Coleman, Assisstant Professor of Entomology and Plant Pathology ABSTRACT Cultivated peanut (Arachis hypogaea L.) is an economically important crop that is produced in the United States and throughout the world. However, there are two major fungal pathogens of cultivated peanuts, and they each contribute to substantial yield losses of 50% or greater. The pathogens of these diseases are Cercospora arachidicola which causes early leaf spot (ELS), and Cercosporidium personatum which causes late leaf spot (LLS). While fungicide treatments are fairly effective for leaf spot management, disease resistance is still the best strategy. Therefore, it is important to evaluate and compare different genotypes for their disease resistance levels. The overall goal of this study was to determine resistance levels of different peanut genotypes to ELS and LLS. The peanut genotypes (Chit P7, C1001, Exp27-1516, Flavor Runner 458, PI 268868, and GA-12Y) used in this study include two genetically modified lines (Chit P7 and C1001) that over-expresses a chitinase gene. This overall goal was addressed with three specific objectives: 1) determine suitable conditions for pathogen culture and spore production in vitro; 2) determine suitable conditions for establishing infection in the greenhouse; 3) compare ELS and LLS disease reactions of young plants to those of older plants.

    4 Introduction The research into fungi on juniper in Belarus was carried out by us since 1997 and were summarized in an article “The fungi in the consortium of common juniper in Belarus” (2002) and a monograph “Mycobiota in the consortium of juniper in Bela- rus” (Belomesyatseva, 2004, in Russian). During the data collection we constantly adverted to the publications of other mycologists concerning juniper-associated fungi outside Belarus. First of all they are L. Holm and K. Holm, O. Petrini, F.D. Kern, R.L. Gilbertson, and A. Bernicchia. In our country we recorded rather not great number of species in comparison with literature data on junipericolous fungi and myxomycetes in global scale. The data on these fungi are spread in many sources, and to simplify the task of subsequent researchers we have compiled a checklist of the taxa mentioned in basic publications, which has included 820 spe- cies. Forestalling the chapters on fungi on juniper we should to write in brief about the host genus. According to R. Pigler (1931) the genus Juniperus L. is the single in the subfamilia Juniperoideae Pigl. of the family Cupressaceae F.W. Neger., ordo Pinales (Coniferales). About 70 species of the genus were described. They are ever- green small trees up to 10–18 m height, bushes, or repent bushes. The genus has wide geographical distribution in Northern Hemisphere from Arctic to Subtropics, and over the all Temperate Zone. Some species occur even in mountain areas of Tropic Zone (Central America, West Indies, East Africa). The distribution patterns together with wide ecological amplitude of the species cause the development of different life forms from tall big trees of savanna type in subtropical climate, small many-trunk bush-like trees in temperate climate to low and repent bushes in high mountains, Arctic, and arid areas.

It has been demonstrated recently that the C. beticola CTB cluster is larger than previously recognized and includes novel genes involved in cercosporin biosynthesis and a partial duplication of the CTB cluster. Several genes in the C. nicotianae CTB cluster are known to be regulated by ‘feedback’ transcriptional inhibition. Expression analysis was conducted in wild type (WT) and CTB mutant backgrounds to determine if feedback inhibition occurs in C. beticola. My research showed that the transcription factor CTB8 which regulates the CTB cluster expression in C. nicotianae also regulates gene expression in the C. beticola CTB cluster. Expression analysis has shown that feedback inhibition occurs within some of the expanded CTB cluster genes.

The qPCR assay was able to consistently detect as few as 10 spores with purified DNA and 25 spores based on crude DNA extraction from rods. In 2019, two spore traps were placed in two peanut fields with a history of LLS. Sampling units were replaced every 2 to 4 days and tested with the developed qPCR assay, while plots were monitored for symptom development. The system detected inoculum 35 to 56 days before visual symptoms developed in the field, with detection related to environmental parameters affecting pathogen life-cycle and disease development. This study develops the framework of the qPCR spore trap system and represents the initial steps towards validation of the performance of the system for use as a decision support tool to complement integrated management of LLS. Natural infection of groundnut foliage with Cercospora arachidicola and Phoma arachidicola resulted in the accumulation of at least six antifungal compounds. The dominant antifungal compound was the pterocarpan medicarpin. Abiotic agents including ultra-violet irradiation, salts of heavy metals and detergents were used to elicit medicarpin and other antifungal compounds.
