The study of the mechanisms regulating the specific features of ES cells led to the discovery of the transcription factors (TFs) governing their unique transcriptome. Among those TFs, Nanog plays a central role in the gene regulatory network that supports ES cells self-renewal. However, the molecular mechanisms by which Nanog exerts its functions are not fully elucidated. We adapted an inducible CRISPR activation (CRISPRa) system in mouse ES cells and demonstrated its functionality to stimulate transcription at endogenous loci. We then accurately determined the list of Nanog responsive genes using gain (CRISPRa cell line) and loss of function experiments and total RNA sequencing. Moreover, the DNA binding profiles of distinct pluripotency TFs were assessed genome-wide by chromatin immunoprecipitation (ChIP) and sequencing upon Nanog depletion and revealed the importance of the latter in the regulation of the pluripotency network activity at thousands of loci. -

NOBEL LAUREATES WITH THEIR CONTRIBUTION IN BIOMEDICAL ENGINEERING Nobel Prizes and Biomedical Engineering In the year 1901 Wilhelm Conrad Röntgen received Nobel Prize in recognition of the extraordinary services he has rendered by the discovery of the remarkable rays subsequently named after him. Röntgen is considered the father of diagnostic radiology, the medical specialty which uses imaging to diagnose disease. He was the first scientist to observe and record X-rays, first finding them on November 8, 1895. Radiography was the first medical imaging technology. He had been fiddling with a set of cathode ray instruments and was surprised to find a flickering image cast by his instruments separated from them by some W. C. Röntgenn distance. He knew that the image he saw was not being cast by the cathode rays (now known as beams of electrons) as they could not penetrate air for any significant distance. After some considerable investigation, he named the new rays "X" to indicate they were unknown. In the year 1903 Niels Ryberg Finsen received Nobel Prize in recognition of his contribution to the treatment of diseases, especially lupus vulgaris, with concentrated light radiation, whereby he has opened a new avenue for medical science. In beautiful but simple experiments Finsen demonstrated that the most refractive rays (he suggested as the “chemical rays”) from the sun or from an electric arc may have a stimulating effect on the tissues. If the irradiation is too strong, however, it may give rise to tissue damage, but this may to some extent be prevented by pigmentation of the skin as in the negro or in those much exposed to Niels Ryberg Finsen the sun. -

This will surprise no one in the community of scientists, within which our reputation ranks among those of the very best life science research institutions worldwide. This fact was reflected once again in an annual survey by Thompson Scientific’s Essential Science Indicators, which reported in 2008 that CSHL is in the top 1% of institutions most cited in published research and that our faculty is ranked by peers among the top three in terms of its influence in shaping the fields of mole- cular biology and genetics. Much of this is due to the sound guidance of our research admin- istration, which con sists of a very effective team headed by Director of Research David L. Spector and includes Research Exec u tive Committee members Greg Han non, Scott Lowe, and Tony Zador, who receive outstanding administrative assistance from Sydney Gary and Walter Goldschmidts. The Laboratory’s achievements in 2008 are particularly noteworthy when considered in the context of the current funding environment. Against a backdrop of limited resources and a contraction in federal grant funding, scientists in all of the areas in which CSHL concen- trates—cancer, neuroscience, and plant biology—reported a steady stream of significant research results throughout the year. Although these are comprehensively described on our website, it is useful to briefly review a few among the many to illustrate the richness of our sci- entists’ cumulative achievements in 2008 and the continuing overall success of our research program. -
