Laboratory Procedures

Spiperone (hydrochloride) is supplied as a solid. A stock solution may be made by dissolving the spiperone (hydrochloride) in the solvent of choice. Spiperone (hydrochloride) is soluble in the organic solvent DMSO, which should be purged with an inert gas. It is also soluble in water. The solubility of spiperone (hydrochloride) in DMSO and water is approximately 100 mM and 10 mM, respectively. We do not recommend storing the aqueous solution for more than one day.

Description

Spiperone is a typical antipsychotic that antagonizes dopamine D2 and serotonin 5-HT2A receptors 1,2 (Kds = 0.18 and 1 nM, respectively). It also activates calcium-activated chloride channels (EC50 = 9.3 µM), stimulating chloride secretion in cystic fibrosis (CF) airway epithelial cells and in CF transmembrane conductance regulator knockout mice.3
