V Praze, 15. 8. 2016 Podpis 2 Abstract Many orchid species are seriously endangered at present. Reasons for their disappearing from natural habitats remain often unclear. Orchids depend on mycorrhizal symbiosis in nature, however only little is known about this symbiosis. Seeds of some species do not germinate in vitro, making their cultivation for scientific and rescue purposes impossible. We found that seed germination of one of such reluctant species, Pseudorchis albida, is strongly inhibited by nitrates even at extremely low concentrations. As this species prefers oligotrophic mountain meadows, nitrate-induced inhibition probably take place in natural conditions. Surprisingly, we found similar but slightly weaker inhibitory effect also in typical mesophillic species and even in slightly eutrophic ones. The sensitivity to nitrates correlates with trophy level of studied species. This inhibitory effect of nitrates could be weakened by application of a range of growth regulators, including auxins, cytokinins and gibberellins, and also by mycorrhizal fungi. The action of nitratereductase is essential for this inhibitory effect of nitrates. Experiments with NO donors, scavengers, and NO quantification are pointing right at NO as compound which perhaps mediates nitrate inhibitory effect. Based on these results, the nitrate inhibitory effect on orchid seed germination seems to be a part of a complex regulatory network. Keywords: nitric oxide, in vitro , asymbiotic, protocorm , PTIO, SNP, tungstate, seed, germination, dormancy 3 Abstrakt Mnoho orchidejí je v součastnosti silně ohrožených a důvody jejich úbytku z přirozených stanovišť v mnoha případech neznáme. Orchideje jsou v přírodě závislé na mykorizní symbióze, ale znalostí o tomto soužití je velice málo. Některé druhy navíc neklíčí v podmínkách in vitro, což znemožňuje jejich kultivaci jak pro vědecké, tak pro ochranářské účely. Zjistili jsme, že klíčení semen jednoho takového druhu, Pseudorchis albida, je silně inhibováno nitráty, a to dokonce při jejich extrémně nízkých koncentracích. Vzhledem k tomu že tento druh osidluje oligotrofní horské louky, pozorovaná inhibice by se mohla uplatňovat i v přírodě. Zajímavé je, že podobný i když slabší inhibiční efekt jsme pozorovali také u mezofilních druhů a dokonce i u mírně eutrofních. Citlivost k nitrátům přitom koreluje s úživností stanoviště studovaných duhů. Pozorovaný inhibiční efekt je možné oslabit aplikací auxinů, cytokininů i giberelinů, anebo také symbiotickými houbami. Pro inhibiční efekt nitrátů je nezbytná aktivita nitrátreduktázy. Experimenty s donory a zhášeči NO a s kvantifikací NO ukazují, že NO pravděpodobně zprostředkovává tento inhibiční efekt. Inhibiční efekt nitrátů na klíčení semen orchidejí se tedy zdá být součástí komplexní signalizační sítě. Klíčová slova: oxid dusnatý, in vitro, asymbiotický, protokorm, PTIO, SNP, wolframan, semeno, klíčivost, dormance 4 Cíle práce Hlavním cílem projektu je zjistit možný fyziologický význam extrémní citlivosti některých orchidejí k dusičnanům. Pro splnění hlavního cíle byly stanoveny dílčí cíle: 1. Zjistit, zda je extrémní citlivost k dusičnanům specifikem druhu Pseudorchis albida, nebo zda se uplatňuje i u jiných druhů orchidejí. + 2. Otestovat vliv NH4 na klíčení semen vybraných druhů orchidejí. 3. Zjistit, zda může být reakce citlivých druhů na dusičnany ovlivněna působením fytohormonů. 4. Pomocí analýzy obsahů endogenních sacharidů v mladých protokormech poodhalit možný účinek dusičnanů na růst protokormů bezprostředně po vyklíčení. 5. Zjistit, zda je pro inhibiční efekt dusičnanů nutná aktivita nitrátreduktázy. 6. Zjistit, zda se při dusičnany vyvolané inhibici klíčení uplatňuje NO. 7. Zjistit, zda NO může ovlivňovat klíčení semen orchidejí. 8. Zjistit, zda mohou inhibiční efekt dusičnanů oslabit mykorhizní houby. 5 Poďakovanie Toto poďakovanie je síce zo srdca, ale nenavštevujem kardiológa a nemožno teda tvrdiť že nasledovné riadky sú zo zdravotného hľadiska v poriadku. Na druhej strane by som rád vynadal operátorovi ktorý zrušil sim kartu ktorá mala tajomníkovi katedry poslat SMS keď sa pokazila 006ka… kedže simku zrušili všetko mi tam chciplo! D. officinale seeds are minute and contain limited energy reserves, and colonization by a compatible fungus is essential for germination under natural conditions. When the orchid mycorrhizal fungi (OMF) initiates symbiotic interactions with germination-driven orchid seeds, phytohormones from the orchid or the fungus play key roles, but the details of the possible biochemical pathways are still poorly understood. In the present study, we established a symbiotic system between D. officinale and Tulasnella sp. for seed germination. RNA-Seq was used to construct libraries of symbiotic-germinated seeds (DoTc), asymbiotic-germinated seeds (Do), and free-living OMF (Tc) to investigate the expression profiles of biosynthesis and metabolism pathway genes for three classes of endogenous hormones: JA (jasmonic acid), ABA (abscisic acid) and SLs (strigolactones), in D.

Among the light emitting diodes (LEDs), the mixture of blue plus red light efficiently enhanced in vitro growth of the plantlets, whereas inhibitory effect on the plantlets growth was observed under the mixture of red plus far-red light. However, the effect of light quality on the growth of plantlets was more pronounced in ‘Bukduseong’ × ‘Hyesung’ hybrid compared to ‘Chunkwang’ × ‘Hyesung’. Of the various gradients of sucrose tested (0, 7.5, 15, 30 and 60 g l-1), 15 g l-1 sucrose was proven suitable concentration for enhancing growth and growth attributes of ‘Bukduseong’ × ‘Hyesung’, while 15 and 30 g l-1 sucrose were regarded as an optimal concentration for in vitro growth of the plantlets of ‘Chunkwang’ × ‘Hyesung’ hybrid. Although, higher sucrose concentration (60 g l-1) enhanced root growth but induced plantlets abnormality. In addition, as a source of natural product, 50 ml l-1 coconut water effectively enhanced plantlets growth of both hybrids compared to the relative control (without coconut water).

Its detailed description supplemented by photo-plates and information on habitat, phenology and distribution is provided. An artificial key to all taxa occurring under the genus Oreorchis in India is also provided. Its threat status in Indian context is accessed by applying IUCN Red Listing Criteria. Résumé Première observation et statut de conservation de Oreorchis patens (Orchidaceae) en Inde – Oreorchis patens (Orchidaceae) est enregistré pour la première fois en Inde. L'article fournit une description détaillée, des planches de photographies et des informations sur son habitat, sa phénologie et sa distribution géographique. Une clé artificielle d'identification de tous les taxons du genre existant en Inde est également proposée. Son statut de menace dans le contexte indien est évalué sur la base des critères de la Liste rouge de l'UICN. Introduction Oreorchis Lindley is an orchid genus comprising 16 or 17 terrestrial species (Freudenstein, 2005; Pearce and Cribb, 1997) distributed in the montane regions of the Western Himalayas, Tibet, South-West China, Siberia, Manchuria, Japan and Taiwan.     Orchid Research Newsletter No. 75 January 2020 Editorial Orchids are perhaps not the first thing that comes to mind when we think about climate change. Record temperatures, catastrophic droughts, melting glaciers, out-of- control bush fires, burning rainforests and other calamities are of more immediate concern. But when we focus on orchid conservation, it is obvious that climate change looms large. It seems likely that orchids are more vulnerable to climate change than most other plant groups, for the following reasons: (1). Since about 70% of all orchids are epiphytes, they are probably more likely to be affected by drought. Even if mature plants would be able to survive unusually severe droughts, one can imagine that seedlings would be much more vulnerable. If such droughts become too frequent, seedling recruitment will be compromised, and the orchids will die out. (2). Since all orchids go through a mycoheterotrophic stage, at least as as seedlings, they depend on the presence of the right fungi for their long-term survival. It could be that climate change affects these fungi in such a way that they are no longer available to particular orchid species. These will then gradually disappear from their habitats. (3). Similarly, since many orchids depend on highly specific pollinators, the effect of climate change on the availability of these pollinators may be significant. A chain is only as strong as its weakest link, and we do not know if it is the orchid, the fungus or the pollinator that is the weakest link. (4). Orchids tend to occur in sparse, widely dispersed populations.

    Beautiful orchids to grow in your garden Jeff Hutchings As owner of one of only a few nurseries in Europe that specialise in hardy perennial orchids, I have battled for several years to educate gardeners about the numerous species and hybrids which will grow happily in the garden in the same way as any other herbaceous perennials – orchids’ exotic flowers hide the fact that many can withstand most of the weather the British climate throws at them. The last two winters have proved the point, with many plants coming through the weather quite successfully, though, as you’d expect, it has exposed the vulnerability of a few species in some northern areas of the country. Here I’ll outline which orchids can, with the minimum of fuss, be grown successfully in a garden, and give you some tips on how to grow them. Of course, it’s difficult to be specific about the hardiness of some species as the climate in the south west is vastly different from, say, Scotland. In areas where there is a lot of winter rain and very low temperatures it may be sensible to grow the plants in pots and put them in a cold frame when they are in their dormant stage. For the purpose, the samples belonging to 19 orchid taxon were collected in the Black Sea Region. SEM and light microscopy photographs were taken with the standard techniques. Variations among taxa were evaluated using various statistical methods such as correlation and discrimination analysis. Among the investigated characteristics, fruit surface ornamentation is related to habitat preferences of the species while morphometric properties of epidermal cells and structural features such as the type of crystal inclusions are important characters at the genus level.
