Sevinç 2 Sitesi DB Blok Kat:3 Daire:14 34303 Halkalı/İSTANBUL, Orcid No: 0000-0002-7556-6549/ [email protected] 2Marmara Üniversitesi Göztepe Kampüsü Fen-Edebiyat Fakültesi Biyoloji Bölümü 34722 Kadıköy/İSTANBUL, Orcid No: 0000-0001-9408-3756/ [email protected] Öz: Bu araştırmada tereyağı numuneleri 2012-2013 yılları arasında yaz ve kış mevsimlerinde Kastamonu ve Erzincan illerinden toplanmıştır. Kastamonu ve Erzincan illerindeki çeşitli evlerden toplanan tereyağı numunelerindeki mikrofungusların izolasyonları ve identifikasyonları amaçlanmıştır. Örnekler steril koşullarda alınmış, steril numune kapları içerisine konulmuş, taşıma çantasıyla +3 - +6 °C’ de saklanmış, laboratuvara getirilmiş ve incelenmiştir. Mikrofunguslar Pepton Dekstroz Agar, Malt Ekstrakt Agar (MEA), Patates Dekstroz Agar (PDA) ve Czapek’s Agar (CZ) gibi suni besin ortamlarinda üretildikten sonra, hazırlanan laktofenollü preparatlarında ölçümleri yapılmıştır. İncelenen türlerin identifikasyonları yabancı eserlere göre verilmeye çalışılmıştır. İzole edilmiş olan türlerin fotoğrafları çekilmiştir. Araştırma süresince 16 örnek incelenmiş, toplam 158 fungus kolonisi ve 4 cinse ait 12 tür izole edilmiştir. Araştırma sahasından izole edilen cinsler; Aspergillus, Fusarium, Penicillium ve Geotrichum’ dur. Bunlar arasından yaygın olanlar Aspergillus ve Penicillium’ dur. Araştırmada izole edilen türler ise Aspergillus flavus, A.parasiticus, A.versicolor, Fusarium oxysporum, Penicillium chrysogenum, P.echinulatum, P.griseofulvum, P.palitans, P.roqueforti, P.solitum, P.verrucosum ve Geotrichum candidum’dur. Sonuç olarak, araştırma sahasında, tereyağlarında zararlar yaparak, ekonomik ve sağlık kayıplarına sebep olan bir mikrofungus florası tespit edilmiştir. The ITS-rDNA of both species was analyzed using polymerase chain reaction (PCR) and DNA sequencing. The target region when amplified using universal fungal primers (ITS1F and ITS4) generated 650-650bp fragments. Consensus sequences of both species were submitted for initial blast analysis which revealed and confirmed the identification of both species by comparing the sequences of these respective species already present in the GenBank. Sequence of Pakistani collection of L. acutesquamosa matched 99% with sequences of same species (FJ998400) and Pakistani L. cristata matched 97% with its sequences (EU081956, U85327, AJ237628). Further, in phylogenetic analysis both species distinctly clustered with their respective groups. Morphological characters like shape, size and color of basidiomata, basidiospore size, basidial lengths, shape and size of cheilocystidia of both collections were measured and compared. Four major subclades(Hysterangiales, Geastrales, Gomphales and Phallales) were also demonstratedto be monophyletic. The interrelationships among the subclades were, however, not resolved, andalternative topologies could not be rejected statistically. Nonetheless,most analyses showed that the Hysterangiales and Phallales do not forma monophyletic group, which is in contrast to traditional taxonomy. The higher-level phylogeny of thegomphoid-phalloid fungi tends to suggest that the Gomphales form a sister group with either the Hysterangialesor Phallales. Until recently, eight species were known from this area. The aim of the present study was to gather new data on the hypogeous fungal mycobiota of this mountain range. In July and September of 2016 and 2017, sporocarps of hypogeous fungi were searched on six permanent plots estab- lished in Pieniny National Park. The dominant plant associations were beech and beech-fir forest with spruce, hazel, hornbeam and elm as accompanying trees. Over 100 samples of hypogeous fungi were gathered during the field work. They included 30 species from ascomycete genera: Elaphomyces (3 species), Genea (1), Hydnobolites (1), Hydnocystis (1), Hydnotrya (1), Leucan- gium (1), Pachyphlodes (1), Tuber (4), and basidiomycetes: Chamonixia (1), Gautieria (3), Hyme- nogaster (5), Hysterangium (5), Melanogaster (2) and Octaviania (1). Nine species were most frequent: Hysterangium nephriticum, Melanogaster broomeanus, Hydnobolites cerebriformis, Hydnocystis piligera, Hymenogaster rehsteineri, H. cf. megasporus, Hysterangium stoloniferum and Gautieria morchelliformis. Three species are first records for Poland: Hydnocystis piligera, Hymenogaster cf. arenarius and H. cf. megasporus. The hypogeous mycobiota of the Pieniny Mts now numbers 35 species.

Acta Mycol. 45 (1): 73–89, 2010. Remarks on ecology of several small agarics growing in carr forests of Querco-Ulmetum minoris and Astrantio-Fraxinetum type are presented and discussed. They all occupy special microhabitat (bare ground), belong to certain functional group (saprotrophs) and seem to be connected with carr forest habitat. The base rich or neutral soil humus, generally not favoured by fungi, together with extreme moisture and fast decomposition of litter causes that some humicolous macrofungi in carr forests form mostly small and short-living fruit bodies of hygrophyte character. They represent several genera: Conocybe, Coprinus, Cystolepiota, Entoloma, Flammulaster, Lepiota, Melanophyllum and Pholiotina of the order Agaricales. New localities are given, ecological amplitude is analised and preliminary mycosynusial consideration is presented.
