EFFECTS OF LIGHT REGIME AND IBA CONCENTRATION ON ADVENTITIOUS ROOTING OF AN EASTERN COTTONWOOD (POPULUS DELTOIDES) CLONE Alexander P. Hoffman, Joshua P. Adams, and Andrew Nelson1 Abstract—Eastern cottonwood (Populus deltoides) has received a substantial amount of interest from in- vitro studies within the past decade. The ability to efficiently multiply the stock of established clones such as clone 110412 is a valuable asset for forest endeavors. However, a common problem encountered is initiating adventitious rooting in new micropropagation protocols. Stem segments were collected from bud-broken 1 year old clone 110412 cuttings, sterilized, and stimulated to initiate shoots. Developed shoots (~2 cm in height) were excised and placed into one of three rooting media that included indole-3-butyric acid (IBA) concentrations (0.5 mg/L, 1 mg/L, or 2mg/L) in full strength DKW Medium, full strength Gamborg B5 vitamins, 2 percent sucrose, 0.6 percent agar, 10 mg/L AMP, 0.2 ml/L of Fungigone. In addition to IBA concentration, cuttings were randomly assigned to light rack positions to test the effects of wide spectrum fluorescent light (100 µmol m-2 s-1 photosynthetically active radiation (PAR), 16/8 hour photoperiod) and light emitting diode light (LED; 4:1 red-to-blue diodes, 250 µmol m-2 s-1 PAR, 16/8 hour photoperiod). After a month of exposure, there was limited rooting exhibited across treatments. However, fluorescents (3.58 ± 1.02) produced significantly better performing microcuttings (judged on morphology, visual vigor, and survival) than LEDs (2.7±0.86) (p<0.005). The high light intensity of the LEDs may be prompting weaker performance through unfavorably high transpiration-induced auxin uptake. -

As the white light hits the paint, which colors does the solution absorb and which colors does it transmit? In this activity students will observe the basic principles of absorption spectroscopy based on absorbance and transmittance of visible light. Concepts • Spectroscopy • Visible light spectrum • Absorbance and transmittance • Quantized electron energy levels Background The visible light spectrum (380−750 nm) is the light we are able to see. This spectrum is often referred to as “ROY G BIV” as a mnemonic device for the order of colors it produces. Violet has the shortest wavelength (about 400 nm) and red has the longest wavelength (about 650–700 nm). Many common chemical solutions can be used as filters to demonstrate the principles of absorption and transmittance of visible light in the electromagnetic spectrum. For example, copper(II) chloride (blue), ammonium dichromate (orange), iron(III) chloride (yellow), and potassium permanganate (red) are all different colors because they absorb different wave- lengths of visible light. In this demonstration, students will observe the principles of absorption spectroscopy using a variety of different colored solutions. Food coloring will be substituted for the orange and yellow chemical solutions mentioned above. Rare earth metal solutions, erbium and praseodymium chloride, will be used to illustrate line absorption spectra. Electromagnetic Spectrum Why do some things have colors? What makes color? Why do fast food restaurants use red lights to keep food warm? Why don’t they use green or blue light? Why do X-rays pass through the body and let us see through the body? What has the radio to do with radiation? What are the night vision devices that the army uses in night time fighting? To find the answers to these questions we have to examine the electromagnetic spectrum. FASTER THAN A SPEEDING BULLET MORE POWERFUL THAN A LOCOMOTIVE These words were used to introduce a fictional superhero named Superman. These same words can be used to help describe Electromagnetic Radiation. Electromagnetic Radiation is like a two member team racing together at incredible speeds across the vast regions of space or flying from the clutches of a tiny atom. They travel together in packages called photons. Moving along as a wave with frequency and wavelength they travel at the velocity of 186,000 miles per second (300,000,000 meters per second) in a vacuum. The photons are so tiny they cannot be seen even with powerful microscopes. If the photon encounters any charged particles along its journey it pushes and pulls them at the same frequency that the wave had when it started. The waves can circle the earth more than seven times in one second! If the waves are arranged in order of their wavelength and frequency the waves form the Electromagnetic Spectrum. They are described as electromagnetic because they are both electric and magnetic in nature. -

They help people tell everyone who can see their badge about their communication preferences. A color communication badge is a name tag holder that can pin or clip onto clothing. In the name tag holder there are three cards: one green card that says “GREEN”, one yellow card that says “YELLOW”, and one red card that says “RED.” The card that is currently visible is the active card; the other two are hidden behind the first one, accessible to the person if they should need them. Showing a green badge means that the person is actively seeking communication; they have trouble initiating conversations, but want to be approached by people who are interested in talking. Showing a yellow badge means that the person only wants to talk to people they recognize, not by strangers or people they only know from the Internet. The badge-wearer might approach strangers to talk, and that is okay; the approached people are welcome to talk back to them in that case. But unless you have already met the person face-to-face, you should not approach them to talk. Showing a red badge means that the person probably does not want to talk to anyone, or only wants to talk to a few people. The person might approach others to talk, and that is okay; the approached people are welcome to talk back to them in that case. Color Schemes Are Combinations of Colors
Color is the reflection of light off of an object into our eyes. Our eyes then read the speed of the light and tell us which color that object is. There are two major categories under the heading of color, they are: 1. Neutrals 2. Colors Neutrals are (combinations of) black and white and all grays Colors consist of: Primary colors Secondary colors Intermediate colors also known as Tertiary colors Primary Colors: are the basic colors that you cannot make by mixing. They are natural colors found in nature. They are red, yellow, and blue. Secondary Colors: are made by mixing any two secondary colors. The secondary colors are orange, violet and green. Intermediate Colors: are made by mixing a primary and a secondary color. The secondary colors are, red-violet, blue-violet, blue-green, yellow-green, yellow-orange and red-orange. Color schemes are combinations of colors. There are many different types of color combinations, however, only four of the most basic are included here. They are: • Complementary colors • Analogous colors • Warm & Cool colors • Monochromatic colors Complementary Colors: are any two colors that are opposite each other on the color wheel. Analogous Colors: are any two colors that are adjacent to (or next to) each other on the color wheel. Warm & Cool Colors: warm colors are those colors that contain combinations of red and yellow. There are six. To help you remember what a warm color is, think of the sun or fire. Cool colors are those colors that contain green and blue. There are six of these too. -

Photosynthetic pigments in sediments: development of applications in archaeology and compound-specific radiocarbon analysis Angela Carol Ballantyne A thesis submitted in partial fulfilment of the requirements for the degree of Doctor of Philosophy at the University of York University of York Department of Chemistry October 2012 Abstract Photosynthetic pigments derived from oxygenic aquatic photoautotrophs are biosynthesised from dissolved carbon dioxide that reflects atmospheric concentrations of radiocarbon (14C). Sedimentary pigment signatures are not influenced by a terrestrial signal as terrestrial photosynthetic pigments are overwhelmingly destroyed by photo-oxidation. These properties have been exploited by this study to reveal the presence of archaeological water features and to radiocarbon date the timing of a geochemically significant event. A new approach for identifying archaeological structures suggested to represent former aquatic features has been developed. HPLC and LC-MSn analysis of sediment extracts from several suspected water features revealed the presence of photosynthetic pigment derivatives, thus providing evidence of the occurrence of photoautotrophic and heterotrophic aquatic organisms at the time the sediment was deposited. Chlorophyll derivatives diagnostic of heterotrophic communities and bacteriochlorophyll derivatives which provide information on photic zone anoxia and eutrophication have been detected in some sites. Thus, the detection of photosynthetic pigments in archaeological sediments provides a geochemical method for investigating the existence and evolution of water features in past landscapes. Photosynthetic pigments are ideal candidates for compound specific radiocarbon analysis (CSRA) as they have known primary sources of carbon. Sediments from Kirisjes Pond Antarctica, which have been previously radiocarbon dated using bulk organic material, were extracted and individual pigments isolated and purified by a preparative HPLC system that had been validated with test samples. -

Many kinds of carotenoids are found in algae and, recently, taxonomic studies of algae have been developed. In this review, the relationship between the distribution of carotenoids and the phylogeny of oxygenic phototrophs in sea and fresh water, including cyanobacteria, red algae, brown algae and green algae, is summarized. These phototrophs contain division- or class-specific carotenoids, such as fucoxanthin, peridinin and siphonaxanthin. The distribution of α-carotene and its derivatives, such as lutein, loroxanthin and siphonaxanthin, are limited to divisions of Rhodophyta (macrophytic type), Cryptophyta, Euglenophyta, Chlorarachniophyta and Chlorophyta. In addition, carotenogenesis pathways are discussed based on the chemical structures of carotenoids and known characteristics of carotenogenesis enzymes in other organisms; genes and enzymes for carotenogenesis in algae are not yet known. Most carotenoids bind to membrane-bound pigment-protein complexes, such as reaction center, light-harvesting and cytochrome b6f complexes. Water-soluble peridinin-chlorophyll a-protein (PCP) and orange carotenoid protein (OCP) are also established. Some functions of carotenoids in photosynthesis are also briefly summarized.
