Flavoenzymes Riboflavin (vitamin B2)i s anessentia l vitamin for many life forms. Only plants and bacteria are able toproduc e this yellow (lat: flavus =yellow ) pigment denovo. Mammals , on the other hand, have to take up riboflavin in their food. In humans, riboflavin is absorbed by the gut and is transported, partially bound to serum albumin, via the bloodstream (Decker, 1994). By the subsequent action of flavokinase and FAD synthase, riboflavin isconverte d intoth ecofactors : flavin mononucleotide (FMN) andflavi n adenine dinucleotide (FAD), respectively (Scheme 1). These cofactors are incorporated into the so-called flavoenzymes thatnee dthes ecofactor s for their activity.A strikin gfeatur e of this group of enzymes isth e diversity of reactions that are catalyzed which ranges from redox catalysis andDN Arepai rt o light emission (Ghisla andMassey , 1989). Reaction mechanism Biological oxidation reactions mostly involve the rupture of a C-H bond with a concomitant transfer of twoelectron st oa nelectro n acceptor likeNAD(P) +,cytochromes ,o r FAD. Flavin-mediated oxidation reactions vary from relatively simple oxidation reactions, like alcohol oxidations,t omor ecomple x reactions,e.g . stereoselective oxidative cyclization of plant alkaloids (Kutchan andDittrich , 1995). E-FAD S + 02 - P + H202 (1) Reactions catalyzed by flavoprotein oxidases generally include two substrates (Equation 1): an electron donor (substrate) and molecular oxygen. Therefore, these enzymatic reactions can be characterized as two-substrate two-product reactions. As a consequence, most flavoprotein oxidases obey aping-pon g mechanism ora ternar y complex mechanism. The type of kinetic mechanism varies between different flavoprotein oxidases and can depend on the type of substrate (Ramsay, 1991;Fraaij e and van Berkel, 1997). Kinetic analysis of the different enzymes hasreveale d thatth e rate-limiting stepi n catalysis is often represented by either the rate of flavin reduction or the rate of product release. As for the type of mechanism, the rate-limiting step may vary depending on the type of substrate.Furthermore , in the case ofD-amin o acid oxidase, itha sbee n found that with the yeast enzyme therat eo f flavin reduction israte-limitin g while withth e mammalian enzyme product release limits turnover (Pollegioni et al., 1993; Vanoni et al., 1997). Taken together, flavoprotein oxidases arequit e variable with respect tothei r kineticproperties . Flavoprotein oxidase catalysis involves two half-reactions in which first the flavin is reduced by the electron donor (substrate) (reductive half-reaction) and subsequently the reduced flavin is reoxidized by molecular oxygen (oxidative half-reaction). Using the stopped-flow technique,thes etw ohalf-reaction s can beanalyze d separately. - reductive half-reaction + E0x S -«— Eox ~S-«— EreCj~ P *• EreC|+ P (2) k-i k_2 By anaerobically mixing the oxidized enzyme with substrate, the reductive half- reaction can be studied. Thereactio n includes thefirs t step in thecatalyti c cycle:bindin go f substrate to form the Michaelis-Menten complex (E0x~S) (Equation 1).I n the succeeding Introduction step the flavin cofactor is reduced by the transfer of two electrons from the substrate. The mechanism by which these electrons are transferred to the N5 atom of the flavin isoalloxazine ring is still a matter of discussion. Three types of mechanisms have been proposed. A carbanion mechanism is thought to be operative when the C-H bond to be oxidized is activated. In that case, the reaction proceeds by abstraction of ahydroge n as a proton to form a carbanion. Subsequently, a N5 flavin adduct is transiently formed by which the electrons are transferred to the flavin (Porter et al., 1973).A radica l mechanism involvingth etransfe r of aproto n andtw osingl eelectron sfro m the 'non-activated' substrate toth e oxidized flavin hasbee n proposed for e.g. monoamine oxidase andmethano l oxidase (Silverman, 1995;Sherr y and Abeles, 1985). Another alternative mechanism is the direct transfer of a hydride to the flavin ring system similar to the well-established reduction of pyridine nucleotides (Mattevi et al., 1996; Pollegioni et al., 1997). Depending on the enzyme, the formed product or product intermediate may be released from the active-site before the reduced flavin is reoxidized (£3) by molecular oxygen. Like other phenolic substances, vanillin has antimicrobial activity and can be extracted from lignin either by a thermo-chemical process or through microbial degradation. Vanillin, can serve as a model monomer in biodegradation studies of lignin. In the present study, a yeast isolated from decaying wood on the Faroe Islands, was identifed as Cystobasidium laryngis strain FMYD002, based on internal transcribed spacer sequence analysis. It dem- onstrated the ability to convert vanillin to vanillyl alcohol, as detected by ultra-high performance liquid chromatogra- phy–quadrupole-time-of-fight. Structural analysis of vanillyl alcohol was carried out by using gas chromatography– mass spectrometry and 1H NMR spectroscopy, and further verifed by synthesis. The reduction of vanillin to vanillyl alcohol has been documented for only a few species of fungi. However, to our knowledge, this biotransformation has not yet been reported for basidiomycetous yeast species, nor for any representative of the subphylum Pucciniomyco- tina. The biotransformation capability of the present strain might prove useful in the industrial utilisation of lignocel- lulosic residues. The reaction product from vanillyl alcohol was identified as vanillin as judged by its melting point, elemental analysis and IR, mass and NMR spectra. The molecular weight of the enzyme was estimated to be approximately 72,000 as determined by gel filtration and the isoelectric point was pH 6.01. The most characteristic feature of this enzyme is its wide substrate specificity range. The enzyme catalyzes the dehydrogenation of various aromatic and aliphatic alcohols and aldehydes with phenazine methosulfate as electron acceptor. The active substrates of this enzyme are as follows: Vanillyl alcohol, benzyl alcohol, cinnamyl alcohol, 2-phenylethanol, 2-phenoxyethanol, aliphatic alcohols of C2 to G8, rr<m?-cinnamaldehyde, formaldehyde, propionaldehyde and butyraldehyde. The highest activities were obtained with vanillyl alcohol, rc-propanol and ^-butanol at the same level. The apparent Kmvalues were as follows: 112/zm for vanillyl alcohol, 7/jm for benzyl alcohol, 180/im for «-propanol, 14^m for #-butanol, 20/zm for 2-phenoxyethanol, 12/iM for 2-phenylethanol and 105 /zm for butyraldehyde. These activities were confirmed to be catalyzed by a single enzyme by activity staining on polyacrylamide gels.

However, attempts to engineer platform micro- organisms to utilize methanol mainly focus on enzyme and pathway engineering. Herein, we enhanced methanol bioconversion of synthetic methylotrophs by improving cellular tolerance to methanol. A previously engineered methanol-dependent Corynebacterium glutamicum is subjected to adaptive laboratory evolution with elevated methanol content. Unexpectedly, the evolved strain not only tolerates higher concentrations of methanol but also shows improved growth and methanol utilization. Transcriptome analysis suggests increased methanol con- centrations rebalance methylotrophic metabolism by down-regulating glycolysis and up- regulating amino acid biosynthesis, oxidative phosphorylation, ribosome biosynthesis, and parts of TCA cycle. Mutations in the O-acetyl-L-homoserine sulfhydrylase Cgl0653 catalyzing formation of L-methionine analog from methanol and methanol-induced membrane-bound transporter Cgl0833 are proven crucial for methanol tolerance. To create the Aldrich Raman Condensed Phase Library, 14,033 compounds found in the Aldrich Collection of FT-IR Spectra Edition II Library were excited with an Nd:YVO4 laser (1064 nm) using laser powers between 400 - 600 mW, measured at the sample. A Thermo FT-Raman spectrometer (with a Ge detector) was used to collect the Raman spectra. The spectra were saved in Raman Shift format. The immune complexes are deposited in the glomerular mesangium, leading to inflammation and loss of renal function, but the complete pathophysiology of the disease is not understood. Using an integrated global transcriptomic and proteomic profiling approach, we investigated the role of the mesangium in the onset and progression of IgAN. Global gene expression was investigated by microarray analysis of the glomerular compartment of renal biopsy specimens from patients with IgAN (n=19) and controls (n=22). Using curated glomerular cell type–specific genes from the published literature, we found differential expression of a much higher percentage of mesangial cell–positive standard genes than podocyte-positive standard genes in IgAN. Principal coordinate analysis of expression data revealed clear separation of patient and control samples on the basis of mesangial but not podocyte cell–positive standard genes.

Although alcohol oxidases generate hydrogen peroxide when they turn over, this issue can be alleviated by addition of catalase, which, not only removes the peroxide, but also creates more oxygen for cofactor regeneration. Alcohol oxidases are perceived to have a limited substrate scope preventing their wider use in synthesis. Thus, we present the engineering of two alcohol oxidases for increased substrate scope, one for the selective oxidation of primary alcohols and one for secondary alcohol oxidation. Arthrobacter chlorophenolicus choline oxidase and Streptomyces hygroscopicus cholesterol oxidase were selected as the primary and secondary alcohol oxidase, respectively. Examination of crystal structures of homologous alcohol oxidases revealed positions in the active site and entrance channel to target for saturation mutagenesis. Libraries were screened using a high-throughput assay based on the detection of hydrogen peroxide using hexanol as the substrate for evolution of choline oxidase and cyclohexanol for evolution of cholesterol oxidase. Mutations from positive hits were combined and, in these cases, resulted in variants with further improvements in both kcat and conversion to the carbonyl in biotransformations.
