Coccídios (Protozoa: Apicomplexa) em peixes da planície de inundação do Rio Curiaú, Estado do Amapá: Prevalência e Caracterização Molecular. Macapá, 2018. Dissertação (Mestre em Biodiversidade Tropical) – Programa de Pós-graduação em Biodiversidade Tropical – Pró-Reitoria de Pesquisa e Pós-Graduação - Universidade Federal do Amapá. Os peixes são vulneráveis aos coccídios, sendo registrados diferentes estágios desses parasitos em todas as classes de vertebrados. O objetivo deste estudo foi investigar coccídios do gênero Calyptospora em espécies de peixes da planície de inundação do Rio Curiaú, estado do Amapá, Brasil. Foram coletados 130 peixes, das espécies Astyanax bimaculatus, Metynnis lippincottianu, Hoplias malabaricus, Hoplerythrinus unitaeniatus; Astronotus ocellatus, Cichla ocellaris, Crenicichla lepidota, Geophagus proximus, Heros efasciatus, Mesonauta festivus, Pterophyllum scalare e Satanoperca jurupari. Os peixes foram eutanasiados, pesados (g), medidos (cm), e dissecados para coleta de fragmentos de fígado, coração e vesícula e examinados pelo método de exame a fresco entre lâmina e lamínula, em microscópio de luz (ML), para verificar a presença de oocistos de Calyptospora spp. Os fragmentos de fígado de G. proximus e H. malabaricus foram armazenados em álcool etílico 80%. Posteriormente, o DNA total de cada amostra foi extraído e submetido a reação em cadeia da polimerase, mediante o uso de primers designados para detecção de um seguimento do gene SSU rDNA de Calyptospora spp. Os produtos das amplificações foram sequenciados e as seqüências obtidas foram comparadas com outras disponíveis no Genbank quanto à similaridade. Foi registrado a presença de oocistos de Calyptospora em 55% dos 130 animais examinados, ou seja, 10 das 12 espécies estudadas. Não foram observadas infecções em A. bimaculatus e M. lippincottianus, peixes de hábito herbívoro. As taxas de infecção variaram entre os órgãos parasitados, onde em todos os hospedeiros positivos foram encontrados oocistos no fígado, seguido por 30% na vesícula biliar e 9% no coração. As sequencias obtidas na biologia molecular apresentaram 99% de similaridade com Calyptospora serralsami descrita de piranha Serrasalmus rhombeus no Brazil. No presente estudo foi registrado pela primeira vez a ocorrência de Calyptospora sp. em A. Ocellatus, G. proximus, H. efasciatus, H. unitaeniatus, H. malabaricus, M. festivus P. Scalare e S. jurupari. Foi determinado a ocorrência de C. serrasalmi na planície e em dois novos hospedeiros. Fish are vulnerable to coccidia, with different stages of these parasites being recorded in all vertebrate classes. The objective of this study was to investigate coccidia of the genus Calyptospora in fish species from the Curiaú River floodplain, Amapá state, Brazil. A total of 130 fish belonging to species Astyanax bimaculatus, Metynnis lippincottianu, Hoplias malabaricus, Hoplerythrinus unitaeniatus; Astronotus ocellatus, Cichla ocellaris, Crenicichla lepidota, Geophagus proximus, Heros efasciatus, Mesonauta festivus, Pterophyllum scalare e Satanoperca jurupari. The fish were euthanized, weighed (g), measured (cm), and dissected for collection of fragments of liver, heart and vesicle and examined by fresh method between lamina and laminula under light microscopy (ML) to verify the presence of oocysts of Calyptospora spp. The liver fragments of G. proximus and H. malabaricus were stored in ethyl alcohol 80%. Subsequently, total DNA from each sample was extracted and subjected to polymerase chain reaction using primers designated to detect a follow-up of the SSU rDNA gene from Calyptospora spp. The products of the amplifications were sequenced and the sequences obtained were compared with others available from Genbank for similarity The presence of Calyptospora oocysts was recorded in 55% of the 130 animals examined, that is, 10 of the 12 species studied. No infections were observed in A. bimaculatus and M. lippincottianus, herbivorous habit fish. Infection rates varied among the parasitized organs, where in all positive hosts oocysts were found in the liver, followed by 30% in the gallbladder and 9% in the heart. The sequences obtained in molercular biology presented 99% similarity with Calyptospora serralsami described of piranha Serrasalmus rhombeus in Brazil. In the present study, the occurrence of Calyptospora sp. in A. Ocellatus, G. proximus, H. efasciatus, H. unitaeniatus, H. malabaricus, M. festivus P. Scalare and S. jurupari. The occurrence of C. serrasalmi in the plain and two new hosts was determined. Finally, new primers were created in this study and tested successfully for Calyptospora diagnosis. Hosts from the Tapajós River were infected by Posthodiplostomum sp. metacercarie, larvae of Contracaecum sp. and Pseudoproleptus sp., with predominance of Contracaecum sp. Hosts from Iara Lake were infected by Procamallanus spiculastriatus, Proteocephalus sp. and Acanthocephala gen. sp., with predominance of P. spiculastriatus. Parasites had an aggregated dispersion and there were differences in Shannon diversity index and the evenness between both host populations, because the parasite component community showed no similarity. The existence of variation in infracommunity and community of parasites for A. ocellatus from different localities indicates the presence of an uneven distribution in terms of species and density of parasites, as well as of intermediate hosts in the localities surveyed.

Studies of the molecular components of the visual transduction cascade, in particular visual pigments, offer an important window into the relationship between genetic variation and organismal fitness. In this thesis, I use molecular evolutionary models as well as protein modeling and experimental characterization to assess the role of variable evolutionary rates on visual protein function. In Chapter 2, I review recent work on the ecological and evolutionary forces giving rise to the impressive variety of adaptations found in visual pigments. In Chapter 3, I use interspecific vertebrate and mammalian datasets of two visual genes (RH1 or rhodopsin, and RPE65, a retinoid isomerase) to assess different methods for estimating evolutionary rate across proteins and the reliability of inferring evolutionary conservation at individual amino acid sites, with a particular emphasis on sites implicated in impaired protein function. ii In Chapters 4, and 5, I narrow my focus to devote particular attention to visual pigments in Neotropical cichlids, a highly diverse clade of fishes distributed across South and Central America.

Florida 33701-5095, USA ABSTRACT Two new species of coccidia, Epieimeria ocellata n sp. and Goussia floridana n. sp., were found in the intestine of red drum Sciaenops ocellatus (L.) (Sciaenidae) in Florida, USA. Merogony and gamogony stages of both species were 'epicellular' in the microvlllous region at epithelia1 cell apices. In E. ocellata, sporogony was intracellular, with endogenous sporulation. Fresh, mature oocysts were roughly spherical (9.6 pm long X 9.3 pm wide) and had no oocyst residuum. Sporocysts were ellipsoidal (6.9 pm long X 4.1 pm wide) and had a distinct Stieda body. Sporozoites were thick (5.6 pm long x 1.8 pm wide), were aligned side by side, and had flexed ends. In G. floridana, sporogony was extra- cellular, with exogenous sporulation. Fresh, mature oocysts were subspherical (19.9 um long X 15.9pm wide) and had no oocyst residuum. Sporocysts wereellipsoidal (12.6 pm long X 7.5 pm wide) and had an indistinct suture line. The sporocyst residuum consisted of 1 to 14 granules. Sporozoites were thick (11.0 pm long X 3.9 pm wide) and occupied most of the sporocyst. INTRODUCTION January and May 1992.     www.nature.com/scientificreports OPEN Species composition and invasion risks of alien ornamental freshwater fshes from pet stores in Klang Valley, Malaysia Abdulwakil Olawale Saba1,2, Ahmad Ismail1, Syaizwan Zahmir Zulkifi1, Muhammad Rasul Abdullah Halim3, Noor Azrizal Abdul Wahid4 & Mohammad Noor Azmai Amal1* The ornamental fsh trade has been considered as one of the most important routes of invasive alien fsh introduction into native freshwater ecosystems. Therefore, the species composition and invasion risks of fsh species from 60 freshwater fsh pet stores in Klang Valley, Malaysia were studied. A checklist of taxa belonging to 18 orders, 53 families, and 251 species of alien fshes was documented. Fish Invasiveness Screening Test (FIST) showed that seven (30.43%), eight (34.78%) and eight (34.78%) species were considered to be high, medium and low invasion risks, respectively. After the calibration of the Fish Invasiveness Screening Kit (FISK) v2 using the Receiver Operating Characteristics, a threshold value of 17 for distinguishing between invasive and non-invasive fshes was identifed. As a result, nine species (39.13%) were of high invasion risk. In this study, we found that non-native fshes dominated (85.66%) the freshwater ornamental trade in Klang Valley, while FISK is a more robust tool in assessing the risk of invasion, and for the most part, its outcome was commensurate with FIST. This study, for the frst time, revealed the number of high-risk ornamental fsh species that give an awareness of possible future invasion if unmonitored in Klang Valley, Malaysia. As a global hobby, fshkeeping is cherished by both young and old people.
