Il N'DEPO Kevin Patrick-Roch - Master 1 / Microbiologie et Biologie Moléculaire/ 2015-2016 RESUME Les bactéries lactiques (BL) sont des microorganismes de catégorie alimentaire qui jouent un rôle important dans la fermentation des matières premières animales et végétales. Elles sont impliquées dans un grand nombre de fermentations spontanées de produits alimentaires. Elles sont principalement utilisées en tant que starter dans les produits alimentaires fermentés où elles permettent de développer certaines caractéristiques organoleptiques et d'augmenter la durée de conservation. Enterococcus et Lactobacillus sont rencontrés dans les aliments fermentés principalement des produits laitiers et des boissons fermentées. Ils contribuent à améliorer et à conserver certains produits alimentaires. Cependant, le pH du milieu ou de l'aliment influence la croissance de ces bactéries. Ces bactéries résistent au milieu défavorable par la production d'acides organiques et de nombreux composés antimicrobiens telles les bactériocines qui jouent un rôle majeur dans la conservation des produits laitiers fermentés et contribuent à l'inhibition des germes pathogènes et des germes d'altération alimentaire. Elles sont impliquées dans un grand nombre de fermentations spontanées de produits alimentaires (Stiles et al, 1997), ce qui a conduit à la reconnaissance de leur statut GRAS (Generally Recognized As Safe) c'est-à-dire généralement reconnues comme sure (Klaenhammer et al., 2005). Actuellement, les bactéries lactiques regroupent treize genres bactériens différents, grâce aux techniques de biologies moléculaires: Lactobacillus, Bifidobacterium, Leuconostoc, Lactococcus, Enterococcus, Streptococcus, Pediococcus, Carnobacterium, Oenococcus, Weissella, Aerococcus, Tetragenococcus et Vagococcus (Carine et al., 2009). En outre, les BL telles que Enterococcus et Lactobacillus font partie des aliments fermentés principalement des produits laitiers et des boissons fermentées. Ils contribuent à améliorer les propriétés organoleptiques et à conserver certains produits alimentaires. les BL sont principalement utilisées en tant que starter dans les produits alimentaires fermentés où ils permettent de développer certaines caractéristiques organoleptiques et d'augmenter la durée de conservation (Abee, 1995; Hugenholtz et al., 1999). Les bactéries lactiques ont longtemps été utilisées pour la conservation et la fabrication des aliments (Soomro et Kiran, 2002). Le concept d'amélioration des bilans nutritionnels et sanitaires par addition de bactéries bénéfiques (probiotiques) à l'aliment a fait l'objet de recherches approfondies. Le rôle propice des probiotiques en tant que facteur d'équilibre sur la microflore intestinale et en particulier leur action vis-à-vis d'une population microbienne indésirable a été démontré (Ouwehand, 1998; San ni et al., 1999; Soomro et Kiran, 2002). Les BL contribuent, par leur métabolisme et leurs activités enzymatiques variées, à la production de composés volatils qui participent au développement de l'arôme, de la saveur et de la texture des produits fermentés (Cheri-Ho, 1997; Oyewole, 1997). Cependant, le pH du milieu exerce parfois une influence sur la croissance de ces BL. Cela peut résulter en une résistance de ces BL par l'acidification du milieu, la production d'acides organiques (Desmazeaud, 1996; Cheri-Ho, 1997; Oyewole, 1997; Daly et Davis, 1998; Ouwehand, 1998). Dans notre synthèse bibliographique, nous parlerons de l'influence du pH des milieux de culture ou de l'environnement de croissance des bactéries lactiques. NNPD affects piglets during the first week of life and results in impaired welfare, decreased weight gain, and in the worst-case scenario death. Commonly used preventative interventions such as vaccination or treatment with antibiotics, have a limited effect on NNPD. Previous studies have investigated the clinical manifestations, histopathology, and to some extent, microbiological findings; however, these studies were either inconclusive or suggested that Enterococci, possibly in interaction with Escherichia coli, contribute to the aetiology of NNPD. This study examined ileal and colonic luminal contents of 50 control piglets and 52 NNPD piglets by means of the qPCR-based Gut Microbiotassay and 16 samples by 454 sequencing to study the composition of the bacterial gut microbiota in relation to NNPD. Results: NNPD was associated with a diminished quantity of bacteria from the phyla Actinobacteria and Firmicutes while genus Enterococcus was more than 24 times more abundant in diarrhoeic piglets. The number of bacteria from the phylum Fusobacteria was also doubled in piglets suffering from diarrhoea. With increasing age, the gut microbiota of NNPD affected piglet and control piglets became more diverse. Independent of diarrhoeic status, piglets from first parity sows (gilts) possessed significantly more bacteria from family Enterobacteriaceae and species E.

While special attention is paid to comparative genomic analysis among clinical relevant isolates, in this study we performed an exhaustive comparative analysis of enterococcal genomes of food origin and/or with potential to be used as probiotics. Beyond common genetic features, we especially aimed to identify those that are specific to enterococcal strains isolated from a certain food-related source as well as features pre- sent in a species-specific manner. Thus, the genome sequences of 25 Enterococcus strains, from 7 different species, were examined and compared. Their phylogenetic relationship was reconstructed based on orthologous proteins and whole genomes. Likewise, markers associated with a successful colonization (bacteriocin genes and genomic islands) and genome plasticity (phages and clustered regularly interspaced short palindromic repeats) were investigated for lifestyle specific genetic fea- tures.

Manter2, T. L. Weir3, M. C. Silva‑Filho1 & J. M. Vivanco4* Insects are known plant pests, and some of them such as Trichoplusia ni feed on a variety of crops. In this study, Trichoplusia ni was fed distinct diets of leaves of Arabidopsis thaliana or Solanum lycopersicum as well as an artifcial diet. After four generations, the microbial composition of the insect gut was evaluated to determine if the diet infuenced the structure and function of the microbial communities. The population fed with A. thaliana had higher proportions of Shinella, Terribacillus and Propionibacterium, and these genera are known to have tolerance to glucosinolate activity, which is produced by A. thaliana to deter insects. The population fed with S. lycopersicum expressed increased relative abundances of the Agrobacterium and Rhizobium genera. These microbial members can degrade alkaloids, which are produced by S. lycopersicum. All fve of these genera were also present in the respective leaves of either A. thaliana or S. lycopersicum, suggesting that these microbes are acquired by the insects from the diet itself. This study describes a potential mechanism used by generalist insects to become habituated to their available diet based on acquisition of phytochemical degrading gut bacteria. Trichoplusia ni (Hübner, 1803) (Lepidoptera; Noctuidae) larvae are generalist herbivores and considered to be critical agricultural pests, which feed on more than 100 species of plants including tomato, potato, corn, cotton, and many other crops1.
