Samples were transported at 4-8C in transport medium (medium 199 with Hanks' salts (Sigma, M0393), 4 ml Gentamicin solution (Sigma, G 1397), 11 ml BSA, fraction V (Sigma, A-8412), 11 ml 1 M HEPES buffer solution (Sigma, 83264) and delivered to the laboratory within 8 hours of collection. SARS-CoV-2 infection validation: A commercially available Real-Time Fluorescent RT-PCR kit (BGI, Shenzhen) was used to confirm SARS-CoV-2-positive samples. For the NGS sequencing, samples were selected up to the 24th PCR threshold cycle. RNA extraction and amplification: Total RNA was isolated using the GeneJet Viral DNA and RNA Purification Kit (Thermo Scientific) and quantified by fluorometry. Reverse transcription was performed with three primer mixes (see Table in appendix p. 3) and SuperScript™ IV Reverse Transcriptase (Invitrogen™) in a 40 μL reaction mixture containing 1х SSIV Buffer, 0,5 mM each dNTP, 5 mM DTT, 100 nM each primer, 10 U/μL SuperScript® IV Reverse Transcriptase and 20 μL RNA.

They prove that the development of the traditional dress and decorative and applied arts goes through the following historical stages: an everyday item – a museum piece – the style of life (fashion) – the object to be transformed into modern design. The article is mainly concerned with the recreation of artistic values in new historical environment which is also the reproduction of a certain ethnic and cultural identity in modern days. In this respect, the Kazakh national costume is analysed as a piece of traditional art that is implemented in modern culture due to specific stylistic, expressive and constructive methods. This methodology calls for thorough knowledge of the Kazakh history, the development of the national costume, its construction and colour palette, the study of ancient technologies and the introduction of new ones which can enrich projective and constructive methods as they inherit cultural and historical experience. Focusing on child welfare transformation in post-Soviet Kazakhstan, this thesis pursues two research questions: (1) How have child welfare institutions changed in Kazakhstan in the post-Soviet period? (2) What was the role of (trans)national institutional factors (i.e., legal and regulatory environments, organizational policies and practices, and interorganizational relationships) and Soviet institutional legacies in shaping institutions of child welfare provision in Kazakhstan? Using an integrated multidimensional theoretical framework built upon the welfare diamond, a transnational approach, and historical institutionalism, I examine a mix of national and transnational organizations involved in child welfare reform, conceptualizing them as interconnected policy actors embedded in the historically contingent (trans)national institutional (legal and regulatory) environment. However, the city still benefits from increasing retail turnover, a low unemployment rate, the highest standard of living in Kazakhstan and a growing population. Almaty’s retail sector comprises a mix of traditional ‘bazaar‘-style markets, western-style shopping malls and a small element of on-street retail. Consumers are gradually moving away from shopping at markets and the success of shopping malls indicates a growing consumer demand for this type of retail format. More developers are adopting global retail concepts and trends. The market is witnessing a shift from primarily shopping destinations to one-stop shopping, dining and entertainment experience. Based on the total international-standard retail stock in the city of 423,000 sqm of leasable area and using the estimated population of 2 million people, the volume of retail space per 1,000 capita stands at 210 sqm, that is comparatively less ALMATY mature than developed European markets. A major focus of these efforts is improving the transport and trade sectors to spur economic growth and promote social and political cohesion within the region.
