain and Ireland (FRDBI). During this ten year period molecular biology, i.e. DNA analysis has been applied to fungal classification.

Information on the records of a new adventive powdery mildew fungus Erysiphe corylacearum (Erysiphales, Ascomycota) in Ukraine is provided. The fungus was found on Corylus avellana (Betulaceae) in Kyiv, as well as in Crimea. Current distribution of this species outside its primary range is analyzed. It has been shown that the fungus came to Ukraine from the East through the Caucasus. Ukrainian specimens were compared with samples collected in the Far East of Russia. It was found that Ukrainian specimens have slightly larger chasmothecia, somewhat more numerous asci, which are slightly more elongated and often short stalked. It is noted that the fungus has a great invasive potential and in the future can cause considerable damage to the forestry of European countries. The article is illustrated with micrographs obtained under light and scanning electron microscopes.     Polish Botanical Journal 47(2): 215–222, 2002 NEW COLLECTIONS OF POWDERY MILDEWS (ERYSIPHALES) IN POLAND AGATA WOŁCZAN´ SKA & WIESŁAW MUŁENKO Abstract: New hosts and records of powdery mildews (Erysiphales) in Poland are discussed. Erysiphe adunca (Wallr.) Fr. was found on a host new for this fungus: Salix repens L. subsp. arenaria (L.) Hiitonen. Two other species – Golovinomyces cichoracea- rum (DC.) V. P. Gelyuta var. cichoracearum (on Chondrilla juncea L.) and Erysiphe syringae Schwein. (on Ligustrum vulgare L.) – were collected on host plants new for Poland. The anamorphic as well as teleomorphic (cleistothecia with ascospores) state of Erysiphe syringae was observed on Ligustrum vulgare and Syringa vulgaris L.; hitherto this fungus has formed mature cleistothecia only in North America. Erysiphe flexuosa (Peck) U. Braun & S. Takamatsu has been introduced from North America to Europe recently. Aesculus pavia L. and A. × carnea Hayne are new hosts for this fungus in Poland. Golovinomyces cichoracearum var. cichoracearum [on Cirsium canum (L.) All.] and Podosphaera spiraeae (Sawada) U. Braun & S. Takamatsu (on Spiraea cf. japonica L.) are recorded on rare host plants. Characterising plant pathogen communities and their environmental drivers at a national scale A thesis submitted in partial fulfilment of the requirements for the Degree of Doctor of Philosophy at Lincoln University by Andreas Makiola Lincoln University, New Zealand 2019 General abstract Plant pathogens play a critical role for global food security, conservation of natural ecosystems and future resilience and sustainability of ecosystem services in general. Thus, it is crucial to understand the large-scale processes that shape plant pathogen communities. The recent drop in DNA sequencing costs offers, for the first time, the opportunity to study multiple plant pathogens simultaneously in their naturally occurring environment effectively at large scale. In this thesis, my aims were (1) to employ next-generation sequencing (NGS) based metabarcoding for the detection and identification of plant pathogens at the ecosystem scale in New Zealand, (2) to characterise plant pathogen communities, and (3) to determine the environmental drivers of these communities. First, I investigated the suitability of NGS for the detection, identification and quantification of plant pathogens using rust fungi as a model system.

Abstract: Erysiphe flexuosa, a powdery mildew parasite of Aesculus species, is believed to have originated in North America where distribution records were confined to regions east of the Rocky Mountains. The fungus recently was found in eastern Washington State and northern Idaho. The fungus can be distinguished readily from other powdery mildews attacking Aesculus species by the presence of two distinctive kinds of chasmothecial appendages. Consistent with previous reports on similar species, observations determined that subulate appendages formed first, on dorsal sides of chasmothecia. The longer, uncinate appendages formed equatorially during later chasmothecial maturation. There are no Aesculus species native to northwest North America that could have hosted an undetected population of the fungus. It appears likely that E. flexuosa became established relatively recently in the inland Pacific Northwest, possibly being introduced on infected host material, similar to the situation in Europe where several reports documented the presence of the fungus beginning in 2000.
