Cyril and Methodius University, Skopje, Republic of Macedonia *e-mail: [email protected] On different locations of the country, 23 soil profiles of Albic Luvisols were excavated and morphologically de- scribed on the field. More than half of soil profiles (15) were under forest vegetation, 4 soil profiles were under grass and the other 4 on arable land. The survey showed that the cation exchange capacity (CEC) is higher in the humus ac- cumulative horizon A, but lower in the eluvial horizon. Due to increasing amount of clay in a lower part of a soil pro- files, a CEC increases in hor. Bt, BtC and C. The base saturation percentage ranges from 46.93 % to 60.83 %. From these data we can see that the soils are in advanced stage of acidification. In Albic Luvisols, the basic cations (Ca++, Mg ++, K+, Na+) are more common in comparison to acid cations (H+ + Al+++). Exception is the eluvial horizon, where the con- tent of acid cations higher compared to basic cations. Among the exchangeable basic cations, we can see that the con- tent of Ca++ cation has the highest values, Mg++ is less represented, while K+ and Na+ cations have the lowest contents among the exchangeable cations. Liu1 Key words Abstract During a survey of cold-adapted fungi in alpine glaciers on the Qinghai-Tibet Plateau, 1 428 fungal isolates were obtained of which 150 species were preliminary identified. Phoma sclerotioides and Pseudogymnoascus pan- glaciers norum were the most dominant species. Psychrotolerant species in Helotiales (Leotiomycetes, Ascomycota) were Phoma sclerotioides studied in more detail as they represented the most commonly encountered group during this investigation. Two Pseudogymnoascus pannorum phylogenetic trees were constructed based on the partial large subunit nrDNA (LSU) to infer the taxonomic place- Psychrophila ments of these strains. Our strains nested in two well-supported major clades, which represented Tetracladium and psychrotolerant a previously unknown lineage. The unknown lineage is distant to any other currently known genera in Helotiales. Tetracladium Psychrophila gen. nov. was therefore established to accommodate these strains which are characterised by globose or subglobose conidia formed from phialides on short or reduced conidiophores. Our analysis also showed that an LSU-based phylogeny is insufficient in differentiating strains at species level. Additional analyses using combined sequences of ITS+TEF1+TUB regions were employed to further investigate the phylogenetic relationships of these strains. Together with the recognisable morphological distinctions, six new species (i.e. P. antarctica, P. lutea, P. oli- vacea, T. ellipsoideum, T. globosum and T. psychrophilum) were described. Our preliminary investigation indicates a high diversity of cold-adapted species in nature, and many of them may represent unknown species.

A range of definitions exists based on their identity, morphology, location and relationship with their host. Fungal endophytes belong to a wide range of taxa and they are categorized by a variety of characteristics. The detection and identification of these fungal endophytes can be performed using culture-dependent and culture-independent methods. These organisms have a range of application in pharmaceutical discovery and agriculture. Agricultural applications include the exploitation of the growth promoting and protective properties of fungal endophytes in crops such as wheat. This important crop is grown in South Africa where biotic and environmental stresses pose a challenge to its cultivation. Fungal endophytes have demonstrated potential to ameliorate these challenges. Future research will reveal how they can be harnessed to fight food insecurity brought about by stress factors such as climate change. Extraneous DNA interferes with PCR studies of endophytic fungi. A procedure was developed with which to evaluate the removal of extraneous DNA. Wheat (Triticum aestivum) leaves were sprayed with Saccharomyces cerevisiae and then subjected to physical and chemical surface treatments. The fungal ITS1 products were amplified from whole tissue DNA extractions. ANOVA was performed on the DNA bands representing S. cerevisiae on the agarose gel.

In April and May 2019, two ascomycetous species – Vibrissea filisporia (Bonord.) Korf & A. Sánchez 1967 and Cudoniella tenuispora (Cooke & Massee) Dennis 1974 were observed in Tivoli, Rožnik and Šiška hill Landscape Park in central Slovenia. This is the first evidence of their presence in the country. Despite specific growth condition requirements, there is a reasonable probability that these two species grow also elsewhere in Slovenia, but have simply been overlooked. We recommend further studies of suitable habitats for the species, to complete the knowledge on their distribution within the country. Taxa are mainly characterized by asci with a simple pore blueing in Melzer’s reagent, although some taxa have lost this character. The monophyly of this class has been verified in several recent molecular studies. However, circumscription of the orders, families and generic level delimitation are still unsettled. This paper provides a modified backbone tree for the class Leotiomycetes based on phylogenetic analysis of combined ITS, LSU, SSU, TEF, and RPB2 loci. In the phylogenetic analysis, Leotiomycetes separates into 19 clades, which can be recognized as orders and order-level clades.

Schumacher1 Key words Abstract Helvella is a widespread, speciose genus of large apothecial ascomycetes (Pezizomycete: Pezizales) that are found in terrestrial biomes of the Northern and Southern Hemispheres. This study represents a beginning on molecular phylogeny assessing species limits and applying correct names for Helvella species based on type material and specimens in the Pezizales university herbaria (fungaria) of Copenhagen (C), Harvard (FH) and Oslo (O). We use morphology and phylogenetic systematics evidence from four loci – heat shock protein 90 (hsp), translation elongation factor alpha (tef), RNA polymerase II (rpb2) and the nuclear large subunit ribosomal DNA (LSU) – to assess species boundaries in an expanded sample of Helvella specimens from Europe. We combine the morphological and phylogenetic information from 55 Helvella species from Europe with a small sample of Helvella species from other regions of the world. Little intraspecific variation was detected within the species using these molecular markers; hsp and rpb2 markers provided useful barcodes for species delimitation in this genus, while LSU provided more variable resolution among the pertinent species. We discuss typification issues and identify molecular characteristics for 55 European Helvella species, designate neo- and epitypes for 30 species, and describe seven Helvella species new to science, i.e., H. alpicola, H. alpina, H. carnosa, H. danica, H. nannfeldtii, H. pubescens and H.

    A PRELIMINARY CHECKLIST OF ARIZONA MACROFUNGI Scott T. Bates School of Life Sciences Arizona State University PO Box 874601 Tempe, AZ 85287-4601 ABSTRACT A checklist of 1290 species of nonlichenized ascomycetaceous, basidiomycetaceous, and zygomycetaceous macrofungi is presented for the state of Arizona. The checklist was compiled from records of Arizona fungi in scientific publications or herbarium databases. Additional records were obtained from a physical search of herbarium specimens in the University of Arizona’s Robert L. Gilbertson Mycological Herbarium and of the author’s personal herbarium. This publication represents the first comprehensive checklist of macrofungi for Arizona. In all probability, the checklist is far from complete as new species await discovery and some of the species listed are in need of taxonomic revision. The data presented here serve as a baseline for future studies related to fungal biodiversity in Arizona and can contribute to state or national inventories of biota. INTRODUCTION Arizona is a state noted for the diversity of its biotic communities (Brown 1994). Boreal forests found at high altitudes, the ‘Sky Islands’ prevalent in the southern parts of the state, and ponderosa pine (Pinus ponderosa P.& C. Lawson) forests that are widespread in Arizona, all provide rich habitats that sustain numerous species of macrofungi. Even xeric biomes, such as desertscrub and semidesert- grasslands, support a unique mycota, which include rare species such as Itajahya galericulata A. Møller (Long & Stouffer 1943b, Fig. 2c). Although checklists for some groups of fungi present in the state have been published previously (e.g., Gilbertson & Budington 1970, Gilbertson et al. 1974, Gilbertson & Bigelow 1998, Fogel & States 2002), this checklist represents the first comprehensive listing of all macrofungi in the kingdom Eumycota (Fungi) that are known from Arizona.
