Starting from January 2008, the Hellenic Plant Protection Journal is published semiannually, in January and July each year. HPPJ publishes scientifi c work on all aspects of plant health and plant protection referring to plant pathogens, pests, weeds, pesticides and relevant environmental and safety issues. In addition, the topics of the journal extend to aspects related to pests of public health in agricultural and urban areas.     The following supplement accompanies the article Isolates as models to study bacterial ecophysiology and biogeochemistry Åke Hagström*, Farooq Azam, Carlo Berg, Ulla Li Zweifel *Corresponding author: [email protected] Aquatic Microbial Ecology 80: 15–27 (2017) Supplementary Materials & Methods The bacteria characterized in this study were collected from sites at three different sea areas; the Northern Baltic Sea (63°30’N, 19°48’E), Northwest Mediterranean Sea (43°41'N, 7°19'E) and Southern California Bight (32°53'N, 117°15'W). Seawater was spread onto Zobell agar plates or marine agar plates (DIFCO) and incubated at in situ temperature. Colonies were picked and plate- purified before being frozen in liquid medium with 20% glycerol. The collection represents aerobic heterotrophic bacteria from pelagic waters. Bacteria were grown in media according to their physiological needs of salinity. Isolates from the Baltic Sea were grown on Zobell media (ZoBELL, 1941) (800 ml filtered seawater from the Baltic, 200 ml Milli-Q water, 5g Bacto-peptone, 1g Bacto-yeast extract). Isolates from the Mediterranean Sea and the Southern California Bight were grown on marine agar or marine broth (DIFCO laboratories). The optimal temperature for growth was determined by growing each isolate in 4ml of appropriate media at 5, 10, 15, 20, 25, 30, 35, 40, 45 and 50o C with gentle shaking. Growth was measured by an increase in absorbance at 550nm. Statistical analyses The influence of temperature, geographical origin and taxonomic affiliation on growth rates was assessed by a two-way analysis of variance (ANOVA) in R (http://www.r-project.org/) and the “car” package.

To cut down the cost of production and optimize the PHA/dried cell weight different concentrations (1, 2 and 5 %) of refined glycerol and a crude glycerol from biodiesel process, were used as carbon source in a batch fermentation process. Pseudomonas mediterranea 9.1 as well as Pseudomonas corrugata 388 and A1 reference strains, grew and synthesized a medium-chain-length poly(3-hy-droxyalkanoates) elastomer both on crude and refined glycerol carbon sources. The mcl-PHA produced by P. mediterranea 9.1 grown on refined glyc- erol was apparently different as a result of a different metabolic pathway. Orco) from commercial greenhouses near La Plata, Argentina (35 ºS 57 ºW) showed abundant adventitious root production, apical chlorosis of leaves and a brown discoloration of the stem pith (Fig. 1). These symptoms were similar to those reported by López et al. (1994) and Catara et al. (2002) on tomatoes affected by Pseudomonas corrugata or Pseudomonas mediterranea. Bacteria consistently isolated from stem lesions formed cream-coloured, glistening, convex colonies on sucrose peptone agar (SPA) and were non-fluorescent on King’s medium B (KMB). Four isolates were selected for further study. All were aerobic, Gram-negative rods with PHB inclusions. In LOPAT tests, all induced a hypersensitive response in tobacco plants, were oxidase positive, did not cause soft rot of potato tubers, and were negative for levan and arginine dihydrolase. Colonies developed at 28ºC and 37ºC but not at 41ºC. Additional characterisation was achieved by API 20 NE tests strips (Biomerieux®, Argentina). Field monitoring in the region of Bitola show very high infection of around 20-30%. Two types of symptoms where observed during the field mon- itoring. First symptoms were observed on the leaves of the infected plants in the form of wilting, prior to flowering stage. The most characteristic symptoms were observed, at the lower part of the stem in the form of a stem cancer. Big variable sclerotia in size and shape were observed inside the stem. The appear- ance of white mycelium on the infected lower parts of the plant was often observed during the wet weather. Other infected plants showed different symp- toms. The stem was longer and thinner than in uninfected plants, and the pit was very small, around 9 cm. Sclerotia observed inside the stem were not big- ger than 2.5 mm. In vitro investigations confirmed the presence of ascomycetes Sclerotinia sclerotiorum (Lib.) de Bary and Sclerotinia minor Jagger, for the first time in the Republic of Macedonia.

Abstract Host-associated bacteria can have both beneficial and detrimental effects on host health. While some of the molecular mechanisms that determine these outcomes are known, little is known about the evolutionary histories of pathogenic or mutualistic lifestyles. Using the model plant Arabidopsis, we found that closely related strains within the Pseudomonas fluorescens species complex promote plant growth and occasionally cause disease. To elucidate the genetic basis of the transition between commensalism and pathogenesis, we developed a computational pipeline and identified genomic islands that correlate with outcomes for plant health. One island containing genes for lipopeptide biosynthesis and quorum sensing is required for pathogenesis. Conservation of the quorum sensing machinery in this island allows pathogenic strains to eavesdrop on quorum signals in the environment and coordinate pathogenic behavior. We found that genomic loci associated with both pathogenic and commensal lifestyles were convergently gained and lost in multiple lineages through homologous recombination, possibly constituting an early step in the differentiation of pathogenic and commensal lifestyles.
