Warranty 18 - 2 - ENGLISH 1 - INTRODUCTION Gilson’s PLATEMASTER is a 96-channel manual pipetting system designed for laboratory applications using microplates. It can transfer liquids with high throughput, accuracy, and precision. PLATEMASTER is ideal for high throughput applications as it will pipette 96 wells simultaneously and still be easy to use, based on the same working principle as a standard air-displacement pipette. PLATEMASTER is suitable for a wide range of applications: • Pipetting and providing master mixes for PCR/qPCR. • Simultaneous processing of nearly all reaction steps in microtiter plates (coating, starting, stopping, washing, etc.) for applications like ELISA. • Addition and removal of culture media, plate replication in cell culture. • Dropwise addition of solutions for crystallography. • Plate replication in expression tests. • Enhancement of existing automation: assay development and adaptation to 96-channel systems, back-up solution for maintenance and downtime. • Whenever workflows involve pipetting on a 96-well or 384-well scale. Gilson PLATEMASTER P20 - 3 - User’s Guide Before using the unit, the user must know how to set up and operate the PLATEMASTER. Therefore, please make sure you read Chapters 1 to 3, including the safety notes. These chapters also describe some of the methodology required for obtaining accurate and reproducible results. Chapter 1 - Introduction contains general information including the safety notes which should be read before installing and operating the device. Chapter 2 - Description contains a detailed description of the PLATEMASTER. Chapter 3 - Operation describes how to operate the PLATEMASTER. Chapter 4 - Appendices contains details on accessories and technical data (specifications). For further information visit www.gilson.com Read this section carefully before installing and operating PLATEMASTER. The instrument described in this User’s Guide is a manual pipetting system to be operated by qualified personnel in a laboratory or similar indoor environment. Cleaning, installation, dismantling, maintenance, adjustment and repair should be performed by appropriately trained aware of the hazards involved. An efficient ground connection is mandatory for the physical protection of the user. Adequate protection, such as ventilation, must be provided if dangerous liquids are used in analytical work. In case of incidental spillage, carefully clean up the spillage, consider the nature of the spilled liquid, and take all required safety measures, accordingly. - 4 - ENGLISH Safety PLATEMASTER is designed for research purposes only. If PLATEMASTER is used in a manner not specified by Gilson, the protection provided by the instrument may be impaired. Gilson will not be responsible for consequential damages. The use of the transportation safety lock is only dedicated for the in house transport. For all other transportation we recommend to disassemble the pipetting head from the base unit and use the transportation box. Temperature of use: 4 to 40°C - Recommended working temperature: 20 to 25 °C Only use accessories provided. Gilson SAS denies any responsibility for service and repairs carried out by the customer or third- party companies. Neither the equipment nor the filter tips applied can be autoclaved. Gilson does not take liability for any damage resulting from the use of PLATEMASTER. Follow the instructions of this User’s Guide to ensure durability and top performance of your PLATEMASTER. Keep this manual handy for future reference, in accordance with Good Laboratory Practices. Pipetting aggressive liquids may damage PLATEMASTER parts in contact with the solution. In case of contact, immediately clean up aggressive liquids. Pipetting of extremely viscous or highly evaporating liquids is at your own risk. The same applies to aggressive and corrosive reagents. Avoid aspiration of any liquid into the tip-holders. Avoid handling under the pipetting head. It is highly recommended to keep the packaging. If the unit has to be sent back to the factory, the original packaging must be used to return equipment under warranty. Report any loss or damage immediately. 2 - DESCRIPTION Introduction The pipetting head moves both vertically and horizontally. Vertical movements are performed for fitting tips and pipetting liquid, horizontal moves are performed for transferring liquid. This movement is possible only when the head is located completely at the top. Two drawers can hold up to three different microplates and can be used at the same time. - 6 - ENGLISH PLATEMASTER - General view The figure below shows a general view of a PLATEMASTER. Insert the pipetting head has follow and secure the head by screwing the first screw down. Unlock the pipetting head, returning it to the upper position. Secure the head by fastening the screw located below the head. - 8 - ENGLISH 4 - OPERATION Unlocking the pipetting head Whether placed on the lab bench, under a fume hood, in a cold room, or used in the field, the PLATEMASTER is completely portable. The PLATEMASTER pipetting head includes a locking device to lock the head when the instrument being moved. After each use of the PLATEMASTER, remember to lock the pipetting head to prepare for transport. To unlock the pipetting head, pull the black part on the right side of the PLATEMASTER and turn it in your direction. Once the pipetting head is released, check proper movement of the device. To lock the pipetting head, pull the black part and turn it in the opposite direction, make sure that the pipetting head cannot move anymore. PLATEMASTER is now securely locked. Never move the device without locking the pipetting head. Damages caused by movement without locking the device will not be covered by the warranty. Volume setting Do not adjust the volume without tightening both parts of the pipetting handle . The volume of liquid to be aspirated is set using the volume adjustment wheel . The pipetting handle being tightened, the volume is set by turning the button slowly to reach the required setting. Never try to force the volume outside of the volume range permitted by the device. - 9 - Tip fitting For optimum performance, use of PIPETMAN DIAMOND Tips with PLATEMASTER is strongly recommended. 5 These tips have the Gilson logo engraved on their 3 collar, ensuring that you have a genuine Gilson product. Plastic tips are for a single use – they must not be cleaned for reuse. 1 To fit PIPETMAN DIAMOND Tips on PLATEMASTER: 4 • Unlock the pipetting head and place it in the upper position. Place the pipetting head to the left side. 2 • Pull the right drawer and place a new rack of tips on the upper location. The four locations have been specially designed for PIPETMAN DIAMOND Tips TIPACK. The rack will perfectly fit into all the locations and will be stable due to the slots. • Move the pipetting head by using the pipetting handle to the right and move it down far enough so that a contact is made to the tips in the rack. • Press the tip fitting handle with moderate pressure down. The correct grabbing of the tips is indicated by the complete unfolding of the tip-ejector . • Move the pipetting head up and the right drawer back. Check the tip-ejector when fitting tips. If the tips have shifted this implies that they have not been fitted correctly. Pipetting The device is now ready to pipetting liquid. Place a vessel containing the solution to be pipetted in the front location of the right drawer. The solution should be under the pipetting head. Liquids (e.g. protein-containing solutions and organic solvents) can leave a film of liquid on the inside wall of the tip ; pre-rinse the tip to minimize any errors related to this phenomenon. Pre-rinsing consists of aspirating the first volume of liquid and then dispensing it back into the same vessel (or to waste). Subsequent volumes pipetted will have levels of accuracy and precision within specifications. As recommended by ISO 8655 standard, pre-rinse to stabilize dead volume inside the pipetting head. - 10 - ENGLISH Aspiration • Smoothly tighten the pipetting handle to the first stop (this corresponds to the set volume of liquid).

    Increasing the Reproducibility of Serial Dilutions Michael Beier* Abstract Pipetting serial dilutions reproducibly can be difficult because it is a time consuming and consequently error prone methodology, especially in 384 well plates. Based upon the assumption that your pipettes are properly maintained and calibrated, the human influence has the largest impact on pipetting results. Observing good pipetting practice helps immensely to improve reproducibility and accuracy. In this article we focus on a selection of good pipetting techniques, which if maintained offer the best chance of increasing and maintaining the reproducibility of your serial dilutions. Mixing is a key component of dilution protocols and in this article we additionally discuss different mixing parameters to help you achieve a homogenous sample. To maximize consistency of dilution assays, automating the process offers many benefits. For ensuring highly reproducible serial dilutions we outline what features an automated pipetting system should have and introduce the VIAFLO ASSIST – a product purpose designed for this task. Introduction to good pipetting practice Good pipetting practice describes guidelines covering the basic techniques to ensure that the pipettes are used in an optimal and reproducible manner. Before preparing your assay, you should perform a pre-wet of your pipette tips. Do this by aspirating and dispensing the full volume 2-3 times. This procedure not only equilibrates temperature differences between liquid and tip but also humidifies the dead air space inside the pipette and tip. Neglecting to perform a pre-wet can result in a smaller delivery volume in the first few dispenses, thereby introducing errors into your subsequent dilution steps.

Abstract High-throughput screening (HTS) can efficiently assay multiple discrete biological reactions using multi-well microplates. The choice of microplate is a crucial yet often overlooked technical decision in HTS and quantitative biology. This chapter reviews key criteria for microplate selection, including: well number, well volume and shape, microplate color, surface treatments/coatings, and considerations for specialized applications such as high-content screening. This chapter then provides important technical advice for microplate handling including mixing, dispensing, incubation, and centrifugation. Special topics are discussed, including microplate surface properties, plate washing, well-to-well contamination, microplate positional effects, well-to-well and inter-lot variability, and troubleshooting. This information and best practices derived from academic, government, and industry screening centers, as well as microplate vendors, should accelerate assay development and enhance assay quality.
