Etiologia da mancha ocular na cultura do arroz causada por Bipolaris gigantea e alterações metabólicas desencadeadas nas plantas. 2017. 88f. Tese (Doutorado) – Programa de Pós-Graduação em Fitossanidade. Universidade Federal de Pelotas. Pelotas, RS, Brasil. A Região Sul do Brasil apresenta-se em posição de destaque no cenário orizícola brasileiro, com grande potencial de crescimento. A ocorrência de doenças está entre os fatores que podem restringir a expressão do seu potencial produtivo, reduzindo a área fotossintética, acarretando perdas na produção de grãos e queda na produtividade. Manchas foliares, relacionadas à doença conhecida como mancha ocular, venham sendo observadas desde a safra 2006/07 no estado do Rio Grande do Sul (RS). O objetivo do presente trabalho foi identificar e caracterizar a etiologia do patógeno causador da doença, bem como, informações referentes aos testes de patogenicidade e avaliar os componentes de resistência, variáveis bioquímicas de mecanismos de defesa e respostas citológicas envolvidas no processo de infecção. Para isso o patógeno foi isolado de plantas sintomáticas e analisado quanto as suas estruturas reprodutivas e colônias, comparando-as com a literatura padrão. A extração de DNA, amplificação por PCR e sequenciamento e análise de regiões conservadas do genoma, consistiram nas etapas para obter a descrição molecular do isolado. Componentes de resistência, atividade enzimática, peroxidação lipídica e quantificação de lesões foram analisados em diferentes períodos de inoculação e estádios fenológicos, para as cultivares BRS Querência e Puitá Inta CL. O presente estudo conclui, satisfatoriamente, que o agente causal da manha ocular do arroz é o fungo Bipolaris gigantea. Ainda, independente do estádio, a cv BRS Querência mostrou maior suscetibilidade à doença, causando danos celulares, a nível de membrana, devido a ação desencadeada pelo fungo, através do acúmulo de espécies reativas de oxigênio. Puitá Inta CL apresentou resposta de defesa através da ação de enzimas antioxidantes. Os resultados esclareceram à correta taxonomia do agente causal da mancha ocular da cultura do arroz e demostraram as primeiras evidências das respostas de defesa de cultivares ao patógeno. Universidade Federal de Pelotas. Pelotas, RS, Brasil. The lands in southern Brazil are in a highlighted position of the rice cropping area, with great potential to increase. The occurrence of diseases is among the factors that can restrict its potential, reducing the photosynthetic area, causing losses on the grains’ production and reduction of yield. Foliar leaf spots related to the eyespot disease are being observed since 2006/07’s rice harvest, in the state of Rio Grande do Sul (RS).The objective of the study is identify and characterize the etiology of the pathogen causing the disease, as well, acquire information regarding the pathogenicity tests and evaluate the resistance components, biochemical variables of defense mechanisms and cytological responses involved in the infection process. In order to accomplish that, the pathogen was isolated from symptomatic plants and its reproductive structures and its colonies were analyzed and compared to the literature. The DNA extraction, amplification by PCR and sequencing and analysis of genome consisted on the steps to obtain the molecular description of the isolate. Components of resistance, enzymatic activity, lipid peroxidation and lesion quantification were analyzed in different periods of inoculation and phenological stages, for cultivars BRS Querência and Puitá Inta CL. The present study concludes that the agent which is causing the eyespot on rice is the Bipolaris gigantea fungus. Moreover, independently of stage the cv BRS Querência showed major susceptibility to the disease, causing cellular damages at membrane level, due to the action triggered by the fungus, through the accumulation of reactive oxygen species. Puitá Inta CL showed defense response by action of antioxidant enzymes. The results showed the correct taxonomy of the causal agent of zonate eyespot on rice crop culture and demonstrate the first evidence of defense responses of cultivars pathogen. McMullin ABSTRACT A comprehensive investigation of the secondary metabolites produced by Chaetomium globosum, Wallemia sebi, Penicillium corylophilum and four Trichoderma species obtained from Canadian buildings is presented. Atopic and non-atopic individuals occupying damp, moldy buildings are at increased risk of both allergic and non-allergic adverse health effects. There is now strong toxicological evidence showing that secondary metabolites, including mold specific glucan, present on spores and mycelial fragments are in part responsible for these effects. At the low doses that could be experienced by the human lung indoors, metabolites from fungi have been demonstrated to alter the expression of genes involved with asthma in vivo and in vitro. These genetic alterations are accompanied by histological disruptions and inflammatory responses. The primary focus of this study was to identify and isolate the dominant toxins produced by the mentioned fungi obtained from Canadian buildings. Isolates were grown in liquid culture and screened for metabolite production. Metabolites were purified by various chromatographic methods and their structures were unambiguously determined by mass spectrometry and detailed analysis of spectroscopic data. In this work, C. globosum primarily produced chaetoglobosin A, C and F, chaetomugilin D and chaetoviridin A.

Thus, it is crucial to understand the large-scale processes that shape plant pathogen communities. The recent drop in DNA sequencing costs offers, for the first time, the opportunity to study multiple plant pathogens simultaneously in their naturally occurring environment effectively at large scale. In this thesis, my aims were (1) to employ next-generation sequencing (NGS) based metabarcoding for the detection and identification of plant pathogens at the ecosystem scale in New Zealand, (2) to characterise plant pathogen communities, and (3) to determine the environmental drivers of these communities. First, I investigated the suitability of NGS for the detection, identification and quantification of plant pathogens using rust fungi as a model system.

    An Inventory of Fungal Diversity in Ohio Research Thesis Presented in partial fulfillment of the requirements for graduation with research distinction in the undergraduate colleges of The Ohio State University by Django Grootmyers The Ohio State University April 2021 1 ABSTRACT Fungi are a large and diverse group of eukaryotic organisms that play important roles in nutrient cycling in ecosystems worldwide. Fungi are poorly documented compared to plants in Ohio despite 197 years of collecting activity, and an attempt to compile all the species of fungi known from Ohio has not been completed since 1894. This paper compiles the species of fungi currently known from Ohio based on vouchered fungal collections available in digitized form at the Mycology Collections Portal (MyCoPortal) and other online collections databases and new collections by the author. All groups of fungi are treated, including lichens and microfungi. 69,795 total records of Ohio fungi were processed, resulting in a list of 4,865 total species-level taxa. 250 of these taxa are newly reported from Ohio in this work. 229 of the taxa known from Ohio are species that were originally described from Ohio. A number of potentially novel fungal species were discovered over the course of this study and will be described in future publications. The insights gained from this work will be useful in facilitating future research on Ohio fungi, developing more comprehensive and modern guides to Ohio fungi, and beginning to investigate the possibility of fungal conservation in Ohio. Abstract Common buckthorn (Rhamnus cathartica) is a competitive Eurasian woody shrub currently invading North America. Buckthorn thickets reduce native diversity and may reduce mycorrhizal diversity through the release of allelochemicals. Two aspects of buckthorn’s invasional biology are explored: 1) identifying fungi associating with buckthorn, and 2) determining buckthorn’s allelochemical impacts on arbuscular mycorrhizae in forest soils and an open-greenhouse experiment. Twenty-three fungi were found growing on buckthorn, including Armillaria mellea s.l., Hypoxylon fuscum, H. perforatum, Nectria cinnabarina, and Cylindrobasidium evolvens. Data from invaded and uninvaded sugar maple (Acer saccharum) soils revealed that arbuscular mycorrhizal fungi (AMF) diversity fluctuated as a function of season or potting disturbance, but the presence of buckthorn had little effect on AMF development in maple roots.
