Rok obhajoby práce: 2017 ABSTRAKT: Tato studie se zabývá klasifikací rodu Agrilus (Coleoptera: Buprestidae), nejpočetnější, značně morfologicky uniformní skupiny hmyzu, která obsahuje více než 3000 druhů. Mnohé z těchto druhů jsou významnými škůdci, kteří se za příhodných podmínek stávají v některých oblastech invazivními. Značná morfologická podobnost jednotlivých druhů znesnadňuje jejich jednoznačnou identifikaci, která je pro rychlé zachycení výskytu neznámého škůdce nezbytná. Určit neznámý druh rodu Agrilus, zvláště pokud je nepůvodní, je bez rozsáhlé srovnávací sbírky téměř nemožné a specialistů zabývajících se rodem Agrilus je velmi málo. Tato práce nabízí možnost identifikace neznámých vzorků pomocí databáze molekulárních markerů zahrnující téměř 100 druhů rodu Agrilus, jež je dostupná v databázi GenBank a použitelná pro molekulárně založenou identifikaci nových vzorků. V této studii byly použity dva přístupy: byly vytvořeny fylogenetické stromy na základě molekulárních markerů s použitím metody maximum likelihood a dále byly srovnány genetické párové vzdálenosti. Analýza 'barcode' fragmentu neumožňuje identifikace, pokud daný druh již není zachycen v databázi. V případě analýzy více fragmentů, je možné po nenalezení odpovídajícího jedince pomocí fylogenetické hypotézy o příbuznosti druhů nalézt nejpříbuznější sesterskou linii s jistým geografickým původem a odhadnout tak původ neznámého vzorku. V databázi jsou rovněž zachyceny nepůvodní druhy zavlečené do Severní Ameriky z Evropy a Asie. Ladislav Bocák Ph. D. Year of defense: 2017 ABSTRACT: This work tests the methods for DNA-based identification of the largest, morphologically uniform insect lineage. The genus Agrilus (Coleoptera: Buprestidae), which includes more than 3000 morphologically identified species, contains economically important pests, which become invasive in some areas with suitable conditions. The unambiguous identification, which is necessery for quick interception and disposal, is complicated by high morphological similarity of individual species. The identification of the unknown species of Agrilus is impossible without an extensive reference collection and expertize knowledge and additionally, there are few trained entomologists available. This study demonstrates the effectiveness of various methods for identification of unknown samples. I used a newly produced molecular marker database, which represents about 100 species of the genus Agrilus from the Northern Hemisphere and the data available in GenBank database. The tree-based and distance based methods were used for identification of species limits and estimation of a cryptic genetic diversity. In this study, phylogenetic hypotheses were also constructed based using molecular markers and maximum likelihood optimality criterion. If the closely similar sequence is absent in the database, to data can identify the most closely related sister species of population, inable estimation of the geographic origin based on the phylogenetic hypothesis. Jejich rozšíření je kosmopolitní a největší diversita je v oblastech s vysokou rozmanitostí potenciálních hostitelských rostlin, především v subtropických částech holoarktické oblasti (Bellamy 2010). Všechny druhy této skupiny jsou fytofágní a jejich diversita je pravděpodobně výsledkem četných přesunů z jedné hostitelské rostliny na druhou. Příkladem druhu, jehož diversifikace je ovlivněna přesuny mezi hostitelskými rostlinami je polyfágní středoevropský druh Agrilus viridis (Bernhard et al. 2005). Analýza molekulárních dat tohoto druhu získaného z odlišných živných rostlin naznačuje, že by se ve skutečnosti mohlo jednat o více specializovaných populací, poddruhů nebo druhů, a nikoliv pouze o jediný takto široce polyfágní druh (Bernhard et al. 2005, Pentinsaari et al. 2014). Larvy rodu Agrilus se živí rostlinnými pletivy a daří se jim především pod kůrou rozmanitých druhů stromů a keřů (Jendek 1994). Větší počty jedinců se vyvíjejí na jednotlivých rostlinách zejména v případě, že jsou stresovány suchem (Dunn

These insects can cause impressive economic and ecological damage in the invaded environment. Thus, several countries use traps baited with pheromones and plant volatiles at ports of entry and surrounding natural areas to intercept incoming exotic species soon after their arrival and thereby reduce the likelihood of their establishment. In this study, we investigated the performance of eight trap colors in attracting jewel beetles and bark and ambrosia beetles to test if the trap colors currently used in survey programs worldwide are the most efficient for trapping these potential forest pests. In addition, we tested whether trap colors can be exploited to minimize inadvertent removal of their natural enemies.

    ISRAEL JOURNAL OF ENTOMOLOGY, Vol. 34, 2004, pp. 109-152 New records of Buprestidae (Coleoptera) from Israel with description of a new species MARK VOLKOVITSH Zoological Institute, Russian Academy of Sciences, 199034 St. Petersburg, Russia ABSTRACT In two expeditions to Israel (in 1994 and 1996), 123 species of Buprestidae (Coleoptera), including one with two subspecies, were collected. Data col- lected, general distribution, and known larval host plants are given for each species. Fifteen Buprestidae species, and new hosts for four species, are recorded from Israel for the first time, including three new host associations for species that already had other known hosts. Xantheremia (Xantheremia) freidbergi is described as a new species. Acmaeodera (Palaeotethya) macchabaea Abeille de Perrin is resurrected from synonymy with A. rubro- maculata sensu Volkovitsh 1986, in part, and two new synonyms, A. pharsaea Obenbberger and A. houskai Obenberger, are established for this species. Acmaeoderella sefrensis Pic is a new status for Acmaeodera rufo- marginata var. sefrensis Pic, and two new synonyms are established for it: A. rufomarginata var. stramineipennis Chobaut and A. virgulata var. lucasi Théry. Acmaeodera miribella Obenberger is a new subspecies of Acmaeo- derella elegans (Harold) n. comb., and Acmaeodera eucera Obenberger is its synonym. Acmaeoderella adspersula arabica Cobos is elevated to species status. Lectotypes are designated for A. rufomarginata var. sefrensis, A. rufomarginata var. stramineipennis, and A. virgulata var. lucasi. INTRODUCTION The study of buprestid fauna of Israel has been relatively poor. Bodenheimer (1937) listed 84 species. There are only a few publications on Israeli buprestids in the 20th century that either summarize data collected (Obenberger, 1946; Niehuis, 1996) or are devoted to buprestid fauna of certain localities (Chikatunov et al., 1999; Volkovitsh et al., 2000).

  • Neufund Von Coraebus Elatus Und Wiederfund Von Trachys Problematica in Sachsen (Coleoptera: Buprestidae)
    30 Kwast: Buprestidae Sächsische Entomologische Zeitschrift 7 (2012/2013) 31 Neufund von Coraebus elatus und Wiederfund von Trachys am 22.07.2009) wurde folglich für Sachsen gestrichen, da zu diesem Zeitpunkt keine problematica in Sachsen (Coleoptera: Buprestidae) Literaturangaben bzw. Belegtiere aus Sachsen bekannt waren. Ende desselben Jahres konnte C. elatus jedoch tatsächlich aus der östlichen Oberlausitz nach- gewiesen werden. Eine kurze Notiz hierzu findet sich bereits in Hornig et al. (im Tom Kwast Druck). Der Nachweis erfolgte auf ungewöhnliche Weise. Das Einzeltier wurde Anfang November tot aus nassem Buchenmulm von einer Baumhöhle gesiebt Elisenstraße 34, 01307 Dresden; E-Mail: [email protected] (Richter mündl. Mitt.). Es handelt sich dabei um einen zufälligen Aufenthaltsort des Käfers bzw. dieser könnte im Spätsommer von austretendem Baumsaft angelockt Zusammenfassung. In this study, 297 beetle species from various taxonomic groups were screened with the use of molecular markers for the presence of infection by any of these endosymbionts. Wolbachia was found to be the most common “male-killer” among beetle hosts as it infected approx. 27% of species. Rickettsia, Spiroplasma and Cardinium were much less prevalent as they infected: 8%, 3% and 2%, respectively, of the studied beetle species. This is the first report of Cardinium presence in beetle hosts. Incidences of co-infection of two bacteria taxa were very rare and only two weevil species were found to be infected by three different bacteria. These findings suggest that endosymbiotic bacteria inhabit their host at various levels of prevalence and that particular taxa usually infect different hosts, suggesting some competition among “male-killers”.

In the second year, the differences in spider species composition and activity density diminished. The main conclusion of our study is that the large-scale intensive summer fire caused no profound changes in cursorial beetle and spider assemblages of this steppe plot. Mitigation of the fire effect is explained by the small plot area, its location at the edge of the fire site and the presence of adjacent undisturbed habitats with herbaceous vegetation.

In this study, we present a survey of ecological associations between herbivorous beetles and host plants from seriously threatened European steppic grasslands. We determined host plants for the majority (65%) of steppic leaf beetles (55 species) and weevils (59) known from central Europe using two barcodes (trnL and rbcL) and two sequencing strategies (Sanger for mono/ oligophagous species and Illumina for polyphagous taxa). To better understand the ecological associations between steppic beetles and their host plants, we tested the hypothesis that leaf beetles and weevils differ in food selection as a result of their phylogenetic relations (within genera and between families) and interactions with host plants. We found 224 links between the beetles and the plants. Beetles belonging to seven genera feed on the same or related plants. Their preferences were probably inherited from common ancestors and/or resulted from the host plant’s chemistry. Bee- tles from four genera feed on different plants, possibly reducing intrageneric competi- tion and possibly due to an adaptation to different plant chemical defences.
