Characters of Fungi Some of the most important characters of fungi are as follows: 1. Occurrence 2. Thallus organization 3. Different forms of mycelium 4. Cell structure 5. Nutrition 6. Heterothallism and Homothallism 7. Reproduction 8. Classification of Fungi. 1. Occurrence: Fungi are cosmopolitan and occur in air, water soil and on plants and animals. They prefer to grow in warm and humid places. Hence, we keep food in the refrigerator to prevent bacterial and fungal infestation. 2. Thallus organization: Except some unicellular forms (e.g. yeasts, Synchytrium), the fungal body is a thallus called mycelium. The mycelium is an interwoven mass of thread-like hyphae (Sing, hypha). Hyphae may be septate (with cross wall) and aseptate (without cross wall). Some fungi are dimorphic that found as both unicellular and mycelial forms e.g. Candida albicans. 3. Different forms of mycelium: (a) Plectenchyma (fungal tissue): In a fungal mycelium, hyphae organized loosely or compactly woven to form a tissue called plectenchyma. It is two types: i. Prosenchyma or Prosoplectenchyma: In these fungal tissue hyphae are loosely interwoven lying more or less parallel to each other. ii. Pseudoparenchyma or paraplectenchyma: In these fungal tissue hyphae are compactly interwoven looking like a parenchyma in cross-section. (b) Sclerotia (Gr. Skleros=haid): These are hard dormant bodies consist of compact hyphae protected by external thickened hyphae. Each Sclerotium germinates into a mycelium, on return of favourable condition, e.g., Penicillium. (c) Rhizomorphs: They are root-like compactly interwoven hyphae with distinct growing tip. They help in absorption and perennation (to tide over the unfavourable periods), e.g., Armillaria mellea. -

Abstract: The present study introduces a novel biotechnology for cost effective and economically feasible production of natural fungal rennin in Egypt to fulfill the local requirements of cheese industry instead of the calf rennet that is highly expensive and imported from abroad. In this study, Rhizomucor miehei NRRL 2034 exhibited the highest enzyme productivity under solid state fermentation. Among twelve industrial by-products used as nutrient substrates for Rhizomucor miehei growth under solid state fermentation, wheat bran yielded the highest milk clotting activity (33350 SU/g fermented culture). The optimum conditions for milk clotting enzyme production were moisture content of 50%, incubation temperature 40°C, incubation period 3 days and inoculums size of 1.8 × 108 CFU/g wheat bran. Addition of whey powder as a nitrogen source at 2% increased the enzyme productivity about 42% compared to the control medium. Wheat bran particle size more than 300 µm was the most suitable size for the highest production of the enzyme. The maximum milk clotting activity was achieved by using mineral solution as a moistening agent for wheat bran. Large scale production of the enzyme in tray showed the maximum activity when 125 grams wheat bran were distributed in an aluminum foil trays ( 20×25 ×5 cm 3) under the optimum cultural conditions. -

Accepted December 26, 2008) SUMMARY: Mucor spp. are rarely pathogenic in healthy adults, but can cause fatal infections in patients with immuosuppression and diabetes mellitus. Documented mucor fungemia is a very rare condition in the literature. We described a fungemia and cutaneous mucormycosis case due to Mucor circinelloides in an 83-year-old woman with diabetes mellitus who developed acute left frontoparietal infarctus while hospitalized in a neuro- logical intensive care unit. The diagnosis was made based on the growth of fungi in the blood, skin biopsy cultures, and a histopathologic examination of the skin biopsy. The isolates were identified as M. circinelloides by molecular methods. This case is important in that it shows a case of cutaneous mucormycosis which developed after fungemia and provides a contribution to the literature regarding Mucor fungemia. Culture-based methods for clinical samples are often negative, difficult and time-consuming and mainly identify isolates to the genus level, and sometimes only as Mucorales. Therefore, applying fast and accurate diagnosis methods such as molecular approaches seems necessary. This study aims at isolating Mucorales for determination of Rhizopus genus between the isolates using molecular methods. Methods: In this descriptive observational study, a total of 500 samples were collected from air and different surfaces and inoculated on Sabouraud Dextrose Agar supplemented with chloramphenicol. Then, the fungi belonging to Mucorales were identified and their pure culture was provided. DNA extraction was done using extraction kit and the chloroform method. After amplification, the samples belonging to Mucorales were identified by observing 830 bp bands. For enzymatic digestion, enzyme BmgB1 was applied for identification of Rhizopus species by formation of 593 and 235 bp segments. Results: One hundred pure colonies belonging to Mucorales were identified using molecular methods and after enzymatic digestion, 21 isolates were determined as Rhizopus species. -

Plant Pathology & Quarantine 8(2), 116–121, Doi 10.5943/ppq/8/2/2 Abstract During spring 2017, red delicious and Granny Smith apples with soft rot symptoms were collected from commercial markets in Saudi Arabia. The causal agent was isolated from infected fruits and its pathogenicity was confirmed by inoculation assays. To confirm the identity, total genomic DNA was extracted and multi-locus sequence data targeting two gene markers (ITS, ACT) was sequenced. Phylogenetic analysis confirmed the presence of two distinct clades; Saudi isolate was placed within a clade comprising Rhizopus oryzae and R. delemar reference isolates. Based on morphological characteristics, molecular identification and pathogenicity test, the fungus was identified as Rhizopus oryzae. This is the first report of Rhizopus soft rot caused by R. oryzae from stored apple fruits in Saudi Arabia. ABSTRACT BACKGROUND Fungi are ubiquitous soil saprophytes often involved in various human ailments. Fungal diseases are emerging worldwide. However, the mycological health risks associated with dumping yard is not much investigated, especially from developing countries where such sites are very common. Aims and Objectives- The major objective of the study was to analyse the possible mycological threat posed by the municipal dumping yard. The study also aims to assess the health status of interacting community in relation to the dumping yard. MATERIALS AND METHODS A descriptive study was conducted from April to June 2015 in Kottayam Municipal dumping yard at Vadavathoor. Two set of soil samples, a total of 50 from the dumping yard were collected, before and after burning of the dumping yard. One set of samples from leachate and neighbouring open wells were collected after burning of the dumping yard. Samples were collected in sterile containers and transported immediately to the laboratory. The samples were cultured on to suitable fungal culture medium and the growths of the fungus were identified by the microscopic and macroscopic features. RESULTS The isolated fungal pathogens from the soil shows that 49% isolates belonged to the Aspergillus genus; and the remaining 51% was almost equally shared by four other different species Geotrichum, Humicola, Microsporum, Rhizopus. -

Historically, this was done by morphologic characteristics and other phenotypic traits. However, with the advent of the molecular age in mycology, phylogenetic analysis based on DNA sequences has replaced these classic means for grouping related species. This, along with the abandonment of the dual nomenclature system, has led to a marked increase in the number of new species and reclassification of known species. Although these evaluations and changes are necessary to move the field forward, there is concern among medical mycologists that the rapidity by which fungal nomenclature is changing could cause confusion in the clinical literature. Thus, there is a proposal to allow medical mycologists to adopt changes in taxonomy and nomenclature at a slower pace. In this review, changes in the taxonomy and nomenclature of medically relevant fungi will be discussed along with the impact this may have on clinicians and patient care.
