(Head of Department) Signature Date ii ABSTRACT The purposes of the study were to identify Colletotrichum species, detect their variability, to investigate their cross infectivity potential and determine farmers' awareness about the diseases that are associated with mango and citrus. Culture characteristics, size and shape of conidia and mycelial growth rates were used to characterize the isolates into different morphological groups. Those grouped isolates were further studied for their genetic similarity and differences, using multigene loci primers and restriction enzymes. ITS region, β- tub gene and GPDH primers were also used to characterize the isolates. Farmers were aware of mango anthracnose but did not know any such disease on citrus. Colletotrichum species isolated from mango and citrus had overlap in their cultural characteristics. All of the isolates were assigned into ten morphogroups. Seven of the morphogroups were characterized by their cylindrical conidia and three of the morphogroups (G1, G3 and G6) had curved conidia. Apart from the conidial shape, most of the morphogroups had unique characters to stand as a group on their own. However, G2, G5 and G9 showed overlap in many of their characters. The results from molecular analysis were different, depending on the gene/region targeted. The ITS region amplification showed high genetic similarity among the ten morphogroups, irrespective of their conidial or other cultural characteristic variations. Further investigation on the same ITS region, using Colletotrichum species-specific primer, CgInt, confirmed the similarity for six of the morphogroups such as (G1, G2, G4, G5, G8 and G9) but the other four morphogroups were not picked. The overall results of the ITS region were very helpful in understanding the close relationship of the different iii morphogroups. Further, analysis of the same group of isolates, using β-tubulin primer, clearly showed existence of genetic variability in between the groups. The variability expressed on β-tub gene analysis was in line with the characterization made, based on conidial shape. It gave two distinct groups; one group having cylindrical conidia and the other having curved conidia. This result is an indicator that conidial shape is governed by a genetic factor. The third gene, GPDH analysis gave high genetic variability results. It amplified nine distinct groups from the ten morphogroups studied. Eight of them were in line with eight of the morphogroups. Therefore, GPDH results were more detailed than the other studied regions of Colletotrichum species in showing genetic variability. However, the combined results of the morphological characterization and the multigene analyses played their own role for a better understanding of species variability. The pathogenicity tests on mango and citrus fruits confirmed most of the morphogroups as pathogenic. However, all the curved morphogroups isolated from diseased mango tested negative on mango. The cross-infectivity potential of citrus and mango isolates of Colletotrichum were confirmed. The cross tests on papaya also showed that most of the morphogroups were pathogenic. From the study, it is understood that critical investigation of Colletotrichum spp., using the traditional techniques of morphological characterization, can assist in determining the genetic variability among isolates but it is tedious, time consuming and wrong conclusions could be drawn if it is first time study. Once the organisms are sequenced and named, the conventional investigation could play a great role to detect the variability. Among the varieties Ranjitha N tested, none of them showed immune and resistant reaction to the disease under shade house condition.     PEST AND DISEASES IN MANGO (MANGIFERA INDICA L.) J. González-Fernández, J.I. Hormaza IHSM la Mayora CSIC-UMA, 29750 Algarrobo, Malaga, Spain EXECUTIVE SUMMARY In this work, we review the most important pests and diseases that affect mango production worldwide as well as the main measures implemented to control them. Pests and diseases are the main factors that can impact sustainable mango fruit production in the tropics and subtropics worldwide. Commercial cultivation of mango, characterized by expansion to new areas, changing crop management, replacement of varieties and increased chemical interventions, has altered significantly the pest and disease community structure in this crop in the different mango producing regions. In addition, climate change is inducing the emergence of new pests and, whereas globalization and trade liberalization have created wide opportunities for mango commercialization growth, at the same time, this can result in faster dispersion of pests and diseases among different mango growing areas if proper sanitary measures are not implemented. This review covers different topics related to pests and diseases in mango. First, a thorough description of the main pests and diseases that affect mango is provided. Second, the different approaches used in different mango producing countries for chemical and biological control are described. Third, recommendations for appropriate mango management techiques that include integrated pest and disease management, reduction in the use of chemicals and the implementation of a good monitoring and surveillance system to help control the main pests and diseases, are also discussed. Finally, the current knowledge on agrohomeopathy and Korean Natural Farming is analyzed and recommendations on future lines of research to optimize mango pest and disease control are discussed.

    Model Profile for 1.0 ha Mango Cultivation 1. Introduction Mango (Mangifera indica) is the leading fruit crop of India and considered to be the king of fruits. Besides delicious taste, excellent flavour and attractive fragrance, it is rich in vitamin A&C. The tree is hardy in nature, can be grown in a variety of soil and requires comparatively low maintenance costs. Mango fruit is utilised at all stages of its development both in its immature and mature state. Raw fruits are used for making chutney, pickles and juices. The ripe fruits besides being used for desert are also utilised for preparing several products like squashes, syrups, nectars, jams and jellies. The mango kernel also contains 8-10 percent good quality fat which can be used for soap and also as a substitute for cocoa butter in confectionery. 2. Scope for Mango Cultivation and its National Importance Mango occupies about 36% of the total area under fruits (2010-11) comprising of 22.97 lakh hectares, with a total production of 151.88 lakh tonnes. Uttar Pradesh and Andhra Pradesh are having the largest area under mango each with around 23% of the total area followed by Karnataka, Bihar, Gujarat and Tamilnadu. Fresh mangoes and mango pulp are the important items of agri-exports from India. India's main export destinations for mango are UAE, Bangladesh, UK, Saudi Arabia, Nepal, Kuwait, USA and other Middle East countries with a limited quantity being shipped to European market. Although, India is the largest mango producing country, accounting about 45% of world production, the export of fresh fruit is limited to Alphonso and Dashehari varieties.

Chambers Abstract Fresh mangoes are highly perishable, and therefore, they are often processed to extend shelf-life and facilitate exportation. Studying the transformation that mango cultivars undergo throughout processing can aid in selecting appropriate varieties for products. In the 1st part of this study, the flavor and texture properties of 4 mango cultivars available in the United States (U.S.) were analyzed. Highly trained descriptive panelists in the U.S. evaluated fresh, purée, and sorbet samples prepared from each cultivar. Purées were made by pulverizing mango flesh, passing it through a china cap, and heating it to 85 °C for 15 s. For the sorbets, purées were diluted with water (1:1), sucrose was added, and the bases were frozen in a batch ice cream freezer. Much of the texture variation among cultivars was lost after fresh samples were transformed into purées, whereas much of the flavor and texture variation among cultivars was lost once fresh mangoes and mango purées were transformed into sorbets. Compared to the other cultivars, Haden and Tommy Atkins underwent greater transformations in flavor throughout sorbet preparation, and processing reduced the intensities of some unpleasant flavors in these cultivars.

Mango (Mangifera indica L.) germplasm can be classifi ed by origin with the primary groups being cultivars selected from the centers of diversity for the species, India and Southeast Asia, and those selected in Florida and other tropical and subtropical locations. Accessions have also been classifi ed by horticultural type: cultivars that produce monoembryonic seed vs. cultivars that produce polyembryonic seed. In this study we used 25 microsatellite loci to estimate genetic diversity among 203 unique mangos (M. indica), two M. griffi thii Hook. f., and three M. odorata Griff. accessions maintained at the National Germplasm Repository and by Fairchild Tropical Botanic Garden in Miami, Fla. The 25 microsatellite loci had an average of 6.96 alleles per locus and an average polymorphism information content (PIC) value of 0.552 for the M. indica population. The total propagation error in the collection (i.e., plants that had been incorrectly labeled or grafted) was estimated to be 6.13%. When compared by origin, the Florida cultivars were more closely related to Indian than to Southeast Asian cultivars.
